1.Effect of Neuritin on angiogenesis of chicken embryo alantoic membrane and humar umbilical vein endo-thelial cells

Objective To investigate the effects of neurotrophic factor Neuritin overexpression on the angiogenic effects of chicken embryonic allantoic membrane (CAM) and human umbilical vein endothelial cells (HUVECs),and to provide a new direction for the treatment of angiogenic diseases. Methods Thirty fresh yellow-skinned breeding eggs were selected to establish a CAM model,which were divided into three groups by randomized numerical table method:positive control group (bFGF),negative control group (NS) and experimental group (Neuritin),with 10 eggs in each group. The positive control group was loaded with 2500 U/mL of bFGF,the experimental group was loaded with 10 μg/mL of Neuritin protein,and the negative control group was loaded with NS. 10 μL loading volume was loaded into each group,and all CAMs were incubated at the same temperature,relative humidity,and time,and the vascular branching,number,and size of the CAMs in each group were recorded after 72 h of incubation. Fresh umbilical cords from healthy pregnant women were selected to produce primary HUVECs,which were divided into three groups:transfected with recombinant plasmid (HUVEC-neu group),transfected with empty vector (HUVEC-3.1 group),and untransfected (HUVEC group). Primary HUVECs in the HUVEC-neu group were transfected with the recombinant plasmid Neuritin,and those in the HUVEC-3.1 group were transfected with the empty vector. HUVEC-3.1 group was transfected with the empty vector plasmid,and HUVEC group was not given any special treatment,and all three groups received the same culture regimen. Western blot was used to detect the protein expression level of Neuritin in HUVEC-3.1 and HUVEC-neu groups. CCK-8 assay,cell scratch assay,Transwell assay,and tube formation assay were used to detect protein expression level of Neuritin in HUVEC-3.1 group and HUVEC-neu group,and HUVEC-neu groups for cell proliferation,migration and tube formation. Results (1) The number of CAM vessel branch points and microvessels in the experimental group was significantly increased compared with that in the negative control group (P<0.01),but there was no statistically significant difference in the number of large and medium-sized vessels between the two groups (P>0.05);(2) Neuritin was successfully overexpressed in HUVECs in the HUVEC-neu group. (3) Compared with the HUVEC-3.1 group,the proliferation vigor of cells in the HUVEC-neu group was decreased (P<0.05),but their migration and tube formation abilities were significantly enhanced (P<0.01). Conclusion Neuritin overexpression promotes angiogenesis and participates in the regulation of neovascularization by affecting cell prolif-eration,migration,and tube formation ability.

Objective To explore the influence of different anesthesia methods on the circulation state and T lymphocyte subsets of patients with abdominal operation.Methods A total of 120 patients with abdominal operation were divided into group A(general anesthesia group)and group B(epidural combined anesthesia group).The hemodynamic indexes,cardiac output indexes and peripheral blood T lymphocyte subsets at different time points were analyzed and compared between two groups.Results The hemodynamic indexes at 5 min and 30 min during the operation and 5 min afteRthe operation in the group B were significantly loweRthan those in the group A (P<0.05),and the peripheral blood T lymphocyte subsets of group B at first and third day afteRthe operation were significantly betteRthan group A (P<0.05).Conclusion The effect of epidural general anesthesia on the circulatory status and T lymphocyte subsets in patients with abdominal operation is less,which is more suitable foRpatients with abdominal surgery.

5.Comparison between the quantitative and semi-quantitative fluorescence RT-PCR assays for the detection of XDH/XO mRNA expression in different organ tissues of rhesus monkey

Objective To analyze the differences between the semi-quantitative RT-PCR and real time quantitative fluorescence RT-PCR assays for detecting XDH/XO mRNA expression in various organ tissues of rhesus monkey, and provide useful reference in methodology of experimental studies.Total RNA was extracted from the myocardium, kidney, testis, skin, and liver tissues, respectively, for detecting XDH/XO mRNA expression in rhesus monkey by semi-quantitative RT-PCR and real time quantitative fluorescence RT-PCR assays.The sensitivity and specificity of the two assays were compared with each other using the same primer sequences and reference genes.Results The expression of XDH/XO mRNA in different organ tissues were detected by both the two PCR assays.The sensitivity of quantitative fluorescence real-time RT-PCR for the XDH/XO mRNA expression in the liver tissue was 39 times higher than that by semi-quantitative RT-PCR.Conclusions Both the quantitative and semi-quantitative fluorescence RT-PCR assays can be used to detect the expression of XDH/XO mRNA in different organ tissues of rhesus monkey.The sensitivity of quantitative fluorescence real-time PCR assay is more sensitive than that of the semi-quantitative RT-PCR assay.
6.Effect of team-based rehabilitation education on daily life ability of elderly patients having undergone operations on hip fractures

Objective To evaluate the effect of team-based rehabilitation education on daily life ability of old patients having undergone operations on hip fractures.Methods Sixty elderly reconvalescents from operations on hip fractures were equally assigned into the intervention and control groups according to admission sequence:The intervention group received rehabilitation education by community nurses at months 1,3 and 6 and the control group were managed with convalesce at home.The two groups were compared in terms of daily life ability at the three time points. Result The daily life ability of the intervention group was better than that of the control group at all the three time points(P<0.05). Conclusion The team-based rehabilitation education is effective for the enhancement of daily life ability of old patients having undergone operations on hip fractures.
7.The effects of pulsed electromagnetic field on healing of tibia and fibula fractures in rats

Objective To study the effects of pulsed electromagnetic field (PEMF) in promoting tibia and fibula fracture healing in rats. Methods Thirty rats were divided randomly into two groups after establishing the animal model of artificial tibia and fibula fracture: a PEMF treated group and a control group. Radiographs were taken immediately postoperatively and once a week thereafter until being sacrificed after week 5. The blood was sampled to measure the activity of alkaline phosphatase (ALP) and the concentration of bone gamma-carboxyglutamic-acid-con-taining proteins (BGP), calcium and phosphate in serum once a week, respectively. Histological sections were taken at weeks 3 and 5 to observe the pathological change of bony callus. Results PEMF of 15 Hz and 1.0 mT could sig-nificantly increase the amount of bony callus, significantly increase the amount of bony callus, promote the disappea-ring of fracture lines and the appearance of endochondral ossification and mature bone trabecula. The amount of colla-gen in bony callus raised and the process of callus moulding accelerated in PEMF treated group. In addition, the ac-tivity of ALP(P<0.04) and the concentration of BGP(P<0.05) in serum increased. Conclusions The PEMF of 15 Hz and 1.0 mT can obviously promote fracture healing of tibia and fibula in rats.
8.Herpes Simplex Virus 1 Infection Alters the mRNA Translation Processing in L-02 Cells

HSV-1 infection-mediated regulation of mRNA translation in host cells is a systematic and complicated process. Investigation of the details of this mechanism will facilitate understanding of biological variations in the viral replication process and host cells. In this study, a comparative proteomics technology platform was applied by two-dimension electrophoresis of HSV-1 infected normal human L-02 cell and control cell lysates. The observed protein spots were analyzed qualitatively and quantitatively by the PDQuest software package. A number of the different observed protein spots closely associated with cellular protein synthesis were identified by matrix-assisted laser-desorption ionization-time of flight-mass spectrometry (MALDI-TOF-MS). The expression levels of the RPLP1 protein, which is required for mRNA translation, and KHSRP protein, which is involved in rapid decay of mRNA, were up-regulated, whereas the expression level of RNP H2, which is involved in positive regulation on the mRNA splicing process, was down-regulated. All of these results suggest that HSV-1 infection can influence cellular protein synthesis via modulation of cellular regulatory proteins involved in RNA splicing, translation and decay, resulting in optimisation of viral protein synthesis when cellular protein synthesis is shut off. Although there is need for further investigations regarding the detailed mechanisms of cellular protein control, our studies provide new insight into the targeting of varied virus signaling pathways involved in host cellular protein synthesis.
9.Evaluation of the impact in longterm application of finished product of homogenate meal on senior patients with chronic diseases

BACKGROUND: Homogenate meals, as a replacement of normal diet, can be used in either hospitals or families. The homogenate meal as nutritious supplements should contain multiple components when it is applied in senior patients with chronic diseases because of the longterm application and complex complications.OBJECTIVE: To investigate the impact of longterm tube feed of finished product of homogenate meal on the nutritious supplements of senior patients.DESIGN: A cross-sectional study based on diagnosis.SETTING, PARTICIPANTS and METHODS: Totally 24 senior patients with chronic diseases who had been used self-made finished products of homogenate meal for more than 1 year were selected for nutritious supplement from Guangzhou General Hospital of Guangzhou Military Area Command of Chinese PLA. The changes of nutritious index at before and 4, 8 and 12 months after supplement were observed.MAIN OUTCOME MEASURES: Anthropometric measurement index and laboratorial index of 24 senior patients with chronic diseases before and after intervention were compared.RESULTS: Serous albumin at before nutritious supplements, and 4, 8 and 12 months after supplements were[ (36.4 ± 4.03), (36.60 ± 4.42),(38.20 ± 3.77), and(40.40 ± 3. 33) ] g/L respectively. Phosphorus and peripheral lymphocytes were significantly heightened while blood sugar began to decrease since 4 months after intervention, which all had significant differences compared with that of before intervention( t =2. 196 -2. 356,P <0.05; t=2.958-3.431. P <0.01) .Anthropometric measurement index also improved compared with that of before intervention.CONCLUSION: Longterm application of finished product of homogenate meal can improve the nutritious status of senior patients with chronic diseases.
