Transarterial radioembolization (TARE) is a widely utilized therapeutic approach for hepatocellular carcinoma (HCC), however, the clinical implementation is constrained by the stringent preparation conditions of radioembolization agents. Herein, we incorporated the superstable homogeneous iodinated formulation technology (SHIFT), simultaneously utilizing an enhanced solvent form in a carbon dioxide supercritical fluid environment, to encapsulate radionuclides (such as 131I,177Lu, or 18F) with lipiodol for the preparation of radiolipiodol. The resulting radiolipiodol exhibited exceptional stability and ultra-high labeling efficiency (≥99%) and displayed notable intratumoral radionuclide retention and in vivo stability more than 2 weeks following locoregional injection in subcutaneous tumors in mice and orthotopic liver tumors in rats and rabbits. Given these encouraging findings, 18F was authorized as a radiotracer in radiolipiodol for clinical trials in HCC patients, and showed a favorable tumor accumulation, with a tumor-to-liver uptake ratio of ≥50 and minimal radionuclide leakage, confirming the feasibility of SHIFT for TARE applications. In the context of transforming from preclinical to clinical screening, the preparation of radiolipiodol by SHIFT represents an innovative physical strategy for radionuclide encapsulation. Hence, this work offers a reliable and efficient approach for TARE in HCC, showing considerable promise for clinical application (ChiCTR2400087731).
2.Trends of Incidence and Mortality of Cancers from 2018 to 2022 in Changsha City,Hunan Province

[Purpose]To analyze the trends of cancer incidence and mortality in Changsha City from 2018 to 2022.[Methods]Based on the annual cancer registry data of Changsha City from 2018 to 2022,SAS 9.2 software was used to calculate the crude incidence/mortality rates and age-standardized incidence/mortality rates by Chinese standard population(ASIRC/ASMRC).The age-standardized rate was standardized using the population composition of China in 2000.Join-point 4.9.0.0 software was used to fit a Log-linear regression model to calculate the average annual percentage change(AAPC)and its 95%confidence interval(CI)for trend analysis.A linear regression model was applied to calculate the contribution rate of each cancer type to the trend change,with a significance level of α=0.05.[Results]From 2018 to 2022,the crude incidence rate of malig-nant tumors in Changsha City was 367.98/105(ASIRC:232.28/105),and the crude mortality rate was 192.36/105(ASMRC:106.11/105).Both incidence and mortality rates were higher in male than those in female.The incidence and mortality rates remained relatively low before age 50 but rose rapidly thereafter:the incidence peaked in the age group of 80~84 years old,while mortality peaked in the age group of 85 years old and above.The top 5 cancers by incidence were lung cancer,female breast cancer,colorectum cancer,cervical cancer,and thyroid cancer;while the top 5 cancers by mortality were lung cancer,colorectum cancer,liver cancer,female breast can-cer,and cervical cancer.The ASIRC increased significantly in the total population(AAPC=4.71%,95%CI:3.20%~6.23%),male(AAPC=2.44%,95%CI:0.35%~4.58%),and female(AAPC=7.17%,95%CI:4.00%~10.43%).The ASMRC increased significantly in the total population(AAPC=2.94%,95%CI:0.73%~5.20%)and male(AAPC=3.37%,95%CI:1.63%~5.15%),but the trend in female was not statistically significant(P>0.05).Contribution rate analysis showed that thyroid cancer(51.25%),female breast cancer(12.18%),lymphoma(6.02%),and oral cavity and pharyngeal can-cer(5.28%)were the main cancer types contributing to the increased ASIRC;lung cancer(26.10%),colorectum cancer(14.89%),oral cavity and pharyngeal cancer(5.73%),lymphoma(5.60%),and thyroid cancer(2.08%)were the main ones contributing to the increased ASMRC;liver cancer(49.82%)and esophageal cancer(19.55%)were the main causes of decreased ASIRC,with their contribution rates to decreased ASMRC reaching 77.69%and 11.16%,respectively.[Conclusion]The disease burden of malignant tumors in Changsha City continued to increase from 2018 to 2022.It is necessary to strengthen prevention and control for cancers with a signifi-cant upward trend(such as thyroid cancer,lung cancer,and colorectum cancer)and high-risk populations(such as male and middle-aged and elderly people),while consolidating the prevention and control effects for liver cancer and esophageal cancer.
3.Effect of zerotime exercise intervention on physical activity levels and sedentary behavior among college students

Objective:
To explore the effects of a zerotime exercise (ZTEx) intervention on physical activity levels and sedentary behavior among college students, providing evidence for improving physical activity and reducing sedentary habits.
Methods:
In September 2023, 45 sedentary college students from a university in Fuzhou were recruited and randomly assigned to either the ZTEx group (23 students) or the control group (22 students) according to a random number table method. ZTEx group received two ZTEx focus group meetings for 3 months, while the control group received safety and health education. The International Physical Activity Questionnaire and a threeaxis accelerometer were used to evaluate the sedentary and physical activity levels of college students. At the same time, evaluations related to physical health and psychological questionnaires were completed. Mixed effects analysis of variance and nonparametric tests were used to statistically analyze the physical health, psychological questionnaire, and sedentary and physical activity data of the college students.
Results:
Postintervention, the ZTEx group showed significant improvements in the duration of lowintensity physical activity [pretest(1 492.78±369.50)min; posttest(1 918.93±354.63)min] and the number of sedentary interruptions [pretest(45.26±13.69)times; posttest(73.78±16.74)times]; grip strength [pretest(28.77±9.23)kg; posttest(31.78±8.00)kg]; sitting up continuously for 30 seconds [pretest(22.52±4.90)times; posttest(26.96±4.87)times]; general selfefficacy [pretest(26.52±4.14)points; posttest(32.96±5.24)points]; body composition summary [pretest(66.44±4.83)points; posttest(72.62±4.88)points]; and psychological composition summary [pretest(61.21±9.88)points; posttest(63.98±9.57)points], while reducing sedentary time [pretest(3 694.28±687.56)min; posttest(2 865.90±493.81)min] in the past 7 days, the differences were statistically significant(P<0.05). The control group exhibited no significant changes(P>0.05).
Conclusion
ZTEx effectively improve the lowintensity physical activity, increases sedentary breaks, and reduces prolonged sitting among college students, fostering healthier habits and improving physical/mental wellbeing.
4.Construction and performance study of biological treatment experimental system for space wastewater

Objective To address the bottlenecks in the application of wastewater biological treatment technology under space conditions,an experimental system for the biological treatment of space wastewater was constructed and its biochemical performance examined.The findings of this study will provide technical support for the biological treatment of space wastewater.Methods Based on the Membrane Aerated Biofilm Reactor(MABR)process,a biological treatment experimental system for space wastewater was constructed and conducted the continuous flow test for 77 days to investigate the performance of PVDF and PP membrane modules in the treatment of simulated air condensate.Results The results demonstrated that both membrane modules exhibited an average TOC removal rate of 90%,indicative of their effective organic matter removal capacity.In the air supply mode,the ammonia oxidation capacity was observed to be comparatively lower,whereas in the oxygen source without bubbling mode,the nitrogen oxidation rate and total nitrogen removal rate could be attained above 90%,indicating a notable degree of simultaneous nitrification and denitrification.The results demonstrated that the mode of gas supply had a significant impact on the nitrogen conversion performance.The abundance of nitrogen-converting bacteria in PP membrane module is higher than that in PVDF membrane module,indicating a better nitrogen-converting performance in PP membrane module.Conclusion The constructed wastewater biological treatment system is optimally suited for the treatment of air condensate,thereby offering a novel technical approach for space wastewater treatment.
5.Research progress on clot waveform analysis in thrombotic diseases

In recent years, with the continuous advancement of related research, the clinical value of clot waveform analysis (CWA) in the diagnosis and management of thrombotic diseases has become increasingly prominent. As a dynamic coagulation monitoring technology based on optical principles, CWA overcomes the limitations of traditional coagulation tests (e.g., APTT, PT), which rely on single time-point parameters, in identifying hypercoagulable states, predicting thrombotic risk, and monitoring anticoagulant efficacy. By analyzing the kinetic profiles of the coagulation cascade, CWA provides multidimensional insights. This article elucidates the theoretical basis and principles of CWA, systematically reviews its applications in arterial/venous thrombosis, other hypercoagulability-related disorders, and anticoagulation therapy monitoring, and synthesizes recent advances of CWA for thrombotic diseases. Specifically, multiple studies demonstrate that CWA-APTT parameters (e.g., peak height Min1, Min2, Delta) can sensitively detect hypercoagulability. Combining CWA with the Padua score significantly enhances the predictive power for venous thromboembolism (VTE) risk assessment. CWA shows clinical utility in evaluating hypercoagulability in patients with acute myocardial infarction (AMI) and acute cerebral infarction (ACI), where parameters such as Min1, Min2, and Max2 exhibit greater sensitivity than conventional APTT. These metrics may predict AMI complications and guide clinical management. Additionally, CWA demonstrates value in diverse scenarios including pregnancy, inflammation-associated hypercoagulability (e.g., COVID-19, Kawasaki disease), and rare thrombotic conditions (e.g., chronic spontaneous urticaria). Beyond diagnostic and risk-stratification advantages, CWA serves as a novel tool for personalized anticoagulation monitoring. Its derivative technology, clot-fibrinolysis waveform analysis (CFWA), extends applications to fibrinolysis assessment—aiding in identifying coagulation status in deep vein thrombosis (DVT) patients, tracking coagulation/fibrinolysis dynamics in COVID-19, and evaluating efficacy of anticoagulant/antifibrinolytic therapies. Nevertheless, despite its unique strengths, challenges such as device dependency, insufficient standardization, and heterogeneity in parameter interpretation hinder widespread clinical adoption, necessitating further investigation. Future directions include establishing multidimensional thrombotic risk assessment systems integrating CWA and developing AI-powered automated CWA analysis platforms to enhance clinical accessibility.
6.The relationship between size distribution of circulating extracellular vesicles and coagulation function after cardiac surgery

Objective:To investigate the relationship between the changes in extracellular vesicles (EVs) size distribution before and after cardiopulmonary bypass (CPB) cardiac surgery and postoperative coagulation function.Methods:A total of 103 patients undergoing cardiac surgery with CPB were enrolled. Venous blood samples were collected at preoperation, postoperative 12 h and 3 days. Additionally, 50 age- and gender-matched healthy volunteers served as a control group. EVs were isolated using gradient centrifugation, and their size distribution was assessed by dynamic light scattering (DLS). The relationship between EV size characteristics, including peak diameter, peak height, and interquartile range( IQR), and postoperative coagulation function was analyzed. Results:Compared to patients with normal postoperative coagulation function, those with postoperative coagulation dysfunction had lower size at peak and IQR, and significantly higher peak intensity. Logistic regression analysis indicated that elevated peak intensity and lower size at peak and IQR were risk factors for coagulation dysfunction. The area under the curve ( AUC) for diagnosing coagulation dysfunction with 12 h postoperative EVs peak intensity was 0.76, with a positive predictive value of 85% at the optimal cutoff of 8.2; the AUC for IQR was 0.84, with a sensitivity of 83%, specificity of 82%, and negative predictive value of 86% at the optimal cutoff of 125.05 nm. Conclusion:The size distribution of circulating EVs show a correlation with coagulation function after cardiac surgery with CPB and may serve as a novel biomarker to predict postoperative coagulation dysfunction.
7.Application of CRISPR-based one-pot droplet assay for BK virus nucleic acid detection

Objective:To establish a method for quantitative detection of BK virus (BKV) nucleic acid using microfluidic droplet one-pot Recombinase polymerase amplification-clustered regularly interspaced short palindromic repeats, RPA-CRISPR/Cas13a.Methods:A methodology establishment study. Plasma or urine samples were collected from 50 post renal transplant patients and 40 healthy individuals in Renji Hospital, Shanghai Jiao Tong University School of Medicine, from May 16 to August 31, 2024, and the viral genomic DNA was extracted. In the renal transplant group, there were 28 males and 22 females aged (46.1±11.6) years; in the healthy control group, there were 21 males and 19 females aged (45.6±11.3) years. Three crRNAs were designed for screening the optimal crRNA in CRISPR/Cas13a reaction taking PCR product as templet. Two pairs of RPA primers targeting BKV sequences were designed and selected an optimal primer set from four combinations for RPA-CRISPR/Cas13a reaction. Microfluidic droplet generation chip was designed and fabricated for rapid quantitative detection of BKV nucleic acids, which was combined with a droplet reader. BKV plasmid standards were utilized to assess the detection sensitivity of the one-pot assay. The CRISPR droplet one-pot assay and qPCR were utilized to detect the BKV load in the above samples, respectively. The methodological consistency was then analyzed, and Kappa coefficients of the results were calculated using the Kappa test, and the correlation between the two methods was evaluated using linear regression analysis. Results:The RPA-CRISPR/Cas13a reaction system was combined with a droplet chip to establish a CRISPR microfluidic droplet one-pot assay for BKV detection. The sensitivity of the CRISPR microfluidic droplet one-pot assay for detecting BKV was 10 copies/ml, which surpassed that of qPCR (100 copies/ml) without cross-reaction with JC virus. Methodological consistency analysis demonstrated that the Kappa coefficient of the two methods was 0.96, and the coefficient of determination R 2 was 0.95. Conclusion:In this study, a novel BKV detection technology based on the one-pot CRISPR droplet assay was successfully established.
8.Establishment and application of a one-pot lyophilized CRISPR system for detecting CMV in liver transplant recipients

Objective:To establish a one-pot lyophilized detection system based on recombinase polymerase amplification (RPA) and Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) and CRISPR-associated protein (Cas13a) technology for the rapid diagnosis of cytomegalovirus (CMV) infection in liver transplantation recipients.Methods:This study is a methodology study. CRISPR RNA (crRNA) and RPA primers were designed targeting the CMV gene sequence. Optimal RPA primer sets were screened to establish the RPA-CRISPR/Cas13a-based CMV detection system. The limit of detection (LOD) was evaluated using gradient-diluted CMV plasmid standards. Cross-reactivity was assessed using genomic DNA from common opportunistic viruses in organ transplant recipients. Lyophilized reagents were validated with CMV-negative and positive samples. P-values were computed using two-sample t-tests for pairwise comparisons and one-way ANOVA for multi-group analyses to assess fluorescence value differences. Subsequently, lyophilized reagents were employed to detect 22 plasma samples from liver transplantation recipients collected at Renji Hospital, Shanghai Jiao Tong University School of Medicine, from June 3, 2024, to May 31, 2025. The test results were then compared with those obtained using quantitative real-time polymerase chain reaction (qPCR). Consistency between the two methods was evaluated using the Kappa coefficient calculated by Kappa test.Result:The established RPA-CRISPR/Cas13a system achieved a detection sensitivity of 1 copy/reaction and exhibited no cross-reactivity with other common opportunistic viruses in organ transplantation. Lyophilized RPA-CRISPR/Cas13a reagents demonstrated performance equivalent to non-lyophilized reagents. Concordance between lyophilized reagent detection and qPCR results for 22 clinical samples was 100% (22/22).Conclusion:A lyophilized CMV detection method based on RPA-CRISPR/Cas13a technology was successfully developed and validated for convenient diagnosing CMV infection in liver transplant recipients.
9.Simultaneous one-stop interventional closure treatment for left atrial appendage and congenital atrial septal defect:a long-term follow-up comparison study

Objective To compare the clinical effect of left atrial appendage(LAA)plus atrial septal defect(ASD)closure therapy and ASD closure therapy in treating ASD associated with atrial fibrillation(AF).Methods A total of 102 patients with ASD complicated by non-valvular AF,who were admitted to the General Hospital of Northern Theater Command of China from January 2016 to December 2023,were enrolled in this study.Of the 102 patients,simultaneous one-stop interventional transcatheter LAA plus ASD closure was performed in 52(LAA+ASD closure group)and ASD closure was performed in 50.(ASD closure group).The perioperative and postoperative 30 d,90 d,180 d clinical safety and efficacy were compared between the two groups.Telephone follow-up was conducted,the complications such as embolization and bleeding were recorded,and the medium-to-long-term follow-up results were compared between the two groups.Results The immediate surgical success rate in both groups was 100%.The immediate postoperative monitoring showed that the occlusion effect was satisfactory.In LAA plus ASD closure group,LACBES LAA occluder was used in 27 patients and LAmbre LAA occluder was adopted in 25.There were no statistically significant differences in the patients' baseline characteristics between the two groups(all P>0.05).In the LAA+ASD closure group,3 patients developed cardiac tamponade,among them 2 patients were cured after pericardiocentesis drainage and one patient was referred to the surgery department to receive occluder removal and intracardiac repair.Medium-to-long-term follow-up was conducted in 101 patients with a median follow-up period of 37.6 months.The incidence of embolic events in the LAA+ASD closure group was lower than that in the ASD closure group(3.9%vs.18.0%,P=0.028).The incidence of bleeding events in the ASD closure group was higher than that in the LAA+ASD closure group(16.00%vs.1.96%,P=0.016).Kaplan-Meier analysis indicated that the risk of occurring embolic events and bleeding events in the LAA+ASD closure group was strikingly lower than that in the ASD closure group(HR=4.295 and 7.888 respectively,95%CI:1.317-14.010 and 2.135-29.140 respectively,P=0.040 9 and P=0.020 8 respectively).Conclusion Simultaneous interventional transcatheter LAA plus ASD closure can effectively prevent embolic events such as stroke,etc.in patients with ASD complicated by AF,and its bleeding risk is lower than simple ASD closure.
