1.Discussion on the Effects of Yanghe Decoction Containing Serum on Migration and Invasion of Breast Cancer Cells in Tumor Related Fat Microenvironment Based on the JAK2/STAT3 Signaling Pathway

Objective To investigate the effects and mechanism of Yanghe Decoction containing serum on the migration and invasion of BT-549 cells intervened by conditioned medium of cancer-associated adipocytes(CAAcm)based on the JAK2/STAT3 signaling pathway.Methods 3T3-L1 mouse embryonic fibroblasts were induced to differentiate into mature adipocytes using adipogenic induction medium.Yanghe Decoction containing serum was prepared from SD rats.BT-549 cells were co-cultured with mature adipocytes to obtain CAAcm.Transwell assay was used to detect the migration and invasion ability of BT-549 cells before and after intervention with CAAcm.BT-549 cells after CAAcm intervention were divided into blank serum group,positive drug group and Yanghe Decoction low-,medium-and high-dosage groups,and cultured with 10%corresponding serum.Cell proliferation was assessed using the CCK-8 assay,scratch assay was performed to evaluate the lateral migration of BT-549 cells,Transwell assay was conducted to evaluate the vertical migration and invasion ability of BT-549 cells,Western blot was performed to detect the expressions of proteins including JAK2,STAT3,VEGF and HIF-1α.Results CAAcm intervention had a promoting effect on the migration and invasion of BT-549 cells.Compared with the blank serum group,the growth inhibition rate of BT-549 cells significantly increased in Yanghe Decoction low-,medium-and high-dosage groups(P<0.05,P<0.01),the scratch healing rate was significantly decreased(P<0.05,P<0.01),the number of migrating cells and invading cells significantly reduced(P<0.01),and the expressions of JAK2,STAT3,VEGF and HIF-1α proteins significantly decreased(P<0.05,P<0.01).Conclusion Yanghe Decoction containing serum can inhibit the proliferation,migration and invasion of BT-549 cells intervened by CAAcm,and the mechanism may be related to the inhibition of JAK2,STAT3,VEGF and HIF-1α protein expressions.
2.Correlation analysis of WT1,BCR-ABL genes with IL-6,VEGF,PC and PS in myeloproliferative neoplasms

Objective To investigate the correlation between WT1,BCR-ABL genes and interleukin-6(IL-6),vascular endothelial growth factor(VEGF),plasma protein C(PC),plasma protein S(PS)in patients with myeloproliferative neoplasms(MPN).Methods A total of 54 MPN patients treated in this hospital from January 2021 to January 2024 were selected as the MPN group,and 54 healthy subjects undergoing physical examination during the same period were selected as the control group.The gene relative expression levels of WT1,BCR-ABL and levels of IL-6,VEGF,PC,and PS were detected and compared between the two groups.The correlations between WT1,BCR-ABL gene relative expression levels and IL-6,VEGF,PC,PS levels were analyzed,and the diagnostic efficacy of WT1 and BCR-ABL genes for MPN was evaluated by receiver operat-ing characteristic(ROC)curve.Results Compared with the control group,the gene relative expression levels of WT1,BCR-ABL and the levels of IL-6 and VEGF in the MPN group were higher,while the levels of PC and PS were lower,with statistically significant differences(P<0.05).Pearson correlation analysis showed that WT1 and BCR-ABL gene relative expression levels were positively correlated with IL-6 and VEGF levels,and negatively correlated with PC and PS levels(P<0.05).The combined use of WT1 and BCR-ABL genes has higher diagnostic efficacy for MPN than single indicators,with an area under the curve(AUC)of 0.833 and a sensitivity of 88.69%.Conclusion The relative expression levels of WT1 and BCR-ABL genes are correlated with IL-6,VEGF,PC,and PS levels in MPN patients.The combination of WT1 and BCR-ABL genes can effec-tively diagnose MPN.

OBJECTIVE Aimed to investigate the impact of comorbid asthma on chronic rhinosinusitis with nasal polyps(CRSwNP)and identify key proteins and signaling pathways.METHODS Proteomic methods were employed to analyze differentially expressed proteins(DEPs)in nasal lavage fluid(NLF)from control,CRSwNP,and CRSwNP with asthma groups.DIA quantitative analysis technology was used to assess the gradient changes of DEPs among the three groups to determine key proteins affected by comorbid asthma in CRSwNP.RESULTS Compared to the control group,1 377 and 1 006 DEPs were identified in the CRSwNP and CRSwNP with asthma groups,respectively.Peroxiredoxin-5(PRDX5),Ran-Binding Protein 1(RanBP1)(upregulated),and Keratin 9(KRT9)(downregulated)were identified as key proteins affecting CRSwNP with asthma.CONCLUSION Comorbid asthma may promote the occurrence and development of nasal polyps through specific key proteins and signaling pathways,providing new molecular insights into the interaction between CRSwNP and asthma.
5.A clinical research of endoscopic submucosal dissection for ileocecal valve lipoma

Objective:To evaluate the efficacy and safety of endoscopic submucosal dissection (ESD) for the treatment of ileocecal valve lipoma.Methods:A retrospective cohort study was performed on data of ileocecal lipoma patients who underwent ESD at the Endoscopy Center of Zhongshan Hospital, Fudan University from December 2013 to June 2023. According to the lesion location, the patients were divided into ileocecal valve group and cecum group. The operation time, operation speed, en bloc resection rate, complications, and follow-up outcomes between the two groups were compared.Results:A total of 59 patients with ileocecal lipoma were enrolled, including 31 patients in the ileocecal valve group and 28 patients in the cecum group.There were no significant differences in gender, age, specimen size, or lesion size between the two groups ( P>0.05). Lipomas in both the ileocecal valve group and the cecum group were successfully resected by ESD. The en bloc resection rates were 100.0% (31/31) and 92.9% (26/28) respectively, and the difference was not statistically significant ( χ2=0.033, P=0.133). Median operative duration significantly differed between the two groups ( ileocecal valve group 26 min VS cecum group 20 min, Z=-0.136, P=0.027), as did resection speed (ileocecal valve group 0.14 cm2/min VS cecum group 0.24 cm2/min, Z=-0.223, P=0.022). Adverse events included one postoperative fever in the ileocecal valve group and one delayed bleeding in the cecum group. During the median follow-up of 38 months (7-106 months), there was no case of residual tumor or recurrence. Conclusion:Despite technical challenges in ESD of ileocecal valve lipoma, it is still a safe, feasible and effective treatment method.

Obstructive sleep apnea hypopnea syndrome(OSAHS)is one of the most common sleep disorders in both adults and the elderly.A large number of studies in recent years have shown that OSAHS may increase the risk of cognitive impairment.This article summarizes the latest research findings on cerebrospinal fluid and plasma biomarkers associated with Alzheimer disease(AD)in patients with OSAHS,as well as how various treatment methods for OSAHS can relief the symptoms of OSAHS and alter the levels of AD biomarkers simultaneously,so as to emphasize the importance of treating OSAHS symptoms to prevent and delay the development and progression of AD.
