BACKGROUND:Tyrosine kinase inhibitors,as well as other types of small-molecule cancer drugs, can cause severe cardiotoxicity. OBJECTIVE:To perform a heart safety re-evaluation by observing the effects of antitumor drugs on isolated heart electrocardiograph, cardiac action potential and associated ion channels and cytotoxicity. METHODS:Extracorporeal cardiac perfusion was given to the isolated rabbit heart using Langendorff perfusion:Sunitinib(0.3,3,10 μmol/L),Crizotinib(0.3,1,3 μmol/L),and Doxorubicin(1,30 μmol/L)were perfused sequentially for 120 minutes to record electrocardiograph and left ventricular pressure.A blank control group was set for comparison.Manual patch clamp was used to record the effects of Crizotinib,Sunitinib,Doxorubicin on hERG,Cav1.2,Nav1.5 channel currents and action potential in human induced pluripotent stem cell derived cardiomyocytes.Adenosine triphosphate level in human induced pluripotent stem cell derived cardiomyocytes was detected by CellTiter-Glo luminescent cell viability assay. RESULTS AND CONCLUSION:Isolated rabbit heart using Langendorff perfusion:Compared with the blank ontrol group,Sunitinib and Crizotinib at≥3 μmol/L decreased heart rate(P<0.01)and prolonged QT/QTc interval(P<0.01),and reduced left ventricular pressure to different extents.Manual patch clamp recording:Compared with the blank control group,Sunitinib and Crizotinib at 3 μmol/L inhibited the activities of hERG,Nav1.5 and Cav1.2 channels and significantly prolonged the duration of action potential(P<0.01).According to the analysis of the test article, the difference between the labeled concentration and the measured concentration of the recovered solution was not significant.Cell viability assays:Compared with the blank control group, adenosine triphosphate content in human induced pluripotent stem cell derived cardiomyocytes significantly decreased after treatment with Sunitinib(IC50=4.64 μmol/L),Doxorubicin(IC50=4.21 μmol/L)and Crizotinib(IC50=2.87 μmol/L),indicating that cell viability significantly decreased(P<0.01).To conclude, this study successfully established an early cardiac safety evaluation method for antitumor drugs, which provides good support and help for the subsequent development of antitumor drugs.

Objective To observe the application effect of nurses-dominated rehabilitation management mode in acute stroke patients. Methods A total of 86 patients with acute ischemic stroke (AIS) in the department of neurology were selected for the study and were randomly divided into observation group (n =43) and control group (n =43) according to the order of admission. All patients were given routine treatment and nursing, and the control group was given the routine rehabilitation training for self-choice after in a stable condition, and the observation group was followed by a multidisciplinary team consultation to carry out the early comprehensive rehabilitation training. The activities of daily living in the two groups, nerve function, nutrition risk condition were compared before intervention and at 14d after intervention, and the related complications within 1 month after intervention were analyzed between the two groups. Results At 7 d after intervention, the MBI score and nutritional adequacy rate of the observation group were significantly higher than intervention before and were significantly higher than that of the control group (P < 0.05).The NIHSS score and nutritional risk rate of the observation group were significantly lower than intervention before and were significantly lower than that of the control group (P < 0.05).At 14 d and 1 month after intervention, the MBI score and nutritional adequacy rate of the two groups were significantly higher than intervention before, and those in the observation group were significantly higher than that of the control group at the same time period (P < 0.05).The NIHSS score and nutritional risk rate of the two groups were significantly lower than intervention before, and the observation group were lower than the control group at the same time period (P <0.05).The incidence rates of post-stroke depression,brain-induced spasm, swallowing dysfunction and shoulder-hand syndrome were significantly lower in the observation group than that in the control group within 1 month after intervention (P < 0.05).Conclusion Nurses-dominated ASU rehabilitation management mode can effectively improve the rehabilitation effect of patients.
7.Investigation and analysis of the sources of stress on entrepreneurship and employment of undergraduate nursing graduates from the university of Traditional Chinese Medicine
Chinese Journal of Modern Nursing 2017;23(23):3050-3052
Objective To investigate the sources of stress on entrepreneurship and employment of graduate nursing students from Traditional Chinese Medicine (TCM) and analyze its influencing factors and measures. Methods A questionnaire was designed from June to October in 2016. 82 students among 680 students who completed internship tasks from School of Nursing of Shandong University of TCM were randomly surveyed by random number table method. We statistically analyzed the survey data. Results Among all of the 82 students,58.53% of them expected to obtain employment in a Triple-A Grade-Ⅲ hospital. 21.95% of them expected to work in the colleges and universities. 52.44% of them selected to take up an occupation in major cities. Undergraduate graduates had greater pressure on entrepreneurship and employment. The main sources of stress were the factors of Chinese medicine institutions, individual factors, entrepreneurship and employment environment factors. Conclusions Undergraduate graduates have many sources of pressure on entrepreneurship and employment. It is necessary for the school and the employers to provide guidance on entrepreneurship and employment for nursing students in order to alleviate the pressure of entrepreneurship and employment and improve the quality of employment.

Objective To investigate the effects of global registry of acute coronary events (GRACE) and Can Rapid risk stratification of unstable angina patients suppress adverse outcomes with early implementation of the ACC/AHA guidelines(CRUSADE) score in the nursing care for patients with acute coronary syndrome (ACS).Methods A total of 128 patients with ACS undergoing percutaneous coronary intervention (PCI) who were admitted from March 2015 to May 2016 were enrolled in this study and divided into usual care group (63 cases) and optimized care group (65 cases). Patients in the usual care group received usual care according to the guidelines, while patients in the optimized care group received optimized care according to GRACE and CRUSADE score. The major adverse cardiac events, the incidence of bleeding complications, average hospitalization and hospitalization costs were recorded and compared between the two groups. Results The characteristics of angiography (multiple vessel rate and target vessel distribution) and procedures of PCI (position, number, length and diameter of stents, coronary blood flow classification after the implantation of stents) were similar (all P>0.05). There were tendencies of reduction in the incidence of MACE, bleeding complications and average hospitalization time in patients of optimized care group compared to the usual care group, with no statistically significant differences between the two groups (P>0.05). The hospitalization costs in the optimized care group (33491±1982)RMB were significantly less than those in the usual care group (36562±2395)RMB. The difference was statistically significant (t=-7.914,P<0.001).Conclusions Optimized care according to GRACE and CRUSADE can improve the treatment effects of ACS patients undergoing PCI and decrease the cost.
