Objective:
: The objective of this study was to assess the relationship between spermine synthase (SMS) expression, tumor occurrence, and prognosis in lower-grade gliomas (LGGs).
Methods:
: A total of 523 LGG patients and 1152 normal brain tissues were included as controls. Mann-Whitney U test was performed to evaluate SMS expression in the LGG group. Functional annotation analysis was conducted to explore the biological processes associated with high SMS expression. Immune cell infiltration analysis was performed to examine the correlation between SMS expression and immune cell types. The association between SMS expression and clinical and pathological features was assessed using Spearman correlation analysis. In vitro experiments were conducted to investigate the effects of overexpressing or downregulating SMS on cell proliferation, apoptosis, migration, invasion, and key proteins in the protein kinase B (AKT)/epithelialmesenchymal transition signaling pathway.
Results:
: The study revealed a significant upregulation of SMS expression in LGGs compared to normal brain tissues. High SMS expression was associated with certain clinical and pathological features, including older age, astrocytoma, higher World Health Organization grade, poor disease-specific survival, disease progression, non-1p/19q codeletion, and wild-type isocitrate dehydrogenase. Cox regression analysis identified SMS as a risk factor for overall survival. Bioinformatics analysis showed enrichment of eosinophils, T cells, and macrophages in LGG samples, while proportions of dendritic (DC) cells, plasmacytoid DC (pDC) cells, and CD8+ T cells were decreased.
Conclusion
: High SMS expression in LGGs may promote tumor occurrence through cellular proliferation and modulation of immune cell infiltration. These findings suggest the prognostic value of SMS in predicting clinical outcomes for LGG patients.

5.In vivo and in vitro study on the inhibitory effects and mechanism of aucubin on prostate cancer

OBJECTIVE To investigate the effects of aucubin (AU) on the proliferation and tumor growth of prostate cancer (PC) cells by regulating the protein kinase B (Akt)/murine double minute2 (MDM2)/p53 signaling pathway. METHODS Prostate cancer cell PC3 were separated into control group, 50 μmol/L AU group, 100 μmol/L AU group, SC79 (Akt activator) group (5 μmol/L), and 100 μmol/L AU+SC79 group. The cell cloning and proliferation ability were investigated; the rate of cell apoptosis and the expressions of Akt/MDM2/p53 signaling pathway-related protein were detected. Meanwhile, xenograft tumor models of nude mice were constructed and separated into tumor group, AU group (80 mg/kg), SC79 group (50 mg/kg), and AU+SC79 group (80 mg/kg AU+50 mg/kg SC79), with 10 mice in each group. They were given relevant medicine, once a day, for 21 d. After the last medication, tumor weight was determined, and the expressions of nucleus-associated antigen (Ki-67) and Akt/MDM2/p53 signaling pathway-related protein were detected in tumor tissue. RESULTS In the cell experiment, compared with control group, the cell clonal formation number, proliferation rate and phosphorylation levels of Akt and MDM2 protein in 50 μmol/L AU and 100 μmol/L AU groups were significantly decreased (P<0.05), while the cell apoptosis rate and p53 protein expression levels were significantly increased (P<0.05); however, the change trend of each index in SC79 group was opposite (P<0.05). Compared with 100 μmol/L AU group, the cell clonal formation number, proliferation rate and phosphorylation levels of Akt and MDM2 protein in 100 μmol/L AU+SC79 group were significantly increased (P<0.05), while cell apoptosis rate and p53 protein expression levels were significantly decreased (P<0.05); however, compared with SC79 group, the changing trend of indexes was the opposite (P<0.05). In the in vivo experiment, compared with the tumor group, the tumor mass and Ki-67 positive expression and the phosphorylation levels of Akt and MDM2 protein in nude mice of AU group were significantly decreased (P<0.05), and the expression level of p53 protein was significantly increased (P<0.05), but the changing trend of above indexes of nude mice in SC79 group were opposite (P<0.05). Compared with AU group, the tumor mass, Ki-67 positive expression and phosphorylation levels of Akt and MDM2 protein in tumor tissues of nude mice in AU+SC79 group were significantly increased (P<0.05), while the expression level of p53 protein was significantly decreased (P<0.05); however, compared with SC79 group, the changing trend of above indexes was opposite (P<0.05). CONCLUSIONS AU can inhibit PC cell proliferation and tumor growth by inhibiting Akt/MDM2/p53 signaling pathway.
6.Spectral entropy to evaluate the effectiveness of analgesic sedation in patients with ischaemic stroke

Objective To investigate the reliability and feasibility of spectral entropy in evaluate the effectiveness of analgesic sedation in patients with ischemic stroke.Methods A total of 64 patients with acute ischemic stroke admitted to Hongqi Hospital Affiliated to Mudanjiang Medical University from July 2021 to November 2022 were selected as study objects,and the included patients were divided into control group and experimental group according to random number table method,with 32 cases in each group.Patients in control group adjusted analgesia and sedation regimen according to Richmond agitation-sedation scale(RASS)score and critical-care pain observation tool(CPOT)score.Patients in experimental group adjusted the analgesic and sedation regimen according to the results of spectral entropy.The vital signs,C-reactive protein(CRP),dose of sedative and analgesic drugs and incidence of adverse reactions were compared between two groups.The correlation between spectral entropy and RASS score and CPOT score was used Spearman correlation analysis.Results The spectral entropy values were positively correlated with the RASS score and CPOT score,respectively(r=0.719,0.556,P<0.001).There were no significant differences in mean arterial pressure and percutaneous arterial oxygen saturation between two groups at different time points(P>0.05).The heart rate at T3 in experimental group was significantly lower than that in control group(P<0.05).At T1,T2 and T3,CRP levels in experimental group were significantly lower than those in control group(P<0.05).The dosage of sufentanil and midazolam in experimental group were significantly lower than those in control group(P<0.05).The incidence of adverse reactions in experimental group was significantly lower than that in control group(12.50%vs.34.38%,χ2=4.267,P=0.039).Conclusion Spectral entropy can be used as an objective method to monitor the depth of analgesia and sedation in patients with ischemic stroke,and has a good correlation with RASS score and CPOT score,which can reduce the incidence of adverse reactions,effectively avoid stress reactions,and reduce the application of analgesia and sedation drugs.

Abdominal surgery as a common type of clinical surgery, postoperative treat-ment and rehabilitation has always been the focus of surgeons. Somatostatin drugs are commonly used for postoperative bleeding, portal hypertension bleeding, and prophylactic treatment of com-plications such as acute pancreatitis. However, the standardized application of somatostatin still faces numerous challenges in clinical practice. The famous experts and their teams in the field of digestive surgery, medical insurance expert and lawyer, combined with the experience of their respective centers, under the new national medical insurance policy, explore the significance of treatment and rehabilitation after abdominal surgery and the application strategy of somatostatin drugs from different angles. Professor Zhang Taiping succinctly explained the incidence and harm of gastro-intestinal fistula, pointing out that "how to heal the fistula in a short time" is the main rehabilitation goal and the treatment is mainly comprehensive treatment. Professor Chen Chaowu comprehen-sively introduces diagnostic and treatment processes for several representative cases encountered in clinical practice while sharing his team′s rich clinical experience and posing reflective questions. Professor Li Fei starts with practical cases, focusing on duodenal fistula, including fistula caused by complications after endoscopic retrograde cholangiopancreatography, with special emphasis on Stanford classification and different types of treatment. 8.Correlation between nasal mucosal microbiota diversity and the pathogenesis and prognosis of chronic sinusitis.

Objective:To analyze the correlation between nasal mucosal microbiota diversity and the pathogenesis and prognosis of chronic sinusitis. Methods:A total of 80 patients with chronic sinusitis(CRS) admitted to Second Affiliated Hospital of Suzhou University were selected as the research group, and 80 patients with chronic dacryocystitis and nasal septum deviation without sinus inflammation admitted to our hospital during the same period were selected as the control group, nasal secretory specimens were collected under nasal endoscopic guidance by nasal swab, and matrix-assisted laser desorption ionization classification time mass spectrometry, anaerobic bacterial culture and common bacterial culture were performed to compare the differences in nasal mucosal flora between groups. Patients with chronic sinusitis were followed up for 6 months, and nasal secretions were collected again to detect microflora, and the patients were divided into relapse group(21 cases) and non-recurrence group(59 cases) according to whether the patients had relapse. Compare the diversity of nasal microbiota between groups. Results:There were no significant differences in mean relative abundance(MRA) between the two groups of preoperative phylum Microbacterium verrucobacterium, Cyanobacterium phylum, Phylum Laubia mlaus, Porphyromonas species, Enterococcus species, Fusobacterium species, Enterobacter species, Enterobacter species, Erythrobacterium species, Ralstonia species, Bacteroides and Streptococcus species(P>0.05). The MRA of Acidobacterium, Proteobacteria, Actinomycetes MRA, Moraxia, Cyanobacterium, Corynebacterium and Staphylococcus were significantly lower than those of the control group(P<0.05), and the MRA of Escher-Shigella species, Fusobacterium, Bacteroides, Firmicutes, Neisseria, Pseudomonas, Haemophilus and Lactobacillus was significantly higher than that of the control group(P<0.05), and there was no significant difference in MRA at the level of nasal flora and genus before and after surgery in the relapsed group(P>0.05). The MRA of Bacteroides after surgery was significantly lower in the non-recurrent group than that before surgery(P<0.05), the MRA of Corynebacterium and Actinomycetes was higher significantly than that of preoperative(P<0.05), and there was no significant difference in MRA of other species and phylum(P>0.05). Conclusion:The onset of CRS is related to nasal mucosal dysbacteria, and whether the dysbacteriosis improves after surgery is correlated with the prognosis of patients.

Purpose/Significance To explore the characteristics and clinical significance of differentially expressed genes closely re-lated to HPV E6/E7 by using bioinformatics.Method/Process The cervical tissue and clinical information of cervical cancer in TCGA and GTEx of UCSC are used as the training set.The expression profile chip GSE63514 related to cervical cancer in GEO is used as the validation set.Firstly,the limma package of R software is used to screen DEGs of tumor and normal samples,and Venn map of genes re-lated to E6/E7 protein in MigDB is made.Survival analysis is performed by survival kit and verified by ROC and protein expression lev-els.Secondly,key genes are obtained by copy number variation and methylation correlation.Finally,the specific co-expression network is constructed and enrichment analysis and immune infiltration analysis are performed.Result/Conclusion There are 101 differentially expressed genes related to HPV E6/E7 protein,and three genes are found to have significance after screening,namely E2F1,MCM4 and PCNA.At the same time,it is found that the genes in the specific coexpression network are significantly enriched in the DNA replication and chromosome organization pathways.Immune correlation analysis shows that key genes are significantly associated with CD4 T cells,B cells and neutrophils.DNA replication,chromosome organization,etc.,are the molecular mechanisms and key genes significantly related to the development of cervical squamous cell carcinoma and HPV E6/E7 encoded proteins.
