Liver diseases, particularly cirrhosis and cancer, significantly threat human health for a long time, and their diagnosis and treatment are important topics in medical research. Tradi-tional diagnostic and therapeutic approaches have focused on biochemical properties of liver diseases, yet often neglecting the mechanical microenvironment of liver at molecular, cellular, and tissue levels. This oversight makes it difficult to meet clinical needs. Recent advancements in biomechanics and mechanobiology have underscored the importance of mechanical properties of liver in understanding disease mechanisms, presenting profound implications for both basic research and clinical practice. However, there is still a lack of in-depth exploration of the mechanical properties of liver in both basic research and clinical treatment, making it unclear of the specific mechanisms and application scenarios. The authors propose and introduce the emerging field of liver mechanomedicine, examine the biomechanical properties of liver and their alterations during disease progression, elucidate mechanobiological mechanisms of cellular mechanical responses and signal transduction, explore the application of mechanical characteristics in the diagnosis and treatment of liver diseases, aiming to provide a new perspective for basic research and clinical practice.

Aim To investigate the effects of sodium lactate(NALA)on right heart failure induced by monocrotaline(MCT)-induced pulmonary arterial hy-pertension in rats and to reveal the underlying mecha-nisms.Methods Forty male Sprague-Dawley(SD)rats were randomly allocated into four groups, with ten rats in each group, namely,MCT group,NALA group, and NALA+MCT group;the MCT and NALA+MCT groups were administered a single intraperito-neal injection of MCT at 60 mg·kg-1 to induce pul-monary hypertension, and one week later, the NALA and NALA+MCT groups received intraperitoneal in-jections of NALA at 0.1 g·kg-1(once a day,for 5 weeks),while the CON and MCT groups received e-qual volumes of physiological saline(once a day,for 5 weeks);right heart function was assessed using echo-cardiography,right ventricular and pulmonary artery remodeling were evaluated via histopathological sec-tions,and the expression levels of ANP,BNP,and in-flammatory factors were measured by ELISA,along with assessments of oxidative stress levels,Western blot detection of the expression levels of proteins in the TLR4/NF-κB signaling pathway. Results Compared to the CON group, the MCT group exhibited increased RVSP and RVHI,decreased right heart function,in-creased collagen fiber deposition, and elevated oxida-tive stress and inflammatory factor expression, and the expression levels of proteins in the TLR4/NF-κB signa-ling pathway increased(P<0.05);compared to the MCT group, the NALA+MCT group showed reduced RVSP and RVHI,improved right heart function,atten-uated pulmonary vascular remodeling, decreased ex-pression of ANP,BNP,inflammatory factors, and H2O2,along with increased antioxidant enzyme expres-sion,and the expression levels of proteins in the TLR4/NF-κB signaling pathway decreased(P<0.05).Conclusion NALA can inhibit right ventric-ular remodeling in rats with pulmonary hypertension, and the underlying mechanism may involve the allevia-tion of inflammatory responses and oxidative stress through the inhibition of the TLR4/NF-κB signaling pathway.

Objective To explore the clinical features, treatment and prognosis of traumatic subdural effusion(TSE)in infants. Methods Data of 51 cases of traumatic subdural effusion in infants admitted to the single center of Dalian Women and Children Medical Center(Group)from February 2013 to February 2020 were retrospectively analyzed, and their clinical manifestations, imaging features, treatment methods and prognosis were summarized and analyzed. Results Fifty-one cases(26 males and 25 females),ranging in age from 1 month to 3 years old of traumatic subdural effusion in infants were reviewed in our hospital, all cases were confirmed by Computed Tomography(CT)examination.31 cases were treated conservatively,29 cases were cured, and 2 cases were treated surgically due to poor conservative treatment.Surgical treatment was performed in 22 cases(including 2 cases who received surgical treatment due to poor conservative treatment). One patient underwent puncture and continuous drainage at the lateral Angle of the anterior fontanelle and was cured.Twenty-one cases underwent cranial drilling, subdural space catheterization for external drainage, and 17 cases(80.95%,17/21)were cured at one time. There were 4 cases(19.05%,4/21)of recurrence after external drainage with catheterization. Two cases were cured by external drainage with Ommaya capsule insertion and intermittent aspiration and fluid drainage. It was changed to subdural peritoneal shunt surgery, and 2 cases were cured after the operation. There was no surgical infection or death in all the children in the group. The median follow-up time ranged from 3 months to 60 months, and the conditions were all stable. Conclusion Traumatic subdural effusion is a common complication after craniocerebral injury in infants and young children.Due to its lack of self-expression,the hidden condition is often ignored. Moreover, the brain tissue of infants and young children is in the growth and development stage, which will affect the development of brain tissue after its onset.

Objective Advances in synthetic DNA technology have made it much easier to fake human DNA samples. There are literature reports that fake human DNA can be synthesized by different methods and implanted in the field to confuse the investigation or mislead the trial. Therefore, distinguishing authentic human DNA from synthetic DNA and performing individual identification has become a critical scientific challenge. Methods We define a novel composite genetic marker(methylation-microhaplotype)by combining CpG sites stably hypermethylated or hypomethylated in natural human DNA and nearby immediately adjacent microhaplotype sites.A total of 19 locis were obtained according to the screening criteria, and a composite detection system for methylation-microhaplotypes was established using MPS technology.Random volunteer DNA samples were extracted and synthetic DNA samples were prepared based on whole genome amplification techniques.Population DNA samples were analyzed to evaluate forensic parameters and methylation variability of the methylation-microhaplotype markers.Comparative analyses of human and synthetic DNA were conducted to assess the markers'ability to discriminate between the two and to detect/type both components in mixed mixed samples. Results The composite detection system composed of 19 locis demonstrated high individual identification ability, achieving a cumulative individual identification probability of 0.999 999 999 996 86.12 hypermethylated locis and 7 hypomethylated locis had relatively stable methylation levels in 57 human DNA samples.According to the allele methylation rate(Ram)value, the system can effectively identify natural and synthetic DNA samples.Meanwhile, for mixed DNA samples, the presence of human and synthetic DNA samples can be found and genotyped. Conclusion Methylation-microhaplotype genetic markers, which can discover human DNA and synthetic DNA and can detect the presence and genotyping of them from mixed samples,is a potential useful tool for forensic DNA analysis.
