1.Relationship Between Quadriceps Micro-Perfusion Assessed by IVIM and Muscle Strength After Low-Load Resistance Training in Healthy Volunteers

Purpose To investigate the changes in quadriceps femoris microcirculatory perfusion level after low-load blood flow restriction training and its relationship with muscle strength.Materials and Methods Twenty-five healthy subjects were prospectively recruited in the First Affiliated Hospital of Kunming Medical University from September to November 2022.A 200 mmHg pressure cuff was applied at the root of the left thigh for blood flow restriction,and the subjects completed regular knee extension training within 4 weeks.Before the first training session and within 24 hours after the last training session,all subjects underwent scanning with the 3.0T MRI intravoxel incoherent motion sequence and the multi-echo steady-state acquisition three-dimentional imaging sequence.After image post-processing,the quadriceps femoris cross-sectional area,perfusion fraction and pseudo-diffusion coefficient were obtained,and the peak torque was measured using an isokinetic dynamometer.The MRI and muscle strength test parameters before and after training were compared,and correlation analyses were performed between the change of peak torque and the change of perfusion fraction,cross-sectional area,and pseudo-diffusion coefficient respectively.Results After low-load blood flow restriction training,the cross-sectional area of the left quadriceps femoris in subjects increased(t=-4.515,P<0.05).Among its components,the cross-sectional area of the left rectus femoris,vastus intermedius and vastus lateralis all increased(t=-3.302,-2.877,-3.207,all P<0.05).The perfusion fraction value of the left quadriceps femoris increased(t=-5.447,P<0.05);the perfusion fraction values of the left rectus femoris,vastus intermedius,vastus lateralis and vastus medialis all increased(t=-5.723,-4.621,-3.767,-4.682,all P<0.05);the muscle strength of the left quadriceps femoris increased(t=-3.983,P<0.05).There was a highly positive correlation between change of perfusion fraction and peak torque of the left quadriceps femoris in subjects(r=0.708,P<0.05).Conclusion After low-load blood flow restriction training,the changes in quadriceps femoris muscle microperfusion quantified by intravoxel incoherent motion are related to muscle strength,which is helpful for formulating rehabilitation training strategies for young patients.

3.A qualitative study of experiential health education on complications among middle-aged and elderly hypertensive patients

Objective To explore the experience and feeling of experiential health education for complications in middle-aged and elderly hypertensive patients,and to provide basis for health education model for hypertensive patients. Methods By objective sampling method,10 middle-aged and elderly hypertensive patients who received experiential health education for hypertension complications in Ruikang Hospital Affiliated to Guangxi University of Chinese Medicine from August to September 2023 were selected for semi-structured interviews,and the interview data were sorted out by Colaizzi seven-step analysis method. Results Middle-aged and elderly hypertension patients with complications of experiential health education experience can be summarized as two themes,seven subthemes,namely:the perceived benefits (improve the cognition of hypertension,complications of experience,behavior change and cognitive clarity,promote interpersonal communication),challenges (the lack of early cognitive,the limitations of traditional health education,body burden concerns). Conclusion Worry about long-term complications and lack of knowledge of disease management are common after receiving experiential health education on complications. Therefore,when designing the experiential health education program,clinical medical staff should fully consider the physiological and psychological characteristics of patients,provide personalized support,and gradually guide patients to adapt to the education process.

5.Validation of microfluidic chip for detection of mutation hotspots associated with hereditary hearing loss

Objective:To explore the application of microfluidic chip in detection of hereditary deafness-associated hotspot mutations.Methods:The dedicatedly designed and fabricated microfluidic chip was integrated with kompetitive allele-specific polymerase chain reaction amplification system, scanned via laser-excited confocal fluorescence scanner, and finally analyzed programmatically to acquire the typing results of the 23 mutation sites of the four common genes associated with hereditary hearing loss. Dried blood spots were collected from 276 neonates containing the 131 cases with hearing loss who were born in 2019 in Chengdu (deafness group) and the 145 cases with normal hearing who were born in 2020 in Chengdu (control group), and analyzed by the microfluidic chip to evaluate its clinical performance.Results:By cluster analysis, the microfluidic chip correctly analyzed the 23 positive reference samples and acquired the same typing results as their actual results, with a limit of detection of 1 mg/L. For the 276 newborn blood spots, the detection results of the microfluidic chips were confirmed to be correct by the contrasting methods. Among Deafness Group, 66 (50.4%) tested positive for the selected 23 mutation hotspots; among Control Group, 40(27.6%) were positive. Among these mutations, c.109G>A of the GJB2 gene was the most prevalent one, whose carrier rate in deafness group and control group were 46.6%(61/131) and 23.4% (34/145), respectively.Conclusions:The micro-fluidic chip system was succeeded in fulfilling the hereditary deafness-related mutation detection, and offered many advantages including high specificity, avoiding the amplicon carryover contamination, simplifying the entire experimental operation process and short detection time, so as to better meet the detection requirement of genetic testing for deafness in newborn screening and other fields.
6.Simultaneous Determination of 9 Components in Huoxiang Zhengqi Oral Liquid by HPLC

OBJECTIVE: To establish a method for simultaneous determination of nine components in Huoxiang zhengqi oral liquid, and to improve and perfect the quality standard of Huoxiang zhengqi oral liquid. METHODS: The contents of nine components in 10 batches of Huoxiang zhengqi oral liquid were determined by HPLC, such as licorice coumarin, isorlicin, liquiritinapioside, narirutin, liquiritin, saponins, hesperidin, magnolol and honokiol. The determination was performed on Kromasil Eternity XT-5-C18 column with mobile phase consisted of acetonitrile-0.05% phosphoric acid solution (gradient elution) at the flow rate of 1 mL/min. The detection wavelength was set at 220 nm, and column temperature was 25 ℃. The sample size was 10 μL. RESULTS: The linear range of licorice coumarin, isorlicin, liquiritinapioside, narirutin, liquiritin, saponins, hesperidin, magnolol and honokiolin were 0.000 5-0.007 5, 0.000 8-0.025 0, 0.006 1-0.976 0, 0.001 6-0.250 0, 0.007 8-0.025 0, 0.000 4- 0.062 7, 0.008 6-0.276 0, 0.010 0-0.500 0, 0.010 0-0.500 0 mg/mL (r=0.999 2-1.000 0). The detection limits were 0.001 3, 0.000 1, 0.004 7, 0.005 0, 0.012 0, 0.001 3, 0.007 8, 0.007 7 0, 0.005 8 μg/mL, and the quantitative limits were 0.013 0, 0.000 8, 0.047 0, 0.050 0, 0.120 0, 0.013 0, 0.078 0, 0.070 0, 0.058 0 μg/mL, respectively; RSD of precision, stability and repeatability tests were less than 3.0% (n=6). Average recovery rates were 98.67%, 101.85%, 98.97%, 103.05%, 100.00%, 97.78%, 97.91%, 100.13%, 101.95%; RSDs were 1.14%, 2.18%, 0.40%, 0.17%, 1.38%, 0.85%, 1.38%, 0.10%, 1.35% (n=6). CONCLUSIONS: The established method is accurate and reliable, which can provide reference for the establishment of the overall quality control evaluation system and the improvement of quality standard for Huoxiang zhengqi oral liquid.
7. Cortisol and inflammatory factors in patients with occupational noise-induced hearing loss and their clinical significance

OBJECTIVE: To investigate the levels of cortisol and inflammatory factors and their influencing factors in patients with occupational noise-induced hearing loss(ONID). METHODS: A total of 106 ONID patients were selected as the ONID group, and 50 healthy participants without noise exposure were selected as the control group by judge sampling method. The levels of salivary cortisol in the two groups were detected by enzyme-linked immunosorbent assay.The serum levels of cortisol, tumor necrosis factor alpha(TNF-α), interleukin-6(IL-6) and C-reactive protein(CRP) in the peripheral blood were detected by electrochemiluminescence assay. Tinnitus Handicap Inventory was used to evaluate the disability levels of tinnitus in ONID patients. RESULTS: The level of salivary cortisol in the morning and in the nighttime, and cortisol, TNF-α, IL-6 in the serum were higher in the ONID group compared with that in the control group(P<0.05). There was no significant difference in the serum level of CRP between these two groups(P>0.05). Spearman correlation analysis results showed that the level of cortisol in the saliva and in the serum was not correlated with TNF-α and IL-6(P>0.05). Analysis of multiple linear regression showed that the levels of serum cortisol and salivary cortisol in the morning in the ONID patients were positively correlated with noise exposure level(P<0.05). The level of TNF-α in the serum was positively correlated with tinnitus score(P<0.05). CONCLUSION: Hyperactivity of hypothalamus-pituitary-adrenal axis and inflammation activation may exist in patients with ONID. The cortisol can be used as a biomarker for the effect of noise-stress.

Malaria is one of the major diseases threatening the lives of people in Africa. Over the past decade, China has provided anti-malaria assistance to Africa. In November 2007, fast control malaria team of Guangzhou University of Chinese Medicine cooperated with the Comoros health authority. In Moheli Island of the Comoros, the anti-malaria team adopted a new strategy of Mass Drug Administration (MDA) and active intervention. They established an effective anti-malaria system and reporting system, as well as a local anti-malaria team. Furthermore, they treated patients with malaria in Moheli Island and implemended health education in local people. In a very short time, they have achieved remarkable results. This paper summarized the experiences of the project in order to provide useful reference for China to better carry out anti-malaria action in Africa and expand foreign health assistance.
9. An investigation of trichloroethylene-induced effects on histone methylation in L-02 hepatic cells

Objective:
To further explore TCE-induced hepatotoxicity and its mechanisms by identification of trichloroethylene (TCE) induced abnormal histone methylation in human liver cells.
Methods:
L-02 cells were treated with 0 and 8 mmol/L TCE for 24 h. Histones were extracted by acid. Liquid chromatography electrospray ionization tandem mass spectrometry (ESI-LC-MS/MS) were used to identify and quantify TCE related histone methylations. TCE induced abnormal methylation of H3K79 me2 and H3K79 me3 were validated by Western blot analysis. The further analysis of the function of histone abnormal methylation modifications were done by single cell gel electrophoresis (SCGE) and Western blot analysis of p53 and ɤH2AX.
Results:
After treatment with TCE for 24 h in L-02 cells, the 36 TCE related histone methylation sites in 28 peptide segments were identified by MS. After treatment with TCE in concentrations of 0 and 8.0 mmol/L in L-02 cells for 24 h, the relative expression level of histone H3K79 me3 were 1.00±0.06, 0.70±0.09 (
10.Reconstruction of personalized three-dimensional entity model of hip bone based on reverse engineering and rapid prototyping technology

BACKGROUND:Hip’s anatomy structure is complex, and the traditional two-dimensional imaging, such as X-ray film or CT scan, is susceptible to image overlap and other soft tissue. As a result, there are some limitations in the diagnosis of hip disease. The digital model is built based on reverse engineering and rapid prototyping. It is comprehensive, intuitive and accurate of the stereo configuration of the hip and the spatial relations of other parts’ anatomical structures. This study has a broad application prospect. OBJECTIVE:To reconstruct the personalized three-dimensional entity model of the hip bone with computer aided technology. METHODS: The human hips were scanned with a portable three-dimensional laser scanner, and then the point cloud data of the hips were gotten. The point cloud was processed and the three-dimensional model reconstruction was carried out by using the engineering software Geomagic and the computer aided software CimatronE. Three-dimensional print of hip was completed and the model was gotten, which was consistent with the geometry of hip bone, through the rapid prototyping machine. RESULTS AND CONCLUSION:A model with good geometric similarity and biomechanical characteristics was built by digital modeling method based on reverse engineering and rapid prototyping. This way of digital modeling provided the basis for digital manufacturing of artificial hip, virtual assembly, stress analysis and surgical simulation.
