ObjectiveTo observe the effect of Huayu Mingmu prescription on retinal angiogenesis and phosphatidylinositol 3-kinase/protein kinase B/mammalian target of rapamycin （PI3K/Akt/mTOR）-hypoxia inducible factor-1α/vascular endothelial growth factor A （HIF-1α/VEGFA） signaling pathway in diabetic retinopathy （DR） rats. MethodsSixty-four SPF-grade male SD rats were used in the study. Eleven rats were randomly selected as the normal group， while the remaining 53 rats were fed a high-sugar， high-fat diet combined with low-dose streptozotocin （STZ） intraperitoneal injection to establish a type 2 diabetes mellitus （T2DM） rat model. DR model evaluation was performed after 12 weeks of diabetes. The rats were then divided into model， low-dose， medium-dose， and high-dose groups of Huayu Mingmu prescription （9.29， 18.57， 37.14 g·kg^-1）， and a calcium dobesilate group （0.16 g·kg^-1）， with 10 rats in each group. The rats were orally administered the corresponding doses of Huayu Mingmu prescription and calcium dobesilate. The normal and model groups received equal volumes of physiological saline via gavage for 8 consecutive weeks. Retinal vascular changes were observed through fundus photography， and pathological changes in retinal tissue were evaluated using hematoxylin-eosin （HE） staining. Retinal microvascular pathological changes were examined through retinal vascular network preparation and periodic acid-Schiff （PAS） staining. Immunofluorescence （IF） was used to detect the expression of VEGFA and angiopoietin-2 （Ang-2） in retinal tissue. Western blot was employed to detect the protein expression of PI3K， Akt， mTOR， HIF-1α， VEGFA， and VEGFR2 in retinal tissue. Real-time fluorescence quantitative polymerase chain reaction （Real-time PCR） was used to assess the mRNA expression of PI3K， Akt， mTOR， HIF-1α， VEGFA， and VEGFR2 in retinal tissue. ResultsCompared with the normal group， the model group exhibited significant pathological changes in retinal tissue， including the appearance of acellular capillaries， as well as significant endothelial cell （E） proliferation and pericyte （P） loss （P<0.01）. The E/P was significantly elevated （P<0.01）. Protein and mRNA expression levels of PI3K， Akt， mTOR， HIF-1α， VEGFA， and VEGFR2 in retinal tissue were significantly increased （P<0.01）， and the expression of Ang-2 protein was significantly elevated （P<0.01）. In contrast， retinal tissue in the treatment groups showed alleviated pathological changes， with reduced endothelial cell proliferation and pericyte loss （P<0.05， P<0.01）. Among the treatment groups， the high-dose Huayu Mingmu prescription and the calcium dobesilate group exhibited a decreased E/P （P<0.01）. Protein and mRNA expression levels of PI3K， Akt， mTOR， HIF-1α， VEGFA， and VEGFR2 in retinal tissue were significantly reduced （P<0.05， P<0.01）， and the expression of Ang-2 protein was significantly decreased （P<0.01）. ConclusionHuayu Mingmu prescription can intervene in retinal neovascularization in DR rats， delay the progression of DR， and its mechanism may be related to antagonizing the PI3K/Akt/mTOR-HIF-1α/VEGFA signaling pathway.

Objective: To explore effects and maintenance of school based sexual abuse prevention programs for minors, so as to provide scientific evidences for optimizing intervention design and policy making. Methods: Six Chinese and English databases were searched, including CNKI, Wanfang Database, Medline (via PubMed), Embase, Cochrane Library and Web of Science, with the time frame set from database inception to December 31, 2024. Studies on school based sexual abuse prevention programs for minors were selected, and data on knowledge, attitudes and skills related to sexual abuse prevention were extracted. Meta analysis was performed using Stata 17. Results: A total of 26 studies were included. The Meta analysis results showed that school based sexual abuse prevention programs improved participants knowledge ( SMD=1.24, 95%CI =0.96-1.52), attitudes ( SMD=0.62, 95%CI =0.19-1.04) and skills ( SMD=0.66, 95%CI =0.50-0.83) (all P <0.01). During the overall follow up, the maintenance rates for knowledge, attitudes, and skills were 0.97(95% CI =0.95-1.00), 0.99(95% CI =0.95-1.04) and 1.01(95% CI =0.99-1.04), respectively, with no statistically significant differences (all P >0.05). However, knowledge retention declined significantly when follow up exceeded three months ( R=0.91, 95%CI=0.83-0.99, P <0.01), while skills retention ( R=0.94, 95%CI=0.87-1.02, P = 0.23) remained higher than knowledge and attitudes ( R=0.98, 95%CI=0.96-1.00, P =0.13), demonstrating stronger long term effects. Conclusion School based sexual abuse prevention programs are effective in enhancing participants knowledge, attitudes and skills, but the intervention effects diminish over time, particularly in knowledge retention.

Objective To investigate the impact of dexmedetomidine on epidural fibrosis after spinal surgery in rats through the transforming growth factor-β1(TGF-β1)signaling pathway.Meth-ods A total of 40 rats were selected,with 10 rats assigned to control group.The remaining 30 rats underwent spinal surgery modeling and were randomly divided into model group,dexmedetomidine group,and dexmedetomidine+TGF-β1 agonist group,with 10 rats in each.Rats with unsuccessful modeling were excluded,resulting in 9 rats in each group for subsequent analysis.Pathological char-acteristics of epidural scar tissue,fibroblast count,PCNA-positive cell expression rate,levels of in-flammatory markers[interleukin(IL)-6,IL-1β,IL-8],stress response indicators[malondialdehyde(MDA),superoxide dismutase(SOD)],and expression levels of mRNAs and proteins related to the TGF-β1 signaling pathway were observed and compared among the groups.Results Compared with the control group,the other three groups showed increased fibroblast count,elevated PCNA-positive cell expression rate,higher levels of IL-6,IL-1β,IL-8,and MDA,lower SOD levels,and increased expression levels of TGF-β1 mRNA,Smad3 mRNA,and TGF-β1 and Smad3 proteins,with statistically significant differences(P＜0.05).Compared with the model group,both the dexmedetomidine group and the dexmedetomidine+TGF-β1 agonist group exhibited decreased fibroblast count,re-duced PCNA-positive cell expression rate,lower levels of IL-6,IL-1 β,IL-8 and MDA,higher SOD levels,and decreased expression levels of TGF-β1 mRNA,Smad3 mRNA,and TGF-β1 and Smad3 proteins,with statistically significant differences(P＜0.05).Compared with the dexmedetomidine group,the dexmedetomidine+TGF-β1 agonist group showed increased fibroblast count,elevated PCNA-positive cell expression rate,higher levels of IL-6,IL-1 β,IL-8 and MDA,lower SOD lev-els,and increased expression levels of TGF-β1 mRNA,Smad3 mRNA,and TGF-β1 and Smad3 proteins,with statistically significant differences(P＜0.05).Conclusion Dexmedetomidine can significantly alleviate the inflammatory response and stress response after spinal surgery in rats and reduce postoperative epidural fibrosis by inhibiting the TGF-β1 signaling pathway.

The goal of gastrointestinal endoscopy anesthesia management is to effectively calm and re-lieve pain while minimizing related adverse reactions and ensuring patient safety.Hypoxemia is the most com-mon adverse event during painless gastrointestinalendoscopy,and severe hypoxemia can cause cardiac and brain accidents.Therefore,how to prevent and reduce the occurrence of hypoxemia isa hot topic in clinical re-search.This article reviews the methods of preventing and reducing hypoxemia in general painless gastrointes-tinal endoscopy,and provides a reference for the selection of appropriate sedation and ventilation strategies for general painless gastrointestinal endoscopy anesthesia.

Developing and implementing biosafety standards for pathogenic microbiology laboratories is essential to achieving scientific, efficient, and standardized management and operation. This article analyzes the current standardization construction in biosafety in pathogenic microbiology laboratories domestically and internationally. It proposes a framework for the biosafety standard system of pathogenic microbiology laboratories, which mainly includes four parts: basic standards, management standards, technical standards, and industry applications. It provides a reference for the standardization work of pathogenic microbiology laboratories and helps to standardize the biosafety industry in China.

Objective:To investigate the independent and combined effects of maternal smoking during pregnancy and offspring genetic susceptibility on overall cancer mortality.Methods:Based on the United Kingdom Biobank ( n=419 228) data, the Cox proportional hazard regression model was used to estimate the effect of maternal smoking during pregnancy on offspring overall cancer (including 16 cancers in men and 18 in women) mortality and its combined effect and interaction with offspring genetic factors. Results:Maternal smoking during pregnancy was significantly associated with a 13% increased risk of overall cancer mortality in men [hazard ratio( HR)=1.13, 95% CI: 1.06-1.20] and 19% increased risk in women ( HR=1.19, 95% CI: 1.11-1.27). Participants with high genetic risk had the highest overall cancer mortality than those with low genetic risk (men: HR=1.42, 95% CI: 1.30-1.55; women: HR=1.38, 95% CI: 1.25-1.52). Compared with participants without maternal smoking during pregnancy and low genetic risk, those with maternal smoking during pregnancy and high genetic risk were associated with a 56% increased risk of overall cancer mortality in men ( HR=1.56, 95% CI: 1.37-1.77) and 59% in women ( HR=1.59, 95% CI: 1.39-1.83). Conclusion:Maternal smoking during pregnancy may increase offspring overall cancer mortality and more severe harm in individuals with high genetic risk.

Objective To study the diagnostic value of serum long chain non coding RNA(lncRNA)CB-SLR,α trypsin H4 heavy chain(ITIH4)in early gastric cancer in the elderly.Methods From 2020 to 2022,a total of 90 elderly patients with early gastric cancer diagnosed and treated in the hospital were selected as the early gastric cancer group,while 45 patients with chronic atrophic gastritis diagnosed and treated in the same period were selected as atrophic gastritis group and 45 healthy people undergoing physical examination in the same period were selected as the healthy control group.The serum levels of lncRNA CBSLR and ITIH4 in each group were detected by fluorescence quantitative PCR and enzyme linked immunosorbent assay.Pearson correlation analysis was performed to determine the correlation between serum lncRNA CBSLR and ITIH4 levels with tumor markers carcinoembryonic antigen(CEA)and carbohydrate antigen(CA)19-9.The levels of serum lncRNA CBSLR and ITIH4 among patients with different clinicopathological characteristics were com-pared.The diagnostic value of serum lncRNA CBSLR and ITIH4 in elderly patients with early gastric cancer was analyzed by receiver operating characteristic curve.Results The serum levels of lncRNA CBSLR,ITIH4,CEA and CA19-9 in the early gastric cancer group were significantly higher than those in the atrophic gastritis group and health control group(all P＜0.05).There was significant positive correlation between serum ln-cRNA CBSLR,ITIH4 and CEA,CA19-9 levels in patients with early gastric cancer(r=0.601-0.751,all P＜0.05).In patients with early gastric cancer,serum IncRNA CBSLR and ITIH4 were related to tumor diame-ter,depth of invasion,differentiation type,and lymph node metastasis(all P＜0.05),but not related to age,gender,tumor location,and gross type(all P＞0.05).The area under the curve of combined detection of ser-um lncRNA CBSLR,ITIH4,CEA and CA19-9(0.912,95％CI:0.858-0.949)for the diagnosis of early gas-tric cancer was higher than that of serum lncRNA CBSLR(0.833,95％CI:0.784-0.879),ITIH4(0.806,95％CI:0.760-0.847),CEA(0.791,95％CI:0.742-0.830),and CA19-9(0.766,95％CI:0.710-0.815)alone(Z=4.482,5.130,5.231,6.117,all P＜0.05).Conclusion The elevated levels of serum lncRNA CB-SLR and ITIH4 in elderly patients with early gastric cancer are associated with adverse clinicopathological characteristics of gastric cancer.The combination of serum lncRNA CBSLR,ITIH4,CEA and CA19-9 has high diagnostic value for elderly patients with early gastric cancer.

Objective To investigate whether activation of mitochondrial acetal dehydrogenase 2(ALDH2)alleviates hypoxic pulmonary hypertension by regulating the SIRT1/PGC-1α signaling pathway.Methods Thirty 8-week-old C57 BL/6 mice were randomized into control,hypoxia,and hypoxia+Alda-1(an ALDH2 activator)group(n=10),and the mice in the latter two groups,along with 10 ALDH2 knockout(ALDH2-/-)mice,were exposed to hypoxia(10%O2,90%N2)with or without daily intraperitoneal injection of Alda-1 for 4 weeks.The changes in right ventricular function and pressure(RVSP)of the mice were evaluated by echocardiography and right ventricular catheter test,and pulmonary artery pressure was estimated based on RVSP.Pulmonary vascular remodeling,right ventricular injury,myocardial α-SMA expression,distal pulmonary arteriole muscle normalization,right ventricular cross-sectional area,myocardial cell hypertrophy,and right cardiac hypertrophy index were assessed with HE staining,immunofluorescence staining and WGA staining,and the expressions of ALDH2,SIRT1,PGC-1α,P16INK4A and P21CIP1 were detected.In pulmonary artery smooth muscle cells with hypoxic exposure,the effect of Alda-1 and EX527 on cell senescence and protein expressions was evaluated using β-galactose staining and Western blotting.Results The wild-type mice with hypoxic exposure showed significantly increased RVSP,right ventricular free wall thickness and myocardial expressions of P16INK4A and P21CIP1,which were effectively lowered by treatment with Alda-1 but further increased in ALDH2-/-mice.In cultured pulmonary artery smooth muscle cells,hypoxic exposure significantly increased senescent cell percentage and cellular expressions of P16INK4A and P21CIP1,which were all lowered by treatment with Alda-1,but its effect was obviously attenuated by EX527 treatment.Conclusion ALDH2 alleviates hypoxia-induced senescence of pulmonary artery smooth muscle cells by upregulating the SIRT1/PGC-1α signaling pathway to alleviate pulmonary hypertension in mice.
