1.Whole genome sequencing and analysis of multidrug resistant ST314 Salmonella Kentucky from a broiler slaughterhouse

This study investigated the potential pathogenicity and genetic characteristics of ST314 Salmonella Kentucky(S.Ken-tucky)isolates from a broiler slaughterhouse.Antimicrobial susceptibility testing and whole-genome sequencing(WGS)were used to determine antimicrobial resistance,virulence factors,and the presence of antimicrobial resistance genes(ARGs)and mobile genetic elements(MGEs)among the isolates.The three multidrug resistant(MDR)isolates exhibited high resistance to multiple antimicrobial agents.The F4-2S strain exhibited resistance to 14 drugs across seven categories,whereas the F4T strain showed resistance to 13 drugs in the same number of categories.In contrast,the Y23 strain was resistant to nine drugs in six categories.Notably,F4-2S dem-onstrated high homology with F4T:both possessed 13 ARGs distributed across nine categories,in addition to a wide range of virulence factors,including secretion systems and effector proteins.The presence of IncR and IncX1 plasmids significantly enhanced both the antimicrobial resistance and pathogenicity of the isolates.The genome map of Y23 revealed a chromosome alongside two plasmids.The chromosome containedonly one resistance gene but several virulence factors,including the type III secretion system(T3SS),which is crucial for bacterial invasion.The plasmid pY23-1 contained eight types of 19 ARGs.Comparative analysis indicated that pY23-1 ex-hibited high homology with pZ1323SSL0055 and pSAL-045,all of which contained multiple ARGs,thus suggesting critical roles of these genes in the evolution of bacterial resistance.In conclusion,ST314 S.Kentucky demonstrated a complex mechanism of resis-tance coupled with significant pathogenic potential.The ARGs and MGEs in the plasmid contributed to the emergence and dissemina-tion of antimicrobial resistance.The multiple virulence factors present in the chromosome may be key factors driving the increasing virulence of ST314 S.Kentucky.

This consensus was developed by the Orthodontic Society of the Chinese Stomatological Association to provide a systematic, scientific, and practical guideline for informed consent in orthodontic care. Orthodontic treatment is typically lengthy, highly individualized, and involves multiple factors such as growth and development, occlusal function, and facial esthetics. Rapid technological advances and diverse risk profiles make the traditional reliance on orthodontist experience or institutional templates insufficient to ensure patients′ full understanding and autonomous decision-making. To address this, the expert panel conducted extensive reviews of domestic and international guidelines, analyzed representative dispute cases, and performed multicenter patient-clinician surveys. Using a multi-round Delphi method, the group established a standardized informed consent framework covering the initial consultation, treatment, and retention phases. The consensus emphasizes that informed consent is not only a fundamental legal and ethical requirement but also a key step in building trust, improving patient compliance, and enhancing treatment satisfaction. Orthodontists should clearly and comprehensively explain treatment plans, potential risks, uncertainties, and associated costs, while respecting the autonomy of patients or guardians, and maintain continuous communication and dynamic evaluation throughout the treatment process. The release of this consensus provides unified and authoritative guidance for clinical orthodontics, helping to standardize informed consent, enhance its transparency, safeguard patient rights, reduce medical risks, and promote high-quality, sustainable development of orthodontic practice.
3.Resistance exercise induces release of irisin from skeletal muscle and im-proves function of bone marrow endothelial progenitor cells in type 2 dia-betic rats

AIM:To investigate the effect of resistance exercise on the function of endothelial progenitor cells(EPCs)in type 2 diabetic rats and explore the mechanism of myokine irisin in this process.METHODS:Forty 6-week-old healthy male SD rats were selected,ten of which were used as control group,and the rest were fed with high-fat and high-sugar diet combined with streptozotocin injection for diabetic modeling.The rats were randomly divided into model group and model exercise group,with 15 rats in each group.The animals in the control and model groups were kept quiet-ly,and those of model exercise group underwent tail-loaded ladder climbing training for 12 weeks(3 times/week).72 hours after the last training,plasma,gastrocnemius and femur were separated.The irisin content of each tissue and mRNA as well as protein expression of fibronectin type Ⅲ domain-containing protein 5(FNDC5)in the gastrocnemius and femur were measured.Bone marrow endothelial progenitor cells were isolated and cultivated,and then treated with exoge-nous irisin for 48 hours.The cell viability and adhesion function of endothelial progenitor cells were detected by CCK-8 and adhesion assay,respectively.The endothelial repair ability was detected by carotid artery injury model in nude mice.The protein expression of phosphatidylinositol 3-kinase(PI3-K)/Akt/endothelial nitric oxide synthase(eNOS)signaling pathway was detected by Western blot.RESULTS:(1)In vivo experiment:compared with the control group,the model group showed a decrease in plasma,gastrocnemius muscle,and femoral irisin content,as well as a decrease in gastrocne-mius muscle FNDC5 mRNA and protein expression levels(P<0.05).Compared with the model group,the levels of irisin in plasma,gastrocnemius muscle and femur,as well as the expression of FNDC5 mRNA and protein in gastrocnemius mus-cle,increased in the model exercise group(P<0.05).There was no significant change in the expression of FNDC5 mRNA and protein in the femurs of each group(P<0.05).(2)Cell experiment:① endothelial progenitor cells without irisin treatment:compared with the control group,the cell viability,adhesion function,and endothelial injury repair ability of endothelial progenitor cells reduced(P<0.05),and the expression of PI3-K,Akt,eNOS signaling pathway down-regulated in the model group(P<0.05).Compared with the model group,the model exercise group showed improvement in endothe-lial progenitor cell function(P<0.05)and up-regulation of protein expression in the PI3-K/Akt/eNOS signaling pathway(P<0.05).② Endothelial progenitor cells treated with irisin:supplementation with irisin improved the function of endotheli-al progenitor cells and increased the expression of various proteins in the PI3-K/Akt/eNOS signaling pathway in the model group and model exercise group compared with those without irisin treatment(P<0.05).CONCLUSION:Regular resis-tance exercise can improve the function of endothelial progenitor cells in type 2 diabetic rats,and its mechanism may be re-lated to the regulation of PI3-K/Akt/eNOS signaling pathway by myokine irisin released from skeletal muscle.

Intraoperative neural monitoring (IONM) combines electrophysiology with anatomy to monitor nerve function during thyroid surgery,and has become an important auxiliary technology for neuroprotection. After more than ten years of development,the technology has been widely applied and popularized in China,promoting the development of a number of new technologies in the industry. Combined with the questionnaire survey data of the Chinese Neural Monitoring Study Group,this paper aims to summarize the clinical effect of IONM technology, the application status and existing problems at home and abroad, and propose the possible future development direction.

7.Preliminary study of three disinfectants on the surface of avian embryo eggs without specific pathogen

Objective:To screen out the disinfection procedure and disinfectant suitable for the actual production of specific pathogen free(SPF)chicken embryo eggs,so as to ensure the disinfection effect of specific pathogen free(SPF)chicken embryo eggs in vaccine production.Methods:This study compares the microbial counting methods of soaking,swabbing with cotton swabs and pouring after thin-film filtration for SPF chicken embryo eggs in a GMP production workshop,and selects the most suitable method for SPF chicken embryo egg microbial counting.Experi-mental groups A,B,and C use self-prepared concentrations of 1∶50 sporicidal agent dilution solution,1∶128 alka-line phenol salt dilution solution,and ready-to-use compound quaternary ammonium disinfectant(sterile),respec-tively,and follow the actual disinfection procedures in the workshop to disinfect and sample the surfaces of SPF chicken embryo eggs entering different cleanliness grades,while using sterile water instead of disinfectant as the control group.The average bactericidal rate is calculated by recording the number of colonies and monitoring the viability of chicken embryo cells using microbial culture,and the disinfection effect of the three disinfectants on SPF chicken embryo eggs is evaluated.Results:The comparison of the results from the three methods shows that the method of sampling SPF chicken embryo eggs by immersion and then counting the microbial colonies through membrane filtration is superior to the other two methods.The final cleaning rate of the control group,which used sterilized injection water to clean the SPF chicken embryo eggs,was 91.67%to 96.97%,while the final steriliza-tion rate of the experimental group,which used the above three disinfectants to disinfect the SPF chicken embryo eggs,was 100.00%.By comparing the cell counts of the experimental group and the control group,it was found that the live cell density of the control group was(6.03-6.25)× 105 cells·mL-1,and that of the experimental groups A-C was(6.08-6.17)× 105 cells·mL-1,(5.99-6.25)× 105 cells·mL-1,and(5.87-6.21)× 105 cells·mL-1 respectively;the cell viability of the control group was 90.33%to 91.35%,and that of the experi-mental groups A-C was 88.25%to 92.12%,89.45%to 93.59%,and 88.02%to 92.89%respectively.Through statistical analysis,it was found that the P values of all experimental groups compared with the control group were greater than 0.05,indicating no statistically significant difference.Conclusion:By comparing the dis-infection effects,cell density and cell viability of the three disinfectants and comprehensively considering factors such as cost and risk of the three disinfectants,1∶50 sporicide dilution,1∶128 alkaline phenolate dilution and ready-to-use compound quaternary ammonium salt disinfectant(sterile)can all be used for the daily surface disin-fection of SPF chicken embryo eggs in the production workshop.The selection of an appropriate disinfectant should be based on specific application scenarios and requirements.
