Objective Investigate the expression level of discs large homolog associated protein 5(DLGAP5)in oral squamous cell carcinoma(OSCC)and analyze its effects on cell proliferation, migration, invasion capacity, and the Warburg effect. Methods Bioinformatics analysis was performed to identify the potential therapeutic targets for OSCC.A total of 72 OSCC tissue samples and 40 adjacent non-cancerous tissue samples collected in the First Affiliated Hospital of Shihezi University from 2013 to 2024 were included, and the clinical pathological and prognostic data were collected from patients.Immunohistochemistry assay was applied to detect the protein expression of DLGAP5,and its association with clinical pathological features was analyzed.Kaplan-Meier survival curve was plotted for survival analysis, and Cox regression model was employed to analyze the prognostic factors. The expression of DLGAP5 at mRNA and protein levels was detected in HOK,SCC-9,SCC-15,SCC-25,and CAL-27 cell lines with RT-qPCR and Western blotting, respectively.Four small interfering RNAs(siRNAs)were designed to target the DLGAP5 sequence, and then based on the transfection efficiency, the sequence with optimal silencing effect was selected for subsequent functional studies. After DLGAP5 was silenced in the CAL-27 and SCC-15 cells,Western blotting was applied to detect the expression of hexokinase 2(HK2)and enolase 1(ENO1),CCK-8,scratch healing and Transwell assays were conducted to assess cell proliferation, migration, and invasion capabilities, and glucose, lactate, and ATP detection kits were utilized to determine the glycolytic metabolic levels in OSCC cells. Results Bioinformatics analysis indicates that DLGAP5 is a potential key therapeutic target for OSCC.Experimental validation demonstrated that DLGAP5 was highly expressed in both OSCC tissues and cells(P<0.05).Analysis of clinical pathology and prognostic data revealed that DLGAP5 expression level was significantly correlated with tumor TNM stage, lymph node metastasis, and differentiation grade in OSCC patients, and high DLGAP5 expression was associated with poor prognosis(P<0.05).DLGAP5 silencing resulted in significantly reduced expression of HK2 and ENO1,markedly decreased levels of glycolytic metabolites(P<0.05),and notably declined cell proliferation, migration, and invasion capabilities(P<0.05).Conclusion DLGAP5 is highly expressed in OSCC.Silencing DLGAP5 may inhibit OSCC cell proliferation, migration, and invasion by indirectly regulating the Warburg effect, and the molecule is associated with poor prognosis in the OSCC patients.

Objective :
To explore differential expression of insulin like growth factor 1( IGF1) in oral squamous cell carcinoma (OSCC) tissues and cells , and to analyze its clinical significance of IGF1 .
Methods :
The protein expression of IGF1 was detected in 115 OSCC tissues (OSCC group) and 74 normal tissues (normal group) using immunohistochemical technique , and the relationship with the clinicopathological factors and prognosis of OSCC was analyzed. The IGF1 protein and mRNA levels in HOK and OSCC cell lines ( CAL⁃27 , TCA⁃8113 , SCC⁃15 and SCC⁃25) were detected by Western blot and qRT⁃PCR.
Results :
The high expression rate of IGF1 in OSCC group was 72. 17% , which was significantly higher than that in normal group (2. 70% ) ( P < 0. 001) . The proportion under ROC curve (AUC) of OSCC diagnosed by IGF1 was 0. 81 , the sensitivity was 0. 73 , and the specificity was 0. 82. The expression of IGF1 was related to the degree of differentiation , T stage and depth of invasion (P = 0. 03 , P = 0. 02 , P = 0. 02) , but not to gender, age , N stage , TNM stage , smoking , alcohol consumption , or HPV infection (P > 0. 05) . Kaplan⁃Meier and COX regression analysis showed that the high expression of IGF1 , the degree of differentiation , the T stage and the depth of infiltration were the related factors affecting the prognosis of patients (P < 0. 01 , P = 0. 04 , P = 0. 03 , P = 0. 04) . COX multivariate indicated that high expression of IGF1 was an independent factor affecting the prognosis of patients ( P = 0. 01) . Western blot and qRT⁃PCR results indicated that the expression of IGF1 in OSCC cell lines was higher than that in HOK (P < 0. 05) .
Conclusion
IGF1 can be a potential diagnostic and poor prognostic marker in oral squamous cell carcinoma.

Bone substitute material implantation has become an important treatment strategy for the repair of oral and maxillofacial bone defects. Recent studies have shown that appropriate inflammatory and immune cells are essential factors in the process of osteoinduction of bone substitute materials. Previous studies have mainly focused on innate immune cells such as macrophages. In our previous work, we found that T lymphocytes, as adaptive immune cells, are also essential in the osteoinduction procedure. As the most important antigen-presenting cell, whether dendritic cells (DCs) can recognize non-antigen biomaterials and participate in osteoinduction was still unclear. In this study, we found that surgical trauma associated with materials implantation induces necrocytosis, and this causes the release of high mobility group protein-1 (HMGB1), which is adsorbed on the surface of bone substitute materials. Subsequently, HMGB1-adsorbed materials were recognized by the TLR4-MYD88-NFκB signal axis of dendritic cells, and the inflammatory response was activated. Finally, activated DCs release regeneration-related chemokines, recruit mesenchymal stem cells, and initiate the osteoinduction process. This study sheds light on the immune-regeneration process after bone substitute materials implantation, points out a potential direction for the development of bone substitute materials, and provides guidance for the development of clinical surgical methods.

As a novel and promising antitumor target, AXL plays an important role in tumor growth, metastasis, immunosuppression and drug resistance of various malignancies, which has attracted extensive research interest in recent years. In this study, by employing the structure-based drug design and bioisosterism strategies, we designed and synthesized in total 54 novel AXL inhibitors featuring a fused-pyrazolone carboxamide scaffold, of which up to 20 compounds exhibited excellent AXL kinase and BaF3/TEL-AXL cell viability inhibitions. Notably, compound 59 showed a desirable AXL kinase inhibitory activity (IC50: 3.5 nmol/L) as well as good kinase selectivity, and it effectively blocked the cellular AXL signaling. In turn, compound 59 could potently inhibit BaF3/TEL-AXL cell viability (IC50: 1.5 nmol/L) and significantly suppress GAS6/AXL-mediated cancer cell invasion, migration and wound healing at the nanomolar level. More importantly, compound 59 oral administration showed good pharmacokinetic profile and in vivo antitumor efficiency, in which we observed significant AXL phosphorylation suppression, and its antitumor efficacy at 20 mg/kg (qd) was comparable to that of BGB324 at 50 mg/kg (bid), the most advanced AXL inhibitor. Taken together, this work provided a valuable lead compound as a potential AXL inhibitor for the further antitumor drug development.
