1.Mechanism of glaucocalyxin a in alleviating asthma airway remodeling by inhibiting mitochondrial fission through targeting the TOP2A/CDK1 signaling pathway

Objective To explore the mechanism of Glaucocalyxin A(GLA)in inhibiting ovalbumin(OVA)-induced airway remodeling in asthmatic mice through the topoisomerase Ⅱ α(TOP2A)/cyclin-dependent kinase 1(CDK1)signaling pathway.Methods Forty Balb/c mice were randomly divided into 5 groups:the control group,the model group,the low-dose GLA group,the high-dose GLA group and the Dexamethasone group,with 8 mice in each group.The effect of GLA on airway remodeling was examined by immunohistochemical staining,ELISA and other methods,and bioinformatics methods were used to predict new targets of GLA.The action targets of TOP2A were screened using the STRING database,and the interaction relationship between the two was verified by co-immunoprecipitation.In vitro,GLA and siRNA were used to interfere with interleukin-4(IL-4)-stimulated human airway epithelial cells BEAS-2B.The expressions of TOP2A,epidermal growth factor receptor(EGFR),Integrin β1,focal adhesion kinase(FAK),β-catenin,CDK1 and DRP1 were detected by Western Blot.Results GLA intervention could significantly reduce OVA-induced asthma airway remodeling,airway smooth muscle thickening,collagen deposition around the airway,the number of eosinophils in alveolar lavage fluid,the expression of pro-inflammatory cytokines such as IL-4,and the level of serum IgE.The new target of GLA screened out was TOP2A,which was highly expressed in the lung tissue of the asthma airway remodeling model.GLA intervention could down-regulate its expression.In vitro,intervention with GLA and si-TOP2A could significantly down-regulate the expressions of IL-4-induced TOP2A,EGFR,Integrin β1,FAK and β-catenin.Further studies have found that TOP2A had an interaction relationship with CDK1.si-TOP2A could downregulate the expression of CDK1,and knockdown of CDK1 could significantly down-regulate the expression of phosphorylated DRP1.Conclusion GLA may alleviate asthma airway remodeling by targeting the TOP2A/CDK1 signaling pathway,providing experimental evidence for the clinical diagnosis and treatment of asthma airway remodeling in asthma.

3.Influence of treatment combining feedforward control and trunk exercise on the integrated ability and activity in children with cerebral palsy

Objective To investigate the influence of treatment combining feedforward control and trunk exercise on the integrated ability and activity in children with cerebral palsy (CP). Methods 100 children with CP were divided into control group and observation group, with 50 patients in each group. Patients in control group were treated with routine trunk exercise while patients in observation group were treated with additional feedforward control. The follow-up lasted 6 months. The scores of WeeFIM, GMFM-88, activity of daily living (ADL) and quality of life in two groups before and 6 months after intervention were observed. Results Scores of WeeFIM and GMFM-88 of 6 months after interven tion were higher than those before intervention in two groups respectively(P<0. 05). Scores of WeeFIM and GMFM-88 in observation group 6 months after intervention were higher than those in control group(P<0. 05). Scores of ADL 6 months after intervention in two groups were significantly higher than before intervention respectively (P<0. 05). Score of ADL 6 months after intervention in intervention group was significantly higher than that in control group(P<0. 05). Results of two groups 6 months after intervention were significantly different from their results before intervention. Scores of social function, physiological function, emotional function and total score of PedsQL were higher than those before intervention respectively (P<0. 05). These scores in observation group 6 months after intervention were significantly higher than those in control group (P<0. 05). Conclusion The effectiveness of treatment combining trunk exercise and feedforward control in children with CP was significant. This treatment could improve the motor function, ADL and life quality of patients.
4.Effect of pramipexole in the treatment of Parkinson after stroke

Objective To investigate the effect of pramipexole in the treatment of Parkinson after stroke .Methods 94 cases with Parkinson after stroke in our hospital from June 2013 to November 2015 were randomly selected and divided into two groups,47 cases respectively.Control group received routine etiological and symptomatic treatment, the study group received conventional therapy and symptomatic treatment and pramipexole treatment,two groups were treated for eight weeks.Parkinson’s disease scores,inflammatory cytokines and oxidative stress index,and the clinical effect and complications of contrast were compared after the treatment.Results Compared with before treatment,scores of daily activities of emotion integral integral,the motor function in two groups decreased,levels of serum IL-1β,IL-6,TNF-αand Hs-CRP decreased,levels of CAT,T-SOD,T-GSH in plasma increased (P<0.05),and compared with the control group,scores of daily activities of emotion integral integral,the motor function in the study group were lower,levels of serum IL-1β,IL-6,TNF-αand Hs-CRP were lower,levels of CAT,T-SOD,T-GSH in plasma were higher (P<0.05),and the total effective rate in the study group 91.49% was higher than the control group 74.47% (P<0.05).Conclusion Pramipexole in the treatment of Parkinson after stroke was effective with high safety,and it can reduce Inflammatory factors and increase oxidative stress.
5.Resuscitation and nursing of idiopathic left ventricular tachycardia combined with ventricular fibrillation treated by radiofrequency ablation

Objective The explore resuscitation and nursing of ventricular fibrillation (VF)during the ablation of idiopathic left ventricular tachycardia (ILVT).Methods Radiofre-quency ablation guided by 3-D mapping system treated for ILVT.During the procedure,the de-fibrillator,the patches and the ventilator should be prepared.Results VF happened in 5 patients during ablation,and the resuscitation was successful after defibrillation and countermeasures.Con-clusion electrocardiograph monitoring,fully preparation before operation,active resuscitation and nursing countermeasures are the key to the success during the radiofrequency ablation.

7.The influence of trunk control training on motor function and the ability in the activities of daily living of patients with cerebral palsy

Objective To observe the influence of trunk control training on motor function and the ability of cerebral palsy (CP) patients in the activities of daily living (ADL).Methods Forty patients with CP were randomly divided into a treatment group (n =20) and a control group (n =20).Both groups were treated with routine rehabilitation,while the treatment group also received trunk control training.All patients were assessed with function ambulation category (FAC) classification,time to walk 10 m,the Berg balance scale (BBS),and the modified Barthel index (MBI) at the beginning and eight weeks later.Results Before the intervention there was no significant difference between the two groups in terms of any of the assessments.Eight weeks later,all the assessment scores were significantly better in the treatment group than in the control group.Conclusion Trunk control training can significantly improve motor function and the ADL ability of patients with CP.
8.Study of left ventricular systolic synchrony and sequence in patients with premature ventricular complexes from right ventricular outflow tract

Objective To evaluate left ventricular(LV) systolic synchrony and sequence in patients with premature ventricular complexes(PVCs) from right ventricular outflow tract (RVOT).Methods Thirty patients with frequent isolated PVCs from RVOT and 30 healthy subjects as control were included.Speckle tracking imaging (STI) was performed to assess the time-to-peak segmental systolic strain in longit udinal(TsL), circumferential (TsC) and radial (TsR) direction.The standard deviation (SD) of TsL,TsC and TsR of 18 LV segments were calculated respectively.All values of patients with PVCs were recorded during sinus beats(PVC-S) and PVC beats(PVC-V) respectively.LV systolic sequence in PVC-V was analyzed.Results Significant differences were observed in the SD values between the PVC-V and control subjects in three directions,as well as between the PVC-S and control subjects in circumferential and radial direction.In PVC-V significance difference was seen in TsL and TsR from apical to basal level,as well as in TsL and TsC in different walls.Conclusions LV systolic synchrony was demonstrated in patients with PVCs from RVOT during both sinus beats and PVC beats.Systolic sequence in PVC beats from RVOT exhibit certain rules.
9.Echocardiographic assessment of left ventricular global and segmental myocardial systolic function with pacing at variant right ventricular sites

Objective To evaluate the left ventricular(LV)global and segmental myocardial systolic function in patients with permanent DDD pacemaker implantation by speckle tracking imaging.Methods Myocardial imaging of LV apical long views and parasternal LV short axis views were performed in 9 patients with right ventricular(RV)septum pacing(RV septum group),15 patients with RV apex pacing(RV apex group)and 13 healthy subjects(control group).Peak systolic longitudinal strain(S_L),peak systolic radial strain(S_R)were measured,and the average of S_L and S_R among LV 18 segments were calculated respectively as corresponding GS_L and GS_R.Results The GS_L[-(18.29±2.67)%]of RV apex group showed significant decrease compared with RV septum group[-(20.54±2.29)%]and control group[-(21.07±2.08)%](P<0.05),while no significant difference was found between RV septum and control group.The GS_R of the pacing groups were both lower than control group[(36.26±9.37)%in control group,(28.59±6.06)%in RV septum group,(26.85±7.73)%in RV apex group,P<0.05],but RV apex group showed more decrease trend.The S_L and S_R of the LV myocardial segments close to the pacing sites in both pacing groups showed significant decrease compared with control group,but RV septum group preserved similar LV strain distribution with control group while RV apex group showed abnormal LV strain pattern.Conclusions Speckle tracking imaging is a useful modality to evaluate global and segmental myocardial systolic function of LV with pacing at different right ventricular sites.
10.Pharmacodynamic Studies of Huoluojiangu Capsule on Analgesic Experiments

Objective To observe effect of Huoluojiangu capsule (HJC) on analgesic experiments. Methods The 3 kinds of analgesic experiments of whirling body, electric irritation and hot board were respectively adopted in mice. 60 mice were randomly divided into 6 groups in every analgesic experiment:A group(blank control), normal saline 0.25 mL/10 g. B group (positive control), diclofenac sodium 3.5 mg/kg;C group (positive control), pethidine 15 mg/kg, D,E and F group were respectively as low,mid and high dose of HJC groups (HJC 14, 21, 28 g/kg). Afterward, effects of HJC on ache induced by chemistry, electricity and heat were respectively observed at different time after pretreatments. Results Low, mid and high dose of HJC groups can all inhibit reaction of whirling body by chemical irritation. Compared with blank control group, low dose of group had evidently effect, P 0.05), but it had stronger analgesic effect than positive groups (B and C groups) at 180 min, P
