1.Association between genetic polymorphisms in XRCC1 and XRCC3 genes and cervical cancer susceptibility

Objective To investigate the potential association between XRCC1 and XRCC3 gene polymor-phisms and susceptibility to cervical cancer within populations infected with high-risk human papillomavirus(HPV).Methods A cohort of 176 cervical cancer patients with high-risk HPV infection,diagnosed at the First People's Hospital of Kashgar Region between January 2021 and January 2023,were enrolled as the study group.Concurrently,177 women infected with high-risk HPV but without cervical cancer were enrolled as the control group.All participants were followed up,and genotyping along with HPV subtyping was conducted for subsequent analysis.Results The XRCC1 Arg399Gln genotype showed a significant association with cervical cancer risk among individuals infected with specific HPV subtypes(e.g.,HPV-16,-18,-52,and-58),but no significant correlation was observed with the number of concurrent HPV infections.Compared to non-carriers,individuals carrying the XRCC1 Arg399Gln genotype exhibited an increased risk of cervical cancer,particularly among those infected with HPV-16,-18,-52,or-58.Statistically significant differences were found between the control group and the cervical cancer group for the XRCC1 Arg280His(P<0.001)and XRCC3 Thr241Met(P<0.05)genotypes;however,no significant differences were observed for XRCC1 Arg194Trp and XRCC1 Arg399Gln polymorphisms(P>0.05).Further analysis revealed that the XRCC1 Arg280His polymorphism was associated with a 3.57-fold higher risk of cervical cancer(OR=3.57,95%CI:1.99~6.43),while XRCC1 Arg194Trp showed no significant association(P>0.05).Additionally,XRCC1 Arg399Gln was associated with a reduced risk(OR=0.57,95%CI:0.34~0.96),and XRCC3 Thr241Met was linked to a decreased risk of cervical cancer(OR=0.22,95%CI:0.06~0.84).Conclusions In the HPV-infected population of Kashgar,the XRCC1 Arg280His polymorphism is significantly associated with an increased risk of cervical cancer(OR=3.57),whereas the XRCC3 Thr241Met(GG)genotype exhibits a protective effect(OR=0.16).The XRCC1 Arg399Gln variant demonstrates a protective association specifically among TT homozygotes(OR=0.57).In contrast,the XRCC1 Arg194Trp polymorphism does not show a statistically significant association with cervical cancer risk.
2.Surveillance results of iodine content in drinking water in Shaanxi Province

Objective To investigate iodine content in drinking water and clarify the distribution characteristics of iodine in water in Shaanxi Province.Methods A cross-sectional survey method was used to collect water samples from administrative villages in the province.Two tap water samples were taken from the centralized water supply villages,and 10%samples were taken from the decentralized water supply villages.Water iodine was detected by arsenic cerium catalytic spectrophotometry.Water iodine areas were divided according to national standards:<40 μg/L was iodine deficiency area,40-100 μg/L was iodine adequate area,>100 μg/L was high iodine area.According to the ecological regionification scheme of the Chinese Academy of Sciences,Shaanxi Province was divided into three types:the central and eastern Inner Mongolia Platea-Loess Plateau ecological region,the Fenwei Basin ecological region,and the Qinba Mountains ecological region.SPSS 25.0 was used for statistical analysis.Results The median of water iodine in Shaanxi Province was 6.66 μg/L.The survey of water iodine content was conducted in 22 848 administrative villages,1 309 townships,112 counties and 14 municipalities in the whole province.The median water iodine was less than 40 μg/L in 91.75%(20 963/22 848)of the administrative villages,between 40 and 100 μg/L in 7.40%(1 691/22 848)of the administrative villages,and more than 100 μg/L in 0.85%(194/22 848)of the administrative villages.The median of water iodine in the central and eastern Inner Mongolia Platea-Lose Plateau ecological zone,the Fenwei Basin ecological zone,and Qinba Mountains ecological zone was 12.35,8.88,and 2.00 μg/L,respectively,and the differences among different ecological zones were statistically significant(H=6 616.23,P<0.001).The median of water iodine of centralized and decentralized water supply was 6.72 and 6.21 μg/L,respectively,and differences between different water supply methods were statistically significant(Z=5.638,P<0.01).Conclusion The overall external environment of Shaanxi Province is iodine deficient,and most of the administrative villages are iodine deficient areas.There are a certain proportion of high iodine water source areas and suitable iodine areas.
3.Analysis of CT imaging characteristics of high altitude pulmonary edema

Objective To analyze the CT manifestations of high altitude pulmonary edema(HAPE)to provide radiological evidence for its early and accurate diagnosis.Methods Totally 200 HAPE patients clinically confirmed at some hospital from April 2021 to April 2024 were enrolled into a study group,and 56 individuals undergoing health examinations at the hospital's physical examination center between January and June 2023 were included into a control group.Examinations were carried out with a United Imaging uCT528 40-slice spiral CT scanner.The patients in the study group were observed in terms of HAPE staging,the extent of pulmonary involvement,CT manifestations of different stages including location,distribution,density and morphology of pulmonary lesion.The diameters of the main pulmonary arteries and ascending aortas of the subjects in the two groups were measured,and the ratios of the two diameters were calculated.SPSS 25.0 software was used for statistical analysis.Results In the study group,there were 26 cases(13.0%)at early stage,105 ones at progression stage(52.5%),32 ones at critical outbreak stage(16.0%)and 37 ones at resolution and absorption stage(18.5%),and there were 35.5%with unilateral lung involvement and 64.5%with bilateral involvement.At early stage,HAPE chest CT manifestations included increased and thickened bilateral bronchovascular bundles,widened main pulmonary artery lumen and faint ground-glass opacity in lungs;at progression stage,HAPE chest CT manifestations revealed multiple cloud-like or patchy areas of increased density within lungs;at critical outbreak stage,CT scanning indicated diffuse patchy opacities and consolidation in lungs,white lung-like changes could be found in some severe cases,and bronchial air signs were shown within affected segments in some ones with severer signs in the right lung than in the left lung;at resolution and absorption stage the CT manifestations were similar to those at early stage,with lesions completely resolving after treatment.The study group had the diameters of the main pulmonary arteries greater while the diameters of the ascending aortas less than those of the control group,and the ratios of the diameters of the two diameters in the study group were higher than those in the control group,with the differences being statistically significant(all P<0.05).Conclusion Chest CT is an important examination method for the early diagnosis of HAPE and clarification of its clinical staging,which directly reflects the pulmonary pathological changes in HAPE patients and helps doctors fully understand the disease progression.
4.Asperosaponin Ⅵ promotes osteogenic differentiation of MC3T3-E1 cells under hypoxia environment

BACKGROUND:Asperosaponin Ⅵ has good osteogenic effects,but its ability to promote cellular osteogenesis under hypoxia environment is not yet clear.OBJECTIVE:To investigate the effect and potential mechanism of Asperosaponin Ⅵ on osteogenic differentiation of MC3T3-E1 cells under hypoxia environment.METHODS:MC3T3-E1 cells were divided into three groups.Cells in the control group were cultured in a complete medium under normoxic conditions(volume fraction of 21%O2);cells in the hypoxia group were cultured in the complete medium under hypoxia conditions(volume fraction of 0.5%O2);and cells in the Asperosaponin Ⅵ group were cultured in the complete medium containing Asperosaponin Ⅵ under hypoxia conditions(volume fraction of 0.5%oxygen).After 24 hours of culture,cell counting kit-8 method and EdU staining were used to detect cell proliferation activity,TUNEL staining and western blot assay were performed to detect cell apoptosis,and flow cytometry was used to detect intracellular reactive oxygen species levels and cell cycle distribution.Each group of cells was cultured in an osteogenic induction medium containing 1×10-5,1×10-6,and 1×10-7 mol/L Asperosaponin Ⅵ under hypoxia conditions for 7 days.The optimal concentration of Asperosaponin Ⅵ for intervention was identified using alkaline phosphatase staining under optical microscopy.Western blot was used to detect the expression of bone morphogenetic protein 2,osteopontin,and PI3K/AKT signaling axis-related proteins.RESULTS AND CONCLUSION:(1)Compared with the control group,the proliferation ability of MC3T3-E1 cells decreased under hypoxia conditions.1×10-6 mol/L Asperosaponin Ⅵ could significantly improve the cell proliferation ability under hypoxia conditions and reduce cell apoptosis.(2)Compared with the hypoxia group,the Asperosaponin Ⅵ group showed a decrease in intracellular reactive oxygen species and a significant increase in the proportion of cells in the S phase.(3)Compared with the hypoxia group,the cell morphology in the 1×10-6 mol/L Asperosaponin Ⅵ group was elongated,with more protrusions and darker alkaline phosphatase staining.(4)Compared with the hypoxia group,the expression of bone morphogenetic protein 2 and osteopontin increased in the Asperosaponin Ⅵ group,while the expression of p-PI3K and p-AKT proteins decreased.These findings indicate that under hypoxia conditions,Asperosaponin Ⅵcan promote osteogenic differentiation of MC3T3-E1 cells,possibly by regulating the PI3K/AKT pathway.
5.The effectiveness of applying different tip positions of midline catheters:a Meta-analysis

Objective To evaluate the effectiveness of different tip positions applied to midline catheters(MC)and provide evidence-based evidence for venous catheter tip positioning in clinical practice.Methods Computerized searches of PubMed,Web of Science,Embase,Cochrane Library,CINAHL,CNKI,Wanfang Database,VIP,and CBM for studies on the effectiveness of applying MC with different tip positions were performed from the time of database construction to July 2024.Meta-analysis was performed using Rev Man 5.3 software after 2 investigators independently screened the studies,extracted the information and evaluated the quality of the included studies.Results A total of 9 studies with 2 302 hospitalized patients were included.The quality evaluation results of the included studies are all B-level.Meta-analysis showed that when the tip of the MC was located in the subclavian vein compared with the tip of the MC in the axillary vein,the rate of total catheter-related complications,phlebitis,blood leakage,infiltration,catheter occlusion,catheter dislocation,and catheter-associated thrombosis were lower,with a statistically significant difference(P<0.05).When the tip of the MC was located in the subclavian vein compared with the tip of the MC in the axillary vein,the catheter retention time was longer,with a statistically significant difference(P=0.007).The descriptive analysis showed a lower rate of extubation due to complications when the tip of the MC was located in the subclavian vein compared with when the tip was located in the axillary vein(P<0.05).Conclusion When the tip of the MC is located in the subclavian vein compared to when it is located in the axillary vein,the incidence of total catheter-related complications,phlebitis,blood leakage,infiltration,catheter occlusion,catheter dislocation,catheter-associated thrombosis,and the rate of catheter extractions due to complications were lower,and the catheter was left in place for a longer period of time.Due to the limitations of the quantity and quality of the included studies,more large-sample,high-quality studies are needed to further validate the effectiveness of different tip positions of MC.

7.An Epithelial Senescence Model Induced by Doxorubicin in MCF 10A Cells

This research aims to construct a stable epithelial cell senescence model for screening and e-valuation of senolytics.We explored the optimal conditions for doxorubicin-induced senescence of non-transformed epithelial cells MCF 10A,including the optimal induction concentration,the optimal inter-vention time,and the optimal senescence duration,and confirmed the feasibility of MCF 10A as an epi-thelial senescence model by multiple ways.The optimal condition for Doxorubicin-induced senescence of MCF 10A cells was treatment with 0.6 μmol/L Doxorubicin for 16 h to achieve the best senescence state on the 8th day.Under the optimal induction conditions,the positive rate of senescence-associated β-gal-actosidase(SA-β-gal)staining in the treated group reached 97%.At the same time,biochemical results of detecting the expression of mRNA,proteins,and immunofluorescence demonstrated that the expression levels of senescence-associated secretory phenotype(SASP),p16,p21 and p53 in the treated group were significantly higher than those in the control cells,and Lamin B1 was significantly decreased(P<0.001),which were consistent with the specific characteristics of senescence.In summary,an epithelial senescence model was successfully induced in MCF 10A cells by Doxorubicin in this study,which will promote the screening of senolytics for senescent epithelial cells.
8.Spatial distribution of iodine content in residential drinking water in Shaanxi Province in 2022

Objective:To study the spatial distribution and characteristics of iodine content in residential drinking water in Shaanxi Province, and provide a basis for scientific prevention and control of iodine deficiency disorders.Methods:From March to October 2022, an investigation was conducted in all counties (districts, referred to as counties) of Shaanxi Province, with administrative villages as survey units. For centralized water supply systems, two terminal water samples were collected, and iodine content was measured separately, with the average value representing the drinking water iodine content at that location. For decentralized water supply systems, a 10% sampling method was used to divide each administrative village into five areas (east, south, west, north, and center), with 10% of water wells sampled from each area (if fewer than 10 water wells, the well with the largest drinking population was selected; if fewer than 5 wells, all wells were sampled). One water sample was collected from each water well, and iodine content was determined using the arsenic-cerium catalytic spectrophotometry method. Spatial autocorrelation and spatial scan analysis were used to analyze the spatial distribution and characteristics of drinking water iodine content.Results:A total of 53 990 drinking water samples were collected, with a median water iodine content of 6.66 μg/L, ranging from 0.10 to 779.40 μg/L. Drinking water iodine content was detected in 112 counties, showing a significant spatial positive correlation (global autocorrelation, Moran's I = 0.74, Z = 43.83, P < 0.001). Local autocorrelation analysis showed that the difference in the distribution of iodine content in drinking water among 36 counties was statistically significant ( P < 0.05), with 22 counties showing low-low clustering and 12 counties showing high-high clustering. Spatial scanning identified three water iodine clustering areas, including two high-water iodine cluster areas and one low-water iodine cluster area. Conclusions:The distribution of iodine content in residential drinking water in Shaanxi Province shows significant spatial clustering and heterogeneity, requiring targeted interventions to achieve precise prevention and control of iodine deficiency disorders.
9.Clinical application of an intelligent puncture navigation system for percutaneous lung needle biopsy under CT guidance

Objective To explore the impact of an intelligent puncture navigation used by different physicians with varying years of experience to perform the lung puncture biopsy surgery.Methods A retrospective selection was conducted of 182 patients who completed lung puncture biopsy surgery.The primary parameters were recorded included puncture time,the number of needle adjust-ments,dose length product(DLP),and complications.The physicians were categorized into high-experience and low-experience groups based on their years of clinical practice.The differences of navigation guidance and manual puncture were compared between the two groups.Results The use of navigation guidance significantly reduced the procedure time for both groups of physicians(P<0.05).Additionally,for the low-experience group,navigation guidance notably decreased the number of needle adjustments(P<0.05)and reduced the radiation dose received by patients(P<0.05).Conclusion The application of intelligent puncture navigation can shorten the procedure time,reduce the number of needle adjustments,and lower the radiation dose received by patients in lung puncture biopsy procedures.It also bridges the operational performance gap between low-experience and high-experience physicians,making it a val-uable imaging-guided tool for widespread adoption.
10.Investigation of High-risk HPV Infection in Kashgar and Evaluation of Health Awareness Education Intervention

[Objective]To investigate the prevalence of high-risk HPV infection among women in Kashgar area,analyze related risk factors,conduct follow-up and health education interventions for positive cases in the past three years,and assess intervention outcomes to improve HPV screening awareness and reduce cervical cancer incidence.[Methods]This study randomly selected 571 women from Kashgar who underwent HPV screening as part of a health check-up in the First People's Hospital of Kashgar between January 2021 and June 2023.High-risk HPV-DNA was detected using the PCR-RDB method.Based on the test results,participants were divided into HPV-negative and HPV-positive groups,and the infection rates and associated risk factors were analyzed.Additionally,follow-up was conducted for women who tested positive for high-risk HPV during the screening to assess the effectiveness of health management interventions.[Results]From 2021 to 2023,the high-risk HPV positive rate among women in Kashgar showed a year-on-year decline,recorded at 19.15%,16.43%,and 11.30%,respectively(P<0.001),with a total of 3,921 cases(16.13%)testing positive for high-risk HPV during this period.Uyghur women had a significantly higher infection rate compared with those of Han nationality and other ethnic minorities(P<0.05).Risk factors included age,age at first sexual activity,number of sexual partners,contraceptive methods,and a history of vaginal inflammation(P<0.05).Health management interventions significantly improved participants'awareness scores regarding cervical cancer prevention and the proportion of women voluntarily receiving HPV vaccination(P<0.05).[Conclusions]High-risk HPV infection among Uyghur women in Kashgar area is relatively high,with single infection of HPV-16 and HPV-52 and mixed infections of HPV-16+HPV-52 and HPV-16+HPV-58 being predominant.Risk factors include age at first sexual activity,number of sexual partners,contraceptive methods,and history of vaginal inflammation.Through triple-education interventions,the incidence of cervical cancer in Kashgar has been effectively reduced,and standards for primary-level diagnosis,treatment,and health supervision have been improved.
