1.Multidrug resistance reversal effect of tenacissoside I through impeding EGFR methylation mediated by PRMT1 inhibition.

Cancer multidrug resistance (MDR) impairs the therapeutic efficacy of various chemotherapeutics. Novel approaches, particularly the development of MDR reversal agents, are critically needed to address this challenge. This study demonstrates that tenacissoside I (TI), a compound isolated from Marsdenia tenacissima (Roxb.) Wight et Arn, traditionally used in clinical practice as an ethnic medicine for cancer treatment, exhibits significant MDR reversal effects in ABCB1-mediated MDR cancer cells. TI reversed the resistance of SW620/AD300 and KBV200 cells to doxorubicin (DOX) and paclitaxel (PAC) by downregulating ABCB1 expression and reducing ABCB1 drug transport function. Mechanistically, protein arginine methyltransferase 1 (PRMT1), whose expression correlates with poor prognosis and shows positive association with both ABCB1 and EGFR expressions in tumor tissues, was differentially expressed in TI-treated SW620/AD300 cells. SW620/AD300 and KBV200 cells exhibited elevated levels of EGFR asymmetric dimethylarginine (aDMA) and enhanced PRMT1-EGFR interaction compared to their parental cells. Moreover, TI-induced PRMT1 downregulation impaired PRMT1-mediated aDMA of EGFR, PRMT1-EGFR interaction, and EGFR downstream signaling in SW620/AD300 and KBV200 cells. These effects were significantly reversed by PRMT1 overexpression. Additionally, TI demonstrated resistance reversal to PAC in xenograft models without detectable toxicities. This study establishes TI's MDR reversal effect in ABCB1-mediated MDR human cancer cells through inhibition of PRMT1-mediated aDMA of EGFR, suggesting TI's potential as an MDR modulator for improving chemotherapy outcomes.

2.Establishment and application of colloidal gold double-antibody sandwich assay for detection of Getah virus

Getah virus(GETV)is an arbovirus capable of infecting humans and vertebrates such as horses and pigs via blood-sucking mosquitoes,which is extremely harmful to the livestock indus-try.Current monitoring methods are time-consuming,costly,and dependent on specialized instru-ments.These characteristics hinder rapid detection in clinical samples.Therefore,the development of a simple,rapid,specific,and sensitive method for detecting GETV antigen is crucial for the pre-vention and control of GETV.In this study,a GETV E1 monoclonal antibody strain SD17/09-E1-mAb was prepared by a prokaryotic expression system for GETV E1 protein expression,and a col-loidal gold double-antibody sandwich assay encapsulating two strains of GETV E1 monoclonal an-tibody wasestablished.The results showed that the prepared colloidal gold test strips had good sen-sitivity and did not cross-react with other common porcine virus-positive tissue samples;the test strips had a high compliance rate with the IFA assay for GETV,and could be stored at 37 ℃ for one month and at room temperature for at least three months.In this study,a colloidal gold anti-surveillance test strip for rapid detection of GETV was successfully prepared,which provides a powerful tool for GETV detection.

4.Quality assurance test cases for stereotactic radiation therapy planning of multiple intracranial metastases

Objective:To present a set of clinically representative quality assurance (QA) test cases for stereotactic radiosurgery (SRT) plans of multiple intracranial metastases, in order to assess the plan quality and machine execution capabilities.Methods:Based on the clinical characteristics of multiple brain metastases, four groups of test cases with three target volumes (TVs), six TVs, nine TVs, and TVs near organs at risk (OARs) were designed. For these cases, SRT plans were developed, and plan quality was assessed using metrics including the Radiation Therapy Oncology Group conformality index (RTOG CI), gradient index (GI), homogeneity index (HI), and the volume of normal brain tissue receiving a dose of 24 Gy ( V24 Gy), which was defined as the volume enclosed by the 24 Gy isodose line around the Brain-PTV ( V24 Gy of Brain-PTV). Verification plans were generated for each test case, including the verification of point doses, planar doses (PD), and SRS MapCHECK (SMC) semiconductor matrix planar doses. Compared with the calculated result of the treatment planning system (TPS), the criteria for the γ analysis of planar doses were set at 1 mm/2% and 2 mm/2%. Results:For the four groups of test cases, the mean CI, GI, HI, and V24 Gy of Brain-PTV were 1.04±0.03, 3.79±0.40, 0.73±0.01 and (7.46±3.80) cm 3, respectively. The mean deviations of the point doses were 0.88%±0.98%, 1.47%±0.79%, 1.52%± 0.76%, and 1.17% ± 0.38%, respectively. The mean γ passing rates of the single fields for PDs were greater than 98% at 2 mm/2% and exceeding 96% at 1 mm/2%, and the mean γ pass rates of the SMC semiconductor matrix for PDs were 97.75% ± 2.31% and 99.33% ± 0.62%, at 1 mm/2% and 2 mm/2% respectively. Conclusions:The proposed QA test cases for SRT of multiple intracranial metastases allow for the effective assessments of the plan quality and machine execution capabilities and, thus, can assist various centers in clinical applications.

7.Scoping review of frailty assessment tools for stroke patients

Objective To provide a reference for the selection of frailty assessment tools for stroke patients by conducting a literature review of existing frailty assessment tools for stroke patients. Methods Systematic searches were conducted in PubMed, Web of Science, EMbase, SinoMed, Wanfang, VIP, and China National Knowledge Infrastructure(CNKI) databases from their inception to May 2023. The literature that met the research purpose and question were screened, and the basic information of each article, including the first author, publication year, country of publication, type of research design, name of the assessment tool, dimensions of scale, the number of items, and assessment time, was extracted. Results A total of 1, 729 articles were retrieved in this study. After multiple screenings, 22 articles (including 8 frailty assessment tools) were ultimately included. The analysis results showed that the evaluation indicators, evaluation time, and evaluation methods of the 8 frailty assessment tools were different and had their own advantages and disadvantages. Among them, the frailty index was the most commonly used frailty assessment tool for stroke patients. The selection of effective frailty assessment tools for stroke patients is essential for strengthening prestroke risk stratification and improving poststroke outcomes. Conclusion There are significant differences among different frailty assessment tools, and existing assessment tools have limitations in measuring frailty in stroke patients. Comprehensive assessments should be conducted in combination with clinicians' experience and judgment. New frailty assessment tools are needed in future research to better guide the rehabilitation treatment and management of stroke patients.
8.Simulation and optimization of integrated axial flow blood pump system

Given that current axial flow blood pumps have certain structural defects,resulting in poor performance and inferior blood compatibility,an integrated axial flow blood pump is developed,and its geometry is optimized using computational fluid dynamics method.The study also investigates the effects of different operational parameters on the performance of the blood pump and compares with experimental data to determine the optimal conditions.Additionally,the flow patterns of the blood pump are comprehensively analyzed for further revealing the internal flow phenomena,and the behavior of red blood cells in the blood flow and their response to shear stress are simulated using discrete phase model to evaluate the blood compatibility of the blood pump.The study shows that the novel blood pump performed well in terms of head.Under the optimal condition with a rotational speed of 9 000 r/min and a flow rate of 6.24 L/min,the blood pump improves the head by 16%as compared with the original structure,and reaches 25%efficiency,which can meet the physiological needs of most people.The pressure gradient and velocity gradient in most areas within the blood pump are smooth,and the internal flow patterns are generally stable,effectively avoiding the occurrence of hemolysis.The optimized blood pump can ensure high-level performance and favorable flow field characteristics while maintaining superior blood compatibility,which provides important reference for the structural optimization of axial flow blood pumps.
9.Establishment and preliminary application of prokaryotic expression of BTV16 re-combinant VP2 protein,preparation of polyclonal antibody and indirect ELISA detection method

Bluetongue virus(BTV)is classified as a category Ⅱ animal epidemic disease in China,infecting multiple species and posing significant threats to the ruminant breeding industry.There are 29 serotypes of BTV,with BTV16 being one of the major serotypes currently prevalent in Chi-na.Bluetongue virus infection mainly manifests as a latent infection,making the establishment of ELISA assays crucial for epidemiological detection.In this study;the expression of the BTV16 VP2 protein was achieved using a prokaryotic expression system,and polyclonal antibodies were pre-pared using BALB/c mice.An indirect ELISA assay using VP2 protein as the encapsulated antigen was established and optimized.Clinical samples from the Guangxi Zhuang Autonomous Region were tested and analyzed for compliance with commercial kits.The results showed that the BTV16 VP2 protein was successfully expressed and purified,and the prepared polyclonal antibody exhibi-ted good immunogenicity.The ELISA assay had good specificity,with no cross-reactivity against ruminant diseases such as AKAV,FMDV and GETV.The critical values for negativity and positivity were determined to be 0.314,and the coefficients of variation(Cv)between batches and within batches were both less than 5%,indicating good reproducibility.The ELISA assay revealed a positive rate of 92.4%for 79 samples from the Guangxi Zhuang Autonomous Region,with a compliance rate of 98.7%when compared to the commercialized kit.In conclusion,this study suc-cessfully established an indirect ELISA method for BTV16,facilitating the detection of bovine clin-ical samples.
10.Text Analysis of Financial Compensation Policies for Public Hospitals in China from the Perspectives of Policy Tools and Regional Differences

Objective It analyzes the financial compensation policies for public hospitals in China from 2009 to 2023,aiming to reveal the characteristics of policy issuance,explore the issues in the use of policy tools across different regions,and provide suggestions for policy optimization.Methods Based on the theory of policy tools,a three-dimensional analytical framework combining policy tools,policy objectives and economic regions was constructed.Content analysis was used to conduct quantitative analysis of the 223 policy texts included,resulting in 3142 valid codes.Results The number of policy documents released over time generally showed a"double-peak"pattern.In terms of the policy tool dimension,the supply-oriented,environment-oriented,and demand-oriented policy tools accounted for 50%,39.21%,and 10.79% respectively.Within the policy objective dimension,the improvement of relevant systems accounts for the highest proportion (33.16%),while subsidies for public health services are the least utilized (8.50%).In the economic regions,the eastern region has the highest number of policy documents (94),while the northeastern region has the lowest (12).The four major regional policy tools are mainly supply-oriented,and there are differences in the use of financial support,government purchase and other tools.Conclusion Relevant departments should strengthen their attention and actively innovate local policies.There is a need to optimize the internal and external structures of the three major policy tools to enhance balanced utilization. Furthermore,increased attention should be paid to the objective of compensation for public health services within policies.Financial investment in central and western regions should be further intensified.
