1.Time-dependent diffusion MRI parameters for differentiating invasive breast cancer with ductal carcinoma in situ and simple invasive breast cancer

Objective To explore the value of time-dependent diffusion MRI(td-dMRI)parameters for differentiating invasive breast cancer(IBC)with ductal carcinoma in situ(DCIS)(IBC-DCIS)from simple IBC.Methods A total of 19 patients with IBC-DCIS(IBC-DCIS group)and 53 patients with simple IBC(IBC group)confirmed by surgery and postoperation pathology were retrospectively enrolled.Breast td-dMRI acquired with oscillating gradient spin-echo(OGSE)and pulsed gradient spin-echo(PGSE)sequences before operation were interpreted,and apparent diffusion coefficient(ADC)and microstructure parameters,including OGSE-ADC value,PGSE-ADC value,cellularity,cell diameter,intracellular volume fraction and extracellular diffusion coefficient were obtained and compared between groups.Receiver operating characteristic(ROC)curves of parameters being significantly different between groups were plotted,and the area under the curve(AUC)was calculated to evaluate the efficacy of these parameters for differentiating IBC-DCIS from IBC.Results Significant differences of OGSE-ADC value,PGSE-ADC value,cellularity,cell diameter,intracellular volume fraction and extracellular diffusion coefficient were found between groups(all P<0.05).The AUC of the above parameters for differentiating IBC-DCIS from IBC was 0.81,0.79,0.78,0.68,0.77 and 0.81,respectively.Conclusion td-dMRI parameters could be used to noninvasively and effectively differentiate IBC-DCIS from simple IBC.
2.Exploring mechanism of Lycium barbarum polysaccharides in preventing inflam-matory bowel disease in chicks based on network pharmacology

This study aims to explore protective effects of Lycium barbarum polysaccharides(LBP)on intestinal damage caused by lipopolysaccharide(LPS)-induced inflammatory bowel disease(IBD)in chicks.Network pharmacology was initially employed to determine the target proteins of wolfberry in the prevention and treatment of IBD.Following this,protein-protein interaction analy-sis,GO and KEGG pathway enrichment analysis,and molecular docking studies were conducted.Subsequently,an animal study was conducted:a total of 100 one-day-old male Hy-line brown lay-ing hens were randomly divided into five groups:a blank control group(CON),an LPS treatment group(LPS),a low-dose LBP group(LPS+LBP 0.25 g/L,L-LBP),a medium-dose LBP group(LPS+LBP 0.5 g/L,M-LBP),and a high-dose LBP group(LPS+LBP 1 g/L,H-LBP).Upon reac-hing 21 days old,duodenal,jejunal,ileal,and cecal tissues were collected to determine SOD and GSH-Px levels.Furthermore,the mRNA expression levels of TNF-α,AKT1,IL-6,IL-1β and TP53 in the intestinal tissues were measured using quantitative real-time PCR.The results demonstrated that network pharmacology identified 45 active ingredients in wolfberry that target 116 key protein sites,including TNF,AKT1 and IL6.The primary objectives focus on signaling pathways including AGE-RAGE,IL-17,TNF,HIF-1,and NF-κB.Molecular docking showed excellent ligand-receptor docking scores,with stable binding facilitated by hydrogen bonds and hydrophobic interactions.Compared to the LPS group,the 0.5 g/L LBP exhibited notably higher levels of SOD and T-AOC.In comparison with the LPS group,the medium and high-dose LBP experimental groups showed notably decreased the mRNA expressions of TNF-α,AKT1,IL-6,and IL-1β,while TP53 mRNA expression was significantly upregulated(P<0.01).In summary,wolfberry exerts preventive and therapeutic effects on IBD through a multi-component,multi-target,and multi-pathway mecha-nism.
3.Value of Serum lncRNA KCNQ1OT1 and FGF2 Levels in the Assessment of Disease Control in Children with Bronchial Asthma

Objective To explore the value of serum long non-coding RNA KCNQl opioid strand transcript 1(lncRNA KCNQ1OT1)and fibroblast growth factor 2(FGF2)in the evaluation of the condition and control level of pediatric bronchial asthma.Methods 96 children with bronchial asthma(Asthma group)and 76 healthy children(Control group)admitted to Yulin Hospital of Traditional Chinese Medicine from March 2021 to March 2023 were selected.According to the severity of bronchial asthma,the children were divided into a mild group(n=54)and a severe group(n=42).According to the disease control status after treatment,the children were divided into an asthma Control group(n=61)and an uncontrolled Asthma group(n=35).The serum levels of lncRNA KCNQ1OT1 and FGF2 were detected in each group.Multivariate Logistic regression analysis was performed to identify the factors affecting bronchial asthma disease control.The performance of lncRNA KCNQ1OT1 and FGF2 in predicting the control of bronchial asthma disease was evaluated using the receiver operating characteristic curve.Results The serum levels of lncRNA KCNQ1OT1(2.35±0.71)and FGF2(27.35±6.29 pg/ml)in the Asthma group were higher than those in the Control group(1.51±0.33,15.32±4.08 pg/ml),and the differences were statistically significant(t=9.527,14.437,all P<0.001).The serum levels of lncRNA KCNQ1OT1(2.59±0.35)and FGF2(28.65±1.03 pg/ml)in the severe group were higher than those in the mild group(2.16±0.41,26.34±1.24 pg/ml),and the differences were statistically significant(t=5.429,9.737,all P<0.001).The serum levels of lncRNA KCNQ1OT1(2.63±0.27)and FGF2(29.02±1.41 pg/ml)in the uncontrolled asthma group were higher than those in the controlled asthma group(2.19±0.35,26.39±1.52 pg/ml),and the differences were statistically significant(t=5.417,8.374,all P<0.001).High lncRNA KCNQ1OT1 and FGF2 levels were risk factors for uncontrolled disease in children with bronchial asthma(all P<0.05).The AUC of lncRNA KCNQ1OT1 and FGF2 combined to predict uncontrolled disease in children with bronchial asthma,which was higher than of lncRNA KCNQ1OT1 and FGF2 alone,and the differences were statistically significant(Z=3.250,2.096,all P<0.05).Conclusion Serum lncRNA KCNQ1OT1 and FGF2 are both elevated in children with bronchial asthma,which is related to the worsening of bronchial asthma and uncontrolled disease.Combining lncRNA KCNQ1OT1 and FGF2 can predict the level of bronchial asthma disease control.
4.Application of solution-focused approach combined with empathic nursing in post-PCI patients with acute myocardial infarction

Objective:To explore the application effects of the solution-focused approach combined with empathic nursing in patients with acute myocardial infarction (AMI) after percutaneous coronary intervention (PCI) .Methods:A total of 96 AMI patients who underwent PCI in the Department of Cardiology at Cangzhou Central Hospital from March 2020 to March 2023 were selected using convenience sampling. They were randomly assigned to an experimental group ( n=48) and a control group ( n=48) using a random number table. The control group received routine nursing care, while the experimental group received a solution-focused approach combined with empathic nursing. Medication adherence, coping strategies, and patient satisfaction were compared between the two groups. Results:After the intervention, medication adherence and satisfaction scores in the experimental group were significantly higher than those in the control group, and the differences werestatistically significant ( P<0.05) . Additionally, the experimental group scored higher in confrontation coping, and lower in avoidance and resignation coping than the control group, and the differences were statistically significant ( P<0.01) . Conclusions:The combination of a solution-focused approach and empathic nursing can effectively improve medication adherence, coping strategies, and patient satisfaction in AMI patients after PCI.

Schizophrenia (SZ) stands as a severe psychiatric disorder. This study applied diffusion tensor imaging (DTI) data in conjunction with graph neural networks to distinguish SZ patients from normal controls (NCs) and showcases the superior performance of a graph neural network integrating combined fractional anisotropy and fiber number brain network features, achieving an accuracy of 73.79% in distinguishing SZ patients from NCs. Beyond mere discrimination, our study delved deeper into the advantages of utilizing white matter brain network features for identifying SZ patients through interpretable model analysis and gene expression analysis. These analyses uncovered intricate interrelationships between brain imaging markers and genetic biomarkers, providing novel insights into the neuropathological basis of SZ. In summary, our findings underscore the potential of graph neural networks applied to multimodal DTI data for enhancing SZ detection through an integrated analysis of neuroimaging and genetic features.

6.Impact of FASN-enriched EVs on endothelial cell function in obstructive sleep apnea hypopnea syndrome.

Endothelial dysfunction is a key factor linking obstructive sleep apnea hypopnea syndrome (OSAHS) with cardiovascular diseases. In this study, we used advanced proteomics and metabolomics approaches to investigate the impact of extracellular vesicles (EVs) derived from the serum of OSAHS patients on endothelial function. Our multi-omics analysis identified dysregulated pathways related to fatty acid metabolism, apoptosis regulation, and inflammatory responses, highlighting fatty acid synthase (FASN) as a crucial player in OSAHS-induced endothelial dysfunction. Both in vitro and in vivo experiments demonstrated that FASN-enriched EVs impair endothelial cell viability and disrupt metabolic homeostasis, offering new insights for the development of targeted therapies for cardiovascular complications associated with OSAHS.

8.Establishment of an indirect ELISA method for detection of ECoV antibody in donkey and application

In order to establish a method for the detection of serum antibodies to donkey-derived e-quine coronavirus(ECoV),recombinant ECoV N protein was expressed in E.coli system,purified by nickel column affinity chromatography and identified by Western blot.After optimizing the re-action conditions,the indirect ELISA(iELISA)detection method was established using the puri-fied recombinant protein as coating antigen and used to detect 143 clinical serum samples.The re-sults showed that the recombinant N protein,which has good reaction activity with serum antibod-y,was successfully expressed.The optimum conditions of the established iELISA method were as follows:the amount of antigen coated was 0.2 μg/well and overnight at 4 ℃,10%skimmed milk powder solution was sealed at 37℃ for 1.5 h,the dilution concentration of serum was 1∶200,and the enzyme-labeled secondary antibody diluted at 1∶10 000.The sensitivity test results showed that the positive serum could be diluted to 1∶6 400.The specificity test results showed that all an-tibodies to several donkey pathogens were negative.The repetitive test results showed that the in-tra-and inter-batch coefficients of variation were 2.90%-6.12%and 2.29%-7.88%respectively.The positive rate of clinical donkey serum was 57.3%.The iELISA established in this study pro-vides a technical support for epidemiological investigation and antibody surveillance.

10.Correlations of neutrophil-lymphocyte ratio and platelet-lymphocyte ratio with arteriovenous fistula stenosis in hemodialysis patients

Objective:To study the correlations of neutrophil-lymphocyte ratio(NLR)and platelet-lymphocyte ratio(PLR)with arte-riovenous fistula(AVF)stenosis in hemodialysis(HD)patients.Methods:Data were collected from 625 patients who underwent arterio-venous fistula hemodialysis at the Department of Nephrology,Affiliated Hospital of North Sichuan Medical College between January 2021 and June 2022.Of these,395 eligible patients with complete information were selected as subjects of study.The 245 patients with AVF stenosis were designated as group 1 and the 150 patients with-out AVF stenosis were designated as group 2.The routine biochemi-cal parameters and complete blood count were recorded for all pa-tients.Results:①Compared with patients in group 2,those in group 1 showed significantly higher NLR(5.07(4.00,6.66)vs.3.46(2.63,4.15),P<0.001),PLR(169.52(127.56,227.11)vs.125.66(89.31,165.31),P<0.001),and C-reactive protein(Hs-CRP)(1.90(0.80,2.99)vs.0.82(0.42,1.27),P<0.001).②Multivariate logistic regression analysis,which was corrected for age,sex,body mass index,AVF anastomosis,puncture method,and diabetes,showed that NLR(OR=2.195,95%CI=1.674~2.878,P<0.001),PLR(OR=1.008,95%CI=1.002~1.012,P=0.007),and Hs-CRP(OR=2.170,95%CI=1.607~2.751,P<0.001)were independent risk factors for AVF ste-nosis in HD patients.③Receiver operating characteristic curve analysis showed that the area under the NLR curve(0.799,95%CI=0.756~0.838,P<0.001),PLR(0.694,95%CI=0.646~0.740,P<0.001),and Hs-CRP(0.717,95%CI=0.670~0.761,P<0.001)could be used to predict AVF stenosis.Their optimal critical values for prediction were 4.08,122.49,and 1.62,respectively.Their combina-tion showed improved prediction effect(AUC 0.870,95%CI=0.833~0.901,P<0.001),high sensitivity(79.18%),and high specificity(81.33%).Conclusion:NLR,PLR,and Hs-CRP were independent risk factors and predictors of AVF stenosis,and their combination has higher predictive value.
