Objective:To investigate the factors that influence the number of oocytes retrieved of patients with polycystic ovary syndrome (PCOS) undergoing unstimulated natural cycles and the association between the number of oocytes retrieved and in vitro maturation (IVM) outcomes. Methods:This retrospective cohort study reviewed clinical data of 586 patients with PCOS undergoing unstimulated natural cycle at Reproductive Medicine Center, Department of Obstetrics and Gynecology of Peking University Third Hospital from 2006 January 1 to 2019 December 31. All patients were divided into three groups according to the number of oocytes retrieved (0-8, 9-19, ≥20). Basic information, basal endocrine hormones, oocyte and embryo development index and clinical outcomes were compared among the three groups.Results:Luteinizing hormone (LH)/follicle-stimulating hormone (FSH) [1.00(0.61,1.76), 1.41 (0.86, 1.96), 1.62(0.96, 2.14)], testosterone [1.07 (0.69, 1.91) nmol/L, 1.28 (0.77, 1.95) nmol/L, 1.67 (1.03, 2.75) nmol/L] and antral follicle count (AFC) [24 (19,24), 24 (24, 30), 30 (24, 40)] were significantly different among 0-8 group, 9-19 group and ≥20 group (all P<0.001). Multivariate logistic regression analysis showed that AFC was a significant factor affecting the number of oocytes retrieved ( P9-19/0-8=0.002, P≥20/0-8<0.001), and LH/FSH was an significant internal factor affecting the number of oocytes retrieved ( P9-19/0-8=0.006, P≥20/0-8=0.003) after the removal of AFC. Results of the analysis in patients after propensity score matching (PSM) showed that the number of oocytes retrieved was negatively correlated with oocyte maturation rate [0-8 group: 48.0% (242/504), 9-19 group: 44.8% (539/1202), ≥20 group: 40.4% (1067/2640), P=0.001], fertilization rate [0-8 group: 31.7% (160/504), 9-19 group: 25.5% (306/1202), ≥20 group∶ 23.7% (625/2640), P=0.001] and transferable embryo rate [0-8 group: 19.2% (97/504), 9-19 group: 11.6% (140/1202),≥20 group: 6.0% (153/2540), P<0.001], while positively correlated with transferable embryo formation cycle rate [0-8 group: 51.1% (47/92), 9-19 group: 66.3% (61/92), ≥20 group: 73.9% (68/92), P=0.005]. There were no differences in clinical pregnancy rate and live birth rate per fresh or frozen embryo transfer cycles, cumulative clinical pregnancy rate and cumulative live birth rate among the three groups. Conclusion:AFC has predictive value for the number of oocytes retrieved in natural cycles, and LH/FSH is an important internal factor affecting the number of oocytes retrieved. Oocyte maturation quality and development potential decrease with the increase of the number of oocytes retrieved in natural cycles, which has no direct effect on clinical outcomes.

Objective:The standard treatment for inoperable locally advanced esophageal cancer is concurrent chemoradiotherapy, but the survival was not satisfied. Nituzumab is a humanized IgG monoclonal antibody against EGFR. The purpose of this study is to investigate the toxicity and efficacy of concurrent chemoradiotherapy combined with nituzumab for locally advanced esophageal cancer.Methods:We retrospectively reviewed the clinical data of locally advanced esophageal cancer who were treated with concurrent chemoradiotherapy combined with nituzumab in Peking University Cancer Hospital from June 2015 to June 2020. Kaplan- Meier method was used for analysis. Results:Thirty Patients were enrolled this study.After a median follow-up of 22.5 months, The objective response rate was 93%. The 1-year, 2-year, 3-year overall survival rates were 83%, 57% and 41%, with the progression-free survival rates 75%, 47% and 32%, with the local-recurrence free survival rates 83%, 53% and 37%, with the metastasis-free survival rates 75%, 51% and 36%, respectively.The incidence of grade≥3 hematological toxicity was 32%. There were 16% patients experiencing grade≥3 esophagitis.Conclusion:The preliminary result of concurrent chemoradiotherapy combined with nituzumab is effective and safe for patients with locally advanced esophageal cancer.

Objective:To explore the predictive value of age, anti-Müllerian hormone (AMH) and antral follicle count (AFC) for the outcome of in vitro fertilization/intracytoplasmic sperm injection (IVF/ICSI) in the expected low prognosis population according to the POSEIDON criteria. Methods:This retrospective study was performed in patients who underwent IVF/ICSI and were expected low prognosis of ovarian stimulation from March 1, 2017 to December 31, 2018 in Reproductive Medicine Center, Department of Obstetrics and Gynecology, Peking University Third Hospital. According to three of the POSEIDON classification index, cycles were divided into 5 groups: group A (age<35 years, AFC<5 and AMH<1.2 μg/L), group B (age≥35 years, AFC<5 and AMH<1.2 μg/L), group C (AFC<5 and AMH≥1.2 μg/L), group D (age<35 years, AFC≥5 and AMH<1.2 μg/L) and group E (age≥35 years, AFC≥5 and AMH<1.2 μg/L). The ovarian stimulation and pregnancy outcomes were analyzed.Results:A total of 6745 IVF/ICSI cycles were included. The proportions of mini-stimulation and natural cycles in group A [15.8% (158/1000), 6.4% (64/1000)] and group B [28.1% (659/2342), 17.7% (414/2342)] were significantly higher than those in groups C, D and E [group C 6.4% (54/845), 1.1% (9/845); group D 5.8% (63/1077), 0.8% (9/1077); group E 11.0% (163/1481), 0.9% (14/1481), P<0.001]. The number of retrieved oocytes in groups A and B [4(2,6), 3(2,5)] was significantly lower than that in groups D and E [7(4,10), 6(3,8)] ( P<0.001). The clinical pregnancy rate [39.2% (200/510), 37.2% (272/731)], the implantation rate [28.4% (251/883), 26.7% (355/1332)], and the live birth rate [24.9% (127/510), 22.3% (163/731)] of young patients in groups A and D were significantly higher than those of older patients [group B: 21.5% (238/1109), 14.9% (266/1787), 10.4% (115/1109), group E: 23.5% (212/903), 15.4% (243/1581), 11.8% (107/903)] ( P<0.001). The cancellation rate of transplantation in groups A and B [49.0% (490/1000), 52.6% (1233/2342)] was significantly higher than that in the other groups [group C 34.1% (288/845), group D 32.1% (346/1077), group E 39.0% (578/1481), P<0.001], and among the cancellation reasons, the proportion of no embryos or no retrieved eggs in groups A and B was also higher. Conclusion:The POSEIDON criteria has a good predictive value on expected low prognosis patients. AMH, which is an objective index, represents a higher clinical pregnancy rate, while higher AFC indicate more retrieved eggs. In young patients with low ovarian reserve (POSEIDON group 3), clinical pregnancy rate per cycle is moderate, but due to a high cancellation rate because of no embryos or no retrieved eggs, live birth rate per egg retrieval cycle is still low.

BACKGROUND: The standard treatment for locally advanced non-small cell lung cancer (NSCLC) is radiotherapy concurrent with chemotherapy, but the survival was not satisfied. With the development of intensity modulated radiotherapy, simultaneous integrated boost technique (SIB) becomes the research direction of locally advanced NSCLC. The aim of this study is to investigate the efficacy and safety of SIB intensity modulated radiotherapy technique for locally advanced NSCLC. METHODS: We retrospectively reviewed the clinical data of locally advanced NSCLC who were treated with radiotherapy by SIB technique in Peking University Cancer Hospital from June 2015 to December 2018. Kaplan-Meier method was used for analysis. RESULTS: Ninty-three patients were included in the analysis. After a median follow-up of 34.23 months, 3-year overall survival (OS), progression-free survival (PFS), local-recurrence free survival (LRFS) and metastasis free survival (MFS) rates were 53.0%, 37.0%, 50.5% and 50.5%, respectively. The incidence of grade ≥3 esophagitis was 5.4%. There were 2 (2.2%) patients experiencing grade ≥3 radiation-related pneumonia. CONCLUSIONS Radiation with SIB intensity modulated radiotherapy technique is effective and safe for patients with locally advanced NSCLC.

Objective: To investigate the incidence and risk factors of peripartum mood disorder (PPMD) in order to improve clinical prevention and intervention of this condition. Methods: This was a prospective cohort study recruiting first-trimester pregnant women (<13 gestational weeks) from Beijing Daxing Maternal and Child Care Hospital from October 1, 2016 to December 31, 2017. Zung Self-rating Anxiety Scale (SAS) and Zung Self-rating Depression Scale (SDS) were used to evaluate the anxiety and depression status in the second and third trimesters, respectively. Their life styles, social and environmental factors exposure during pregnancy were also collected. Statistical analysis was conducted using Chi-square test, student's t test, Mann-Whitney U test and multivariate logistic regression. Results: (1) A total of 478 subjects were enrolled in early pregnancy and 460 (96.2%) and 431 (90.2%) of them completed SAS and SDS assessment in the second and third trimesters, respectively, and 413 (86.4%) of the subjects finished both assessments on two occasions. (2) The prevalence of anxiety and depression was 7.1% (30/425) and 13.9% (59/425) in the second trimester, and 11.2% (44/392) and 21.5% (84/390) in the third. The incidence of anxiety and depression in the third trimester was 7.7% (26/336) and 9.6% (30/313). (3) Cross-sectional data analysis showed that prolonged television watching was a risk factor of anxiety in the second (OR=1.216, 95%CI: 1.055-1.402) and third (OR=1.166, 95%CI: 1.044-1.303) trimester, while exercise was a protect factor (OR=0.238, 95%CI: 0.105-0.541; OR=0.432, 95%CI: 0.212-0.879). Pregnant women with longer sleeping time had lower risks of depression in the second trimester (OR=0.725, 95%CI: 0.554-0.950); those who did exercise had lower risks of depression in the third trimester (OR=0.450, 95%CI: 0.252-0.803). (4) Longitudinal-data analysis revealed that longer television watching time in the second trimester was a risk factor for anxiety (OR=1.264, 95%CI: 1.117-1.432) and depression (OR=1.119, 95%CI: 1.005-1.246) in the third trimester. Conclusions The prevalence of anxiety and depression in the third trimester is higher than that in the second trimester. The incidence of depression is higher than that of anxiety in the third trimester. Prolonged television viewing time in the second trimester is a risk factor for both anxiety and depression in the third trimester.

OBJECTIVETo investigate the biological characteristics and genetic features of human placenta mesenchymal stem cells (hPA-MSCs) cultured in vitro in order to assess its safety for clinical use.

METHODSThe shapes of the 1st, 3rd, 5th, 7th, 10th, 13th, 17th and 20th generation hPA-MSCs cultured in vitro using serum-free culture medium were observed. Their cell cycle, cell surface markers, and karyotype were analyzed, and relevant genes and cytokines were measured.

RESULTSThe shape of hPA-MSCs has remained as fusiform or short fusiform, and there was no significant change. About 93% of hPA-MSCs cells were in G0/G1 phase and remained stable. No obvious chromosomal translocation, loss or inversion was noted by karyotyping analysis. Cytokines expression level remained stable. Related gene expression level as a whole was on the decline, but the gene expression level of the first five generations showed very slight variations, with genetic characteristics remaining stable.

CONCLUSIONThe hPA-MSCs cultured in vitro with serum-free medium has retained stable in the first five generations.

BACKGROUND:Currently, the research about effect of non-dextran coated superparamagnetic iron oxide nanoparticles on cellproliferation and cytotoxicity is relatively much less. OBJECTIVE:To evaluate the effects of 0, 25, 50, 75, 100 mg/L non-dextran coated superparamagnetic iron oxide nanoparticles on the proliferation and cytotoxicity of rat bone marrow mesenchymal stem cels. METHODS:Culture media containing 0, 25, 50, 75, 100 mg/L non-dextran coated superparamagnetic iron oxide nanoparticles were prepared for culture of bone marrow mesenchymal stem cels. After 24 hours of culture, the cels were confirmed using Prussian blue staining, and cellcounting was detected using cellcounting kit-8. Meanwhile, lactate dehydrogenase activity in the supernatant and intracelular superoxide dismutase activity were detected. RESULTS AND CONCLUSION:Loading of non-dextran coated superparamagnetic iron oxide nanoparticles in BMSCs was confirmed by Prussian blue staining. The percentage of cels labeled with non-dextran coated superparamagnetic iron oxide nanoparticles was up to 100% when the cels were incubated with a non-dextran coated superparamagnetic iron oxide nanoparticle solution of 50 mg/L and above, but 25 mg/L was insufficient to label al of the cels. Furthermore, as the concentration of non-dextran coated superparamagnetic iron oxide nanoparticles decreased, the cellproliferation rate decreased gradualy. The 25 mg/L group had a minimum cellproliferation rate, but the 25 and 50 mg/L groups showed no statisticaly significant difference (P > 0.05). Therefore, 50 mg/L is considered as the appropriate concentration of non-dextran coated superparamagnetic iron oxide nanoparticles, under which, the labeling efficiency is higher and the cytotoxicity is lower.

Background and purpose:Breast masses is woman's common diease,With the development of people's living.They are eager to find a new method which is efficient and less pain to replace conventional open surgery.So Mammotone appears.We assessed the efficacy of Mammotome biopsy system for the patients with single and multiple breast masses.We assessed the efficacy of Mammotome biopsy system for patients with single and multiple breast masses.Methods:From Janurary 2004 to April 2005,patients with single and multiple breast masses underwent Mammotome and conventional surgery respectively.Two methods has been compared from the aspects of difficulties,side effects,prognosis and degree of patient's satisfaction.Results:The length of excisions,anesthetic dosage,operational time,pain etc with Mammtome group were superior to the conventional group,especially for the patients with multiple breast masses.There were no difference in terms of bleeding during or after operation for two groups.Patients were followed up 3 to 15 months,none of the patients had relapse and patient's satisfaction was very encouraging.Conclusions:The color guided Mammotome showed very promising results for the patients with breast benign masses,and it was very useful for the masses either located deeply or were multiple.
