1.Relationship between MTHFR polymorphism and the effect of hyperbaric oxygenation combined with drug therapy in the patients with profound sudden deafness

Objective To explore the effect of gene mutation on sudden deafness through investigation on the relationship between MTHFR C677T polymorphism and the effect on such patients, so as to provide solid evidence for the treatment of sudden deafness. Methods Samples of venous blood in 99 patients with profound sudden deafness were collected and DNA was extracted from them. Real-time PCR was used for SNP genotyping of MTHFR gene. The patients underwent HBO and drug therapy and therapeutic effects were observed, then, the relationship between different genotypes and therapeutic effects were compared. Results The mutation types and frequencies of MTHFR genotype in the study population were: C/C 17. 17%, C/T 51. 51% and T/T 31. 32%. The effective rate in the patients with profound sudden deafness treated with HBO and drug therapy was 75. 76%, and there was no statistical significance in the distribution of C/C and C/T+T/T genotypes, when comparisons were made between the effective and non-effective groups(P>0. 05). Conclusion Study results revealed that MTHFR gene polymorphism was not associated with the efficacy of HBO combined with drug therapy in the patients with profound sudden deafness.

3.Effects of hyperbaric oxygen on skin ischemia induced by hyaluronic acid

Objective To observe the therapeutic effects of hyperbaric oxygen ( HBO ) on skin ischemia induced by hyaluronic acid .Methods Forty-two patients with skin ischemia induced by hyaluronic acid were divided into 2 groups: The patients in group A were given routine treatment , and some patients received HBO therapy 72 hours later, while the patients in group B all received HBO therapy within 72 hours after injection of hyaluronic acid on the basis of routine therapy .Effectual time was recorded and compared between the patients of the 2 groups, and Pearson analysis was performed with respect to HBO implementation time and effectual time.Effectual rates were compared between the 2 groups, 10 days after injection of hyaluronic acid.Results The average effectual time of group A was (13.2 ±2.8)days, while that of group B was(11.0 ±3.1) days.Statistical significance could be noted when comparisons were made between them ( P<0.05).Results of Pearson analysis concerning HBO implementation time and effectual time indicated that there was a positive correlation between HBO implementation time and effectual time (P<0.05, r>0).Observation at day 10 showed that the effectual time of group A was 28.57%and that of group B was 53.57%.Conclusion Therapeutic effects of early HBO therapy on skin ischemia induced by hyaluronic acid injection was positive , and the earlier HBO intervention was made , the earlier the effectual time was .

5.Effect of hyperbaric oxygen coupled with cisplatin on the apoptosis of Fadu cells in vitro

Objective To investigate the effect of hyperbaric oxygen (HBO) coupled with cisplatin on the apoptosis of human hypopharyngeal cancer Fadu cells in vitro and also to observe the effect of HBO exposure at different time points on the efficacy of cisplatin.Methods Human hypopharyngeal cancer Fadu cells for culture were bought from the Tumor Research Institute,Shanghai Jiaotong University.MTT assay was applied to observe the effect of different concentrations of cisplatin on the proliferation inhibition of Fadu cells in vitro.In accordance with different treatment,Fadu cells were divided into 6 groups:group C (the control group),group HBO (the HBO group),group CHT (the chemotherapy group),group HBO + CHT (the HBO + chemotherapy group),group CHT + HBO (the chemotherapy + HBO group and group IC (the HBO immediately combined with chemotherapy group).Cells of various groups were detected by flow cytometry 48 hours after experiment to observe changes in apoptosis,and adenosine triphosphate (ATP) changes in the cells of various groups were measured by ATP kit.Results Apoptosis rates of group C,HBO,CHT,HBO + CHT,CT + HBO and IC were respectively(2.00 ±0.17)%,(2.07 ±0.22)%,(8.73 ±0.74)%,(11.9 ±0.68)%,(9.92 ±0.27)% and(10.4 ±0.72)%.No statistical significance could be noticed in the apoptosis rates of group C and HBO(P >0.05).The apoptosis rates of group CHT,HBO + CHT,CHT + HBO and IC were all higher that those of group C and HBO,and statistical significance could be noted when comparisons were made between them(P <0.01).Experiments showed that the apoptosis rate of group HBO + CHT was higher than those of the other groups,with statistical significance(P < 0.05).Suppose the ATP level of group C was 100%,the ATP levels of other groups were respectively 98.7%,83.9%,61.3%,78.5% and 77.1%.There was statistical significance in the ATP levels of group C and HBO(P >0.05).However,the ATP levels of group CHT,HBO + CHT,CHT + HBO and IC were all lower than those of group C and HBO,also with statistical significance (P <0.01).The ATP level of group HBO + CHT was significantly lower than those of the other groups,also with statistical significance(P < 0.05).Conclusions Single HBO exposure had no effect on the apoptosis of hypopharyngeal cancer Fadu cells.HBO coupled with cisplatin could enhance the therapeutic efficacy of cisplatin,promote apoptosis,and the efficacy might be associated with the time points of the combined treatment of HBO.

7.Effects of normobaric hyperoxia on the learning and memory abilities of valproic acid autism model rats and the morphology of pyramidal cells in hippocampal CA1 region

Objective To investigate the effects of normobaric hyperoxia intervention on learning and memory abilities of valproic acid(VPA) autism model rats and the morphology of pyramidal cells in hippocampus CA1 area.Methods Animal model groups of autism were obtained in male offspring of the Wistar rats that received intraperitoneal injection of 600 mg/kg VPA at the 12.5 day after pregnancy.According to the eye opening time,behavior,weaning weight and the learning and memory abilities which were evaluated by the Y electricity maze test at the 28th day after birth,40 male VPA autism model rats were randomly selected 20 only and divided into normobaric hyperoxia model group (group A,n =10),atmospheric air model group (group B,n =10).Normal control groups were obtained in male offspring of Wistar rats that received intraperitoneal injection of equivalent physiological saline at the same period pregnancy.(group C,n =10).Rats in group A were treated with oxygen for 1 h per day and lasted 1 week;group B and C were treated with normal air.The learning and memory abilities of three groups were assessed at the 35th day after birth.The immunohistochemistry methods and image analysis were used to observe the pyramidal cells of autism model rats in hippocampal CA1 region.The effect of normobaric hyperoxia therapy on pyramidal cell of autism model rats in hippocampal CA1 region were evaluated by HE staining technique.Results The trying times of group A after treatment were less than those before treatment (31.15 ± 0.99 vs 31.54 ± 0.97,t =2.739,P =0.018).The memory times were more than those before treatment (3.00± 0.58 vs 2.69 ± 0.48,t =-2.309,P =0.040).The trying times of group A after treatment were less than those in group B after treatment (P =0.016).The memory times of group A were not different from that in group B after treatment(P=0.810).The morphology of pyramidal cells in hippocampal CA1 region showed that the pyramidal cells of the autism model rats had apoptosised.The number of apoptotic cells reduced and the number of normal form cells increased after the normobaric hyperoxia intervention compared with the autism model rats.Conclusion Normobaric hyperoxia intervention can improve the learning and memory abilities of the autism model rats.The apoptosis of the pyramidal neurons in hippocampal CA1 might be reduced after the normobaric hyperoxia intervention.
8.Hyperbaric oxygen plus Nimodipine for carbon monoxide poisoning

Four hundred eighty patients with carbon monoxide poisoning were randomly assigned to receive hyperbaric oxygen alone or hyperbaric oxygen plus Nimodipine.Treatment outcomes and the incidence of delayed encephalopathy after acute carbon monoxide poisoning(DEACMP)were observed.We found that the incidence of DEACMP was 14.0%(67/480)in all cases,19.2%(46/67)in hyperbaric oxygen group,and 8.8%(21/67)in hyperbaric oxygen plus Nimodipine group(hyperbaric oxygen group vs hyperbaric oxygen plus Nimodipine group,P<0.05).These results suggested that hyperbaric oxygen combined with Nimodipine could prevent the development of DEACMP.
