1.Mechanism of PRDX3 Involved in Development and Progression in Clear Cell Renal Cell Carcinoma

【Objective】 To investigate the relationship between the expression of PRDX3 (thioredoxin-dependent peroxide reductase)and the occurrence and development of ccRCC (clear cell renal cell carcinoma). 【Methods】 The expression of PRDX3 was first verified in 16 cases of ccRCC tissues and adjacent normal tissues. In the present study , according to the PRDX3 over-expression level,we established the stable PRDX3 overexpression cell lines and knockdown cell lines in 786-O cell lines. We detected the growth rate of tumor cells after overexpression and knockdown of PRDX3. Interaction proteins with PRDX3 were searched by anti-flag pull-down test combined with LC- MS/MS technique. The interaction between PRDX3 and PRDX1(peroxiredoxin 1)was preliminarily explored.【Results】The western blot results showed that PRDX3 were down- regulated in 14 out of 16 ccRCC tissue samples about 1.78 times. Stable PRDX3 overexpression and knockdown cell lines and those control group were successfully established[786O- PRDX3(+)and 786O- PRDX3(-),786O- PRDX3 KN and 786O- PRDX3 NCi]. PRDX3 expression in 786O- PRDX3(+)was 2.1 times higher than 786O- PRDX3(-)at mRNA level and 1.8 times at protein level. PRDX3 expression in 786O- PRDX3 KN was 0.48 times lower than 786O-PRDX3 NCi at mRNA level and 0.51 times at protein level. The cell growth rate of 786O-PRDX3 (+)cell lines was significantly lower than that of 786O-PRDX3(-). Meanwhile ,there was no significant difference in 786O-PRDX3 KN and NCi cell lines. Pull-down results shows that PRDX3 may interact with PRDX1 through disulfide bond and the binding sites of those two proteins were identified respectively.【Conclusion】PRDX3 was down- regulated expression in renal clear cell carcinoma and the interaction with PRDX1 may be involved in the occurrence and development of tumor. Increasing the expression level of PRDX3 can significantly reduce the growth rate of tumor cells. Based on PRDX3 ,it is possible to develop targeted drugs for treating renal clear cell carcinoma.
2.Protective effects of bone marrow mesenchymal stem cell-derived exosomes on smoke inhalation injury in rats

Objective Acute lung injury induced by variety causes can be reduced by mesenchymal stem cells.Some studies have shown that mesenchymal stem cell-derived exosomes have similar features with mesenchymal stem cell,but its role in acute lung injury is less studied.The study was to investigate the protective role and underlying mechanisms of bone marrow mesenchymal stem cell-derived exosomes (BMSC-DEs) on smoke inhalation injury (SⅡ) in rats.Methods Thirty Wistar rats were randomly divided into 3 equal groups:normal control group,smoke inhalation injury (SⅡ) model group and bone marrow mesenchymal stem cell-derived exosomes (BMSC-DEs) treated group.12 h after establishing the SⅡ model,BMSC-DEs treated group was injected with 0.5 mL BMSC-DEs (derived from 4× 106 BMSCs),and normal control group and SⅡ model group were injected with equivalent volume of normal saline.7 days later,samples were collected.The histopathologic changes of lung were observed after HE staining;BCA was used to test the amounts of total protein in bronchoalveolar lavage fluid (BALF);Enzyme linked immunosorbent assay was used to test the levels of tumor necrosis factor-α (TNF-α) and keratinocyte growth factor (KGF) in the lung tissue;Immunohistochemical was used to test the levels of pulmonary surfactant protein C(SP-C).Results The BALF levels of total protein of SⅡ group was significantly higher than those of normal control group (P<0.01) and BMSC-DEs groups(P<0.05);Compared with normal group [(0.164±0.021) ng/L],the levels of tumor necrosis factor-α of SII and BMSC-DEs groups [(0.355±0.106)、(0.234±0.024) ng/L] (P< 0.05) were significantly higher,and SⅡ group was higher than that of BMSC-DEs group(P<0.01);Compared with normal group,the KGF protein expression level in lung tissue of SⅡ group was significantly lower (P<0.05),but BMSC-DEs group was higher (P<0.05).BMSC-DEs group was higher than SⅡ group (P<0.01);Immunohistochemistry showed that the SP-C expression level in lung tissue of SⅡ group was significantly lower than those of other groups (P<0.05).There was no statistically difference between BMSC-DEs group and control group (P>0.05).Conclusion BMSC-DEs has a protective effect of smoke inhalation injury rats,the underlying mechanism may be related to BMSC-DEs to reduce inflammation and promote restoration of the alveolar epithelial type Ⅱ.
3.Exploring the relationship between secreted frizzled-related protein 1 and chronic periodontitis

Objective This study detects the expression of secreted frizzled-related protein 1 (SFRP1) in healthy patients and patients with chronic periodontitis (CP) and explores the relationship between SFRP1 and the occurrence and development of CP. Methods First, 28 patients forming the CP group were further divided into mild, moderate, and severe CP subgroups according to clinical attachment loss (CAL) data. Ten healthy volunteers were recruited in the control group. Gingival crevi-cular fluid (GCF) was collected from all of the patients, and the concentration of SFRP1 in the GCF samples was detected using enzyme-linked immunosorbent assay. Next, gingival lesions were obtained from 22 patients in the CP group and healthy gingival tissues were obtained from the 10 healthy patients in the control group. Immunohistochemical analysis for SFRP1 was used to analyze the correlation between the expression of SFRP1 and the severity of CP based on staining intensities. Results The concentration of SFRP1 in GCF samples taken from of the CP group (281.07 ng·L-1±33.37 ng·L-1) was signifi-cantly higher than that in samples taken from the control group (245.30 ng·L-1±35.69 ng·L-1) (P<0.05). A significant positive correlation was observed between the concentration of SFRP1 in GCF and CAL (r=0.651, P<0.001). Furthermore, the SFRP1 scores in the CP groups (4.500±0.913) were significantly higher than those in the control group (2.800±1.135) (P<0.001). SFRP1 scores did not vary significantly among the CP subgroups (P>0.05). Conclusion SFRP1 expression in the CP groups was significantly higher than that in the control group. Thus, SFRP1 may play a significant role in the development of CP.
4.Expression of the transforming growth factor beta-induced gene in human corneal tissue and cell in vitro

Background Clinical studies indicated that the pathogenesis of most corneal dystrophy is associated with the mutation of the transforming growth factor beta-induced (TGFBI) gene.However,the molecular mechanism of mutated TGFBI gene in corneal dystrophy is unclear. Objective The present study was to investigate the expression of the TGFBI gene in human corneal tissue and cells in vitro.MethodsHuman corneal epithelial cells and keratocytes were cultured and passaged,and donor corneal tissue was obtained for the section preparation.RT-PCR was used to detect the expression of TGFBI mRNA in human corneal tissue and cells.Immunofluorescence was used to test the expression of the TGFBI protein in the human corneal tissue,and immunohistochemistry was used to test the expression of the TGFBI protein in human corneal epithelial cells and corneal stromal cells.ResultsRT-PCR analysis showed that TGFBI mRNA could be detected as a 1274 bp band in human corneal tissue and corneal stromal cells,but no TGFBI mRNA was observed in corneal epithelial cells.Immunofluorescence assay revealed that corneal stromal cells were positive ly expressed for the TGFBI protein,but the corneal epithelial cells did not express the TGFBI protein.Immunohistochemistry indicated that the expression of TGFBI was detected the red fluoressence in the cytoplasm of corneal stromal cells;however,no positive response was found in corneal epithelial cells.ConclusionsThe expression of the TGFBI gene occurs in human corneal stromal cells but not in the corneal epithelial cells.This result might be of helpful for studying the function and role of TGFBI gene in pathogenesis of corneal dystrophy.
5.Accommodative response and nearwork-induced transient myopia induced by simplified and traditional Chinese characters

Background Epidemiologic studies found that the incidence of myopia is higher in Hong Kong and Taiwan regions of China than that of the mainland.So whether the general reading words with traditional Chinese characters and simplified characters is associated with myopia deserves attention.Objective This study was to test the accommodative responses and the regression levels of nearwork-induced transient myopia (NITM) induced by traditional complex Chinese characters and modern simplified ones in the same size,and to explore the differences and inherent relationship of the accommodative regulations based on the structures of the two types of Chinese character in causing reading triggered myopia.Methods Twenty two volunteers aged 24-29 years were included in this study with informed consent.The corrected vision of both eyes from each subject was ≥ 1.0 with a mean spherical equivalence of (-1.86±2.34)D.Accommodative response was tested with 4 different reading texts using the rapid sequence visual presenting model with the GRAND SEIKO-WV5500 infrared autorefractor,and this procedure was performed after full correction of refractive error.An initial test of looking at a certain distance was performed (as baseline),and then the subjects read intensively at the targets for 10 minutes at 33 cm to calculate the accommodative responses.After a 10 second pause,the ocular refractive status was obtained exactly at 15 seconds,20 seconds.The one-way ANOVA method was used to determine the effects of the different font types and sizes on the adjustive responses and the causation of NITM.Results Accommodative response induced by simplified and traditional Chinese characters showed an accommodative lag of (1.11 ±0.38),(0.95 ±0.43),(1.18 ±0.33) and (1.06±0.28) D,showing a significant difference among the 9 pt and 12 pt simplified and traditional Chinese characters (F =1.62,P =0.19),and significantly different accommodative lag values between 12 pt simplified characters and 9 pt traditional characters was found (t =5.56,P =0.02).NITM induced by the four different targets were (-0.45 ±0.45),(-0.47 ±0.46),(0.45 ±0.82) and (-0.46±0.78) D in the 4 types of characters,without a significant difference among them (F=0.01,P =0.99).Conclusions Near-distance reading causes accommodative lag regardless of the type of reading texts.The target demonstrated stimuli spatial frequency and font size play an impact on accommodative responses.NITM appears when one reads simplified or traditional Chinese for 10 minutes.The accommodative lag and NITM trend might be responsible for the onset or regression of myopia,yet it is not supportive for the hypothesis that reading traditional Chinese causes more strain since there is no difference between the two.
6.Evaluation of the effectiveness of health education on iodine deficiency disorders in Hebei province in 2009

Objective To evaluate the effectiveness of health education on iodine deficiency disorders (IDD) in Hebei, and to provide basic information for development of control strategies. Methods A total of 34 project counties were selected in Hebei province, and 3 townships were chosen in each project county. Heath educational activities were carried out in the classes of grade 5 in the central primary school of each selected township. In the meantime, 3 villages were chosen in each selected township where the health education for women of childbearing age in the community was carried out. Sureys on knowledge of IDD control were conducted in the 34 project counties before and after the health educational activities. Results The knowing rates of IDD control among pupils in all 34 project counties increased from 71.10%(7835/11 019) to 94.84%(10 840/11 430) after health education, with a average increase of 23.74%. The knowing rates of IDD control among housewives increased from 77.02%(4531/5883) to 95.32%(5902/6192), with a average increase of 18.30%. Of which, the knowing rates of IDD control among pupils in Hengshui city increased from 55.56% (750/1350) to 94.89% (1281/1350),with a average increase of 39.33%. The knowing rates of IDD control among housewives in Handan city increased from 65.68%(532/810) to 96.50%(909/942), with a average increase of 30.82%. Conclusions The knowing rates of IDD control among pupils and housewives are remarkably increased after implementing the health education projects. They have better life and health habits, and the project achieves desired effect.
7.Value of cardiac CT examination in middle-aged and elderly patients with atrial septal defect before planned transcatheter closure

Objective To investigate the value of the cardiac CT examination for decision making in middle-aged and elderly patients before planned transcatheter atrial septal defect (ASD) closure. Methods Cardiac CT was performed in 63 adult patients [18 males, aged from 50 to 77 years, mean age (56. 87 ±5.79) years]with ASD before planned transcatheter ASD closure. Coronary CT angiography was made for detection of associated cardiovascular diseases, followed by 3D reconstruction of ASD for determination of the defect size in the GE-workstation, results were compared between transthoracic echocardiography measurement, CT measurement, and atrial septal defect occluder waist diameter. Results Cardiac CT identified additional cardiovascular diseases in 14 patients and decision making was changed based on cardiac CT results. Coronary artery stenosis was detected in 8 patients by cardiac CT, and proved by coronary angiography, and all of them were given comprehensive management: percutaneous coronary intervention and thanscatheter ASD closure were successively performed in 2 cases, and 1 case was referred to surgery for both coronary artery bypass graft and surgical ASD repair, and 5 patients were given pharmacological management for coronary artery disease besides thanscatheter ASD closure. Cardiac CT identified large ASD with insufficient rim tissue in 2 cases and transcatheter closures were abandoned.Cardiac CT screened out 1 case from those with insufficient posterior inferior rim by transthoracic echocardiography, and transcatheter ASD closure was successfully pedormed. Cardiac CT ruled out ASD in 1patient. In addition, cardiac CT detected 1 partial abnormalous pulmonary vein connection and 1 ductus arteriosus in this cohort. A correlation on ASD measurements was found between CT size and TTE size(r =0. 80,P < 0. 01 ; Y =0. 84X + 8. 85, R2 =0. 63, P < 0. 05 ), and between ASO size and CT size ( r =0. 92,P < 0. 01 ; Y =0. 93X + 4. 78, R2 =0. 84,P < 0. 05 ). Conclusion In middle-aged and elderly patients with ASD for possible transcatheter closure, cardiac CT is valuable on determing ASD size and morphology and could provide incremental information for optimizing clinical management for ASD patients.
8.Analysis of complications during and post interventional therapy of congenital heart disease

Objective To analyze the incidence and cause of complications during and after interventional therapy for congenital heart disease (CHD). Methods From April 1986 to April 2009, 388 out of 6029 patients with CHD developed complications during and post interventional therapy, another 5 patients died post procedure, clinical data from these 393 patients were retrospectively analyzed. The patients with severe functional insufficiency requiring intervention or surgery during and after interventional therapy were classified as severe complications. Results The overall complication rate was 6. 44% [7.69% post atrial septal defect occlusion, 4.20% post patent ductus arteriosus (PDA) occlusion, 1.31% post percutaneous balloon pulmonary valvuloplasty, 14.94% post veatricular septal defect occlusion, 3.13% post percutaneous closure of aortopulmonary collaterals, 30.95% post catheter embolotherapy of pulmonary arteriovenous malformations, 12.50% post transcatheter closure of coronary artery fistulae, 20.00% post transcatheter closure of ruptured sinus of Valsava aneurysm, 66. 67% post percutaneous balloon aortic valvuloplasty]. The severe complication rate was 0.65% (39/6029). The procedure-related mortality rate was 0.08% (5/6029), 0.26% (2/761) post percutaneous balloon pulmonary valvuloplasty, 0.05% (1/2070)post PDA occlusion, 9.10% (1/11) post balloon atrial septostomy, 33.33% (1/3) post percutaneous balloon aortic valvuloplasty. Emergency Cardiovascular surgery rate was 0.22% (13/6029). Selective surgery was required in 0.13% (8/6029)of patients post procedure. Two patients (0.03%) received permanent pacemaker implantation. Conclusions The severe complications and mortality rate of interventional therapy for CHD are relative low. Post procedure follow-up is needed fro monitoring possible procedure-related complications.
9.Effects of paecilomyces cicadidae total polysaccharides on non-specificity immune regulation in rats

AIM: To study the immune regulation of paecilomyces cicadidae total polysaccharides. METHODS: After subcutaneous injection with 200 mg/L paecilomyces cicadidae total polysaccharides in the back of the rats for 17 days, the white blood cell (WBC) counts, and the levels of lipid peroxide (LPO) and reduced glutathione (GSH) in the spleen and thymus of rats were detected. The alveolar macrophages (AM?) cultured with 10% fetal bovine serum-RPMI-1640 for 2 h in vitro , then the activity of lactate dehydrogenase (LDH) and acid phosphatase (ACP) were detected by automatic biochemistry analyzer, and the ability devouring neutral red in the AM? were examined. RESULTS: Compared with control group, the WBC, the levels of GSH in the spleen and thymus, and the activities of LDH and ACP were significantly increased and the ability of devouring neutral red was also strengthened( P
