1.Effect of silencing MPZL1 on tumor cell stemness and drug resistance of A549 / Tax cells via β-catenin signaling

Objective :
To explore the effect of MPZL1 knockdown in A549 Taxol resistant (A549 / Tax) cells and whether it affect drug resistance and tumor cell stemness by regulating β-catenin.
Methods :
A549 and A549 / Tax cells were treated with different concentrations of doxorubicin and paclitaxel to observe the differences in drug resist- ance between the two cells.Quantitative real-time PCR (qRT-PCR) and Western blot were used to detect the MP- ZL1 expression level in A549 and A549 / Tax cells. After knockdown or overexpression of MPZL1 in A549 / Tax cells,cells were divided into control group,small hairpin RNA negative control ( sh-NC) group,MPZL1 knock- down(sh-MPZL1) group,overexpression negative control ( OE-NC) group,MPZL1 overexpression ( OE-MPZL1) group.Cell counting kit-8 ( CCK-8 ) and clone formation assay were utilized to investigate cell proliferation and clone formation ablity.Western blot assay was used to detect the protein expression after the cells treated with Wnt / β-catenin signaling inhibitor XAV939 and activator CHIR-99201 .
Results :
The half inhibitory concentration ( IC50 ) of doxorubicin and paclitaxel in A549 / Tax cells significantly increased compared to A549 cells(P<0. 01) . MPZL1 presented a higher expression trend in A549 / Tax cells.The IC50 values of A549 / Tax for doxorubicin and paclitaxel were 2. 731 mg / ml and 4. 939 μg / ml after MPZL1 knockdown,compared to 4. 541 mg / ml and 13. 55 μg / ml in the NC group (P<0. 01) .The results of CCK-8 and clone formation assay showed that the knockdown of MPZL1 reduced the viability of cells proliferation and clonal formation ability (P<0. 05) .Western blot results in- dicated that the expression levels of MPZL1 protein,tumor cell stemness associated proteins ( CD44,CD133) ,β - catenin and multidrug resistance protein 1 (MDR1) ,lung resistance-related protein ( LRP) were significantly re- duced in the sh-MPZL1 group. Furthermore ,XAV939 could inhibit the expression levels of MPZL1 ,CD44, CD133,MDR1,LRP and β-catenin(P<0. 01) .The inhibitory effect of knockdown MPZL1 on the aforementioned proteins was significantly reversed by CHIR-99201 treatment.
Conclusion
MPZL1 is highly expressed in A549 / Tax cells.Knockdown MPZL1 suppresses the tumor cell stemness and proliferation,thereby reversing the drug re- sistance of doxorubicin and paclitaxel in A549 / Tax cells.
2.Bilateral adrenal hemorrhage caused by anticoagulant therapy after joint replacement surgery: One case report and literature review

Bilateral adrenal hemorrhage is a rare cause of primary adrenal insufficiency, and bilateral adrenal hemorrhage due to anticoagulant use is even rarer. We describe the case of a 62-year-old woman receiving post total knee arthroplasty anticoagulant therapy who presented fever, vomitting, stomachache, and severe fatigue on the 8th day. It was until 4 months later that the patient was finally diagnosed with adrenal insufficiency resuting from bilateral adrenal hemorrhage, her symptoms were relieved by glucocorticoid replacement therapy. In order to promote the awareness, diagnosis, and mangement of post-surgery anticoagulants induced bilateral adrenal hemorrhage, the clinical characteristics of the reported cases were summarized and analyzed.
4.Investigation on psychological status of patients with acute pancreatitis and analysis of influencing factors

Objective To investigate the pshcyological status and influencing factors of patients with acute pancreatitis. Methods Acute pancreatitis patients admitted in Changhai Affiliated Hospital of Navy Medical University from December 2018 to February 2019 were enrolled, and 120 patients were selected using random number table method. Self-made general questionnaire, self-rating depressing scale, self-rating anxiety scale and coping style questionnaire were used for investigation and patients′ psychological status was evaluated. Results In this survey, 109 questionnaires were collected and eligible for final analysis. The incidence of depression in patients with acute pancreatitis was 44. 0%, and the incidence of anxiety was 20. 2%. Male patients had significantly lower anxiety and depression than women. The levels of anxiety and depression in the 31-50 age group were significantly lower than those in other age groups. The level of anxiety in patients with pancreatitis with children was significantly lower than in patients without children. The coping style of "solving problem" was significantly negatively correlated with depression and anxiety level. The"self-blame" coping style was significantly positively correlated with depression and anxiety level. The"rationalized"coping style was positively correlated with the level of depression. Conclusions The mental health status of patients with acute pancreatitis was not optimistic. Different coping styles were important factors affecting the level of anxiety and depression.
6.Learning from POSTAL study: can levothyroxine improve pregnancy outcomes in women with thyroid autoimmunity?

Whether levothyroxine ( LT4 ) treatment improves outcomes following in vitro fertilization and embryo transfer ( IVF-ET) in euthyroid women who have tested positive for thyroid autoantibodies remains unclear. In Pregnancy Outcome Study in enthyroid women with Thyroid Autoimmunity after Levothyroxine ( POSTAL) trial, which was a randomized controlled study involving 600 euthyroid women undergoing IVF-ET who were tested positive for thyroperoxidase antibodies, the miscarriage rate, clinical pregnancy rate and live-birth rate were not significantly different between the LT4 intervention group and control group. Therefore, LT4 treatment did not appear to improve pregnancy outcomes among women with thyroid autoantibodies undergoing IVF-ET.
7.Clinical analysis on arthroscopic debridement for the treatment of early infection after total knee replacement.

OBJECTIVETo compare the clinical effects of arthroscopic debridement versus open debridement on controlling and treatment of infection after total knee replacement.
METHODSFrom October 2009 to September 2016 in three hospitals, 11 patients with 11 joints which were infected after total knee replacement were randomly divided into two groups:5 cases in arthroscopy group and 6 cases in routine group. Patients in arthroscopy group were treated with arthroscopic debridement to remove the necrotic tissues, then closed-type irrigation with sensitive antibiotics by using two sebific ducts were performed continuously for 2 or 3 weeks until the flushing fluid became clear for 3 or 5 days;other 6 patients in routine group were treated with open surgical debridement and the following procedures in keeping with those in the arthroscopy group. Operation time, blood loss and incision length were recorded during the operation, and pain scores were recorded on the 1st, 3rd and 7th day after the operation. The curative effects were evaluated according to the Hospital for Special Surgery score system.
RESULTSThe local and general symptoms of the 11 patients disappeared, and the test outcomes of biochemistry, blood and synovial fluid were normal. All patients were followed up, and the duration ranged from 6 to 18 months. Infection recurrences were observed in 1 case of arthroscopy group and 2 cases of routine group 3 months later after operation, and all these patients who underwent the second time operation with arthroscopic debridement were cured. According to the Hospital for Special Surgery score system, 3 cases obtained excellent result, 2 good, no poor and bad cases in arthroscopy group;3 cases obtained excellent result, 1 good, 1 poor and 1 bad in routine group.
CONCLUSIONSIf the sensitive antibiotics can be found for the infected joints without obvious destruction of bone and no prosthesis loosening, it has a better therapeutic effect by using arthroscopic debridement combined with continuous drainage and irrigation. The method has a better curative effect with smaller trauma.
8.Retrospective analysis of craniofacial features in patients with cleidocranial dysplasia

Objective:To investigate the craniofacial features in patients with cleidocranial dysplasia (CCD).Methods:The facial features,cervical vertebral bone age and skeletal abnormalities of 8 patients with CCD were studied by analyzing facial photos,cephalo-metric and panoramic radiographs.Results:4 patients were in the early growth stage and the other 4 in the late period of development. The bossing forehead and inclined eye fissure were observed in all patients,but underdevelopment of midfaces were not obviously pres-ented in younger patients.Morphological abnormalities of craniofacial bones,such as ascending ramus,coronoid process,nasal bones and disappearence of gonial angle were observed in all patients.Conclusion:Some craniofacial malformations in patients with CCD may be presented earlier than underdeveloped midface,which can be helpful for early diagnosis of CCD.
9.Advances in radionuclide molecular imaging of pancreatic β-cells

In both type 1 and type 2 diabetes mellitus,β-cell mass (BCM) is lost.Various treatments are developed to restore or reconstruct BCM.The development of non-invasive methods to quantify BCM in vivo offers the potential for early detection of β-cell dysfunction prior to the clinical onset of diabetes.PET imaging with radioligands that directly target the pancreatic β-cells appears promising.The ability to determine the BCM has been investigated in several targets and their corresponding radiotracers,including radiolabeled receptor ligands,antibodies,metabolites and reporter genes.Therefore,we summarize the recent progress in radionuclide molecular imaging of pancreatic β-cells.
10.Effect of working memory training task on visual contrast sensitivity for college students

Objective To explore the influence on college students' contrast sensitivity how dual N-back working memory training task based on Gabor,and to provide new training method for improving the contrast sensitivity.Methods 42 college students were randomly divided into three groups:a control group (17 students) ; a working memory training group (13 students) ; and a Gabor orientation training group (12 students).The control group received no training,while the working memory training group and the Gabor orientation training group received N-back working memory training and Gabor training,respectively,over three weeks (5 days a week,approx.half an hour per day).The above-mentioned two training methods performed with the same visual stimulus,i.e.,the Gabor patch with a fixed spatial frequency and contrast.The contrast sensitivity of the 42 students was measured and recorded using OPTEC6500 before and after training.Results In the working memory training group,a salient gain in contrast sensitivity was observed after the training (before:(1.48 ± 0.26) log,after:(1.87 ± 0.13)log),with a statistically significant difference (t =-6.20,P < 0.05),while both the Gabor orientation training group (before:(1.75 ± 0.19) log,after:(1.68 ± 0.30) log) and the control group (before:(1.67 ±0.25) log,after:(1.61 ± 0.26) log)) showed no statistically significant difference (P > 0.05) in terms of contrast sensitivity.Conclusion N-back working memory training shows a positive effect on improving contrast sensitivity in Gabor identification training,and can be used as a means to improve contrast sensitivity.
