In this paper, by referring to ancient and modern literature, the textual research of Inulae Flos has been conducted to clarify the name, origin, production area, quality evaluation, harvesting, processing and others, so as to provide reference and basis for the development and utilization of famous classical formulas containing this herb. After textual research, it could be verified that the medicinal use of Inulae Flos was first recorded in Shennong Bencaojing of the Han dynasty. In successive dynasties, Xuanfuhua has been taken as the official name, and it also has other alternative names such as Jinfeicao, Daogeng and Jinqianhua. The period before the Song and Yuan dynasties, the main origin of Inulae Flos was the Asteraceae plant Inula japonica, and from the Ming and Qing dynasties to the present, I. japonica and I. britannica are the primary source. In addition to the dominant basal species, there are also regional species such as I. linariifolia, I. helianthus-aquatili, and I. hupehensis. The earliest recorded production areas in ancient times were Henan, Hubei and other places, and the literature records that it has been distributed throughout the country since modern times. The medicinal part is its flower, the harvesting and processing method recorded in the past dynasties is mainly harvested in the fifth and ninth lunar months, and dried in the sun, and the modern harvesting is mostly harvested in summer and autumn when the flowers bloom, in order to remove impurities, dry in the shade or dry in the sun. In addition, the roots, whole herbs and aerial parts are used as medicinal materials. In ancient times, there were no records about the quality of Inulae Flos, and in modern times, it is generally believed that the quality of complete flower structure, small receptacles, large blooms, yellow petals, long filaments, many fluffs, no fragments, and no branches is better. Ancient processing methods primarily involved cleaning, steaming, and sun-drying, supplemented by techniques such as boiling, roasting, burning, simmering, stir-frying, and honey-processing. Modern processing focuses mainly on cleaning the stems and leaves before use. Regarding the medicinal properties, ancient texts describe it as salty and sweet in taste, slightly warm in nature, and mildly toxic. Modern studies characterize it as bitter, pungent, and salty in taste, with a slightly warm nature. Its therapeutic effects remain consistent across eras, including descending Qi, resolving phlegm, promoting diuresis, and stopping vomiting. Based on the research results, it is recommended that when developing famous classical formulas containing Inulae Flos, either I. japonica or I. britannica should be used as the medicinal source. Processing methods should follow formula requirements, where no processing instructions are specified, the raw products may be used after cleaning.

ObjectiveTo analyze the long-term trend of incidence and mortality of type 2 diabetic kidney disease (DKD) in China from 1990 to 2021. MethodsThe Joinpoint regression model was used to analyze the average annual percentage change (AAPC) of standardized incidence rate and standardized mortality rate, and the age-period-cohort (APC) model was constructed to analyze the longitudinal age change, period and cohort effect risk ratio (RR). ResultsFrom 1990 to 2021, the standardized incidence rate of type 2 DKD in males and females showed an overall upward trend, with AAPC of 0.08% and 0.36%, respectively. The age-standardized mortality rate of the total population and female showed a downward trend, with AAPC of -0.61% and -1.03%, respectively. However, there was no significant difference in males. APC model showed that the age effect existed: the peak age was 75-79 years old, the mortality rate of females increased, and the mortality rate of males decreased after 80-84 years old. For the effect of time period, the risk of type 2 DKD incidence in females in 2017—2021 was 1.05 times that in 2002—2006, and the risk of death in males and females in 2017—2021 was 0.84 and 0.71 times that in 2002—2006, respectively. For cohort effects, the highest risk of disease was seen in men and women born in 1967—1971, and the highest risk of death was seen in men born in 1952—1956 and women born in 1912—1916. ConclusionFrom 1990 to 2021, the standardized incidence rate of type 2 DKD in China shows an upward trend, and the standardized mortality rate shows a downward trend. It is necessary to strengthen the health behavior publicity and education of type 2 DKD, and actively carry out early screening to reduce the disease burden.

ObjectiveTo investigate the effects of targeted silencing of Runt-related Transcription Factor 3 (RUNX3) on the proliferation and migration of Mouse Hepatic Stellate Cells (HSCs), as well as subsequent collagen deposition. MethodsMouse hepatic stellate cell line (JS-1) was selected and then morphologically observed and identified under a microscope. After the cells had fully adhered, they were treated with 5 ng/mL of transforming growth factor beta 1 (TGF-β1) for 24 hours to induce hepatic stellate cell activation. Furthermore, a RUNX3 silencing model was established using RUNX3 lentiviral infection. The experiment was divided into four groups: Control group, TGF-β1 group, TGF-β1+siRNA-NC group, and TGF-β1+siRNA-RUNX3 group. Protein expression changes of RUNX3, alpha-smooth muscle actin (α-SMA), and Alpha 1 type I collagen (Collagen I) were detected using Western blot method. Cellular immunofluorescence assays were employed to investigate the deposition changes of α-SMA and RUNX3 in hepatic stellate cells. RT-qPCR was utilized to examine the mRNA expression changes of RUNX3, α-SMA, and Collagen I. The proliferative capacity of hepatic stellate cells was assessed using Edu staining. The migratory ability of hepatic stellate cells was evaluated through wound healing assays and Transwell migration experiments. ResultsCompared with Control group, a significant elevation in RUNX3 was observed in the TGF-β1-induced activated HSCs (P<0.01). Meanwhile, the protein and mRNA levels of fibrosis-related markers and α-SMA and Collagen I were significantly upregulated (P<0.001). Additionally, the proliferation and migration capabilities of HSCs were significantly enhanced (P<0.001). In contrast, when compared to TGF-β1+siRNA-NC group, TGF-β1+siRNA-RUNX3 group exhibited a notable decrease in RUNX3 and other related indicators, such as the protein and mRNA levels of α-SMA and Collagen I (P<0.05). Concurrently, the proliferation and migration capabilities of HSCs were significantly inhibited in TGF-β1+siRNA-RUNX3 group (P<0.01). ConclusionSilencing RUNX3 can inhibit the deposition of collagen and the proliferation and migration of hepatic stellate cells. Conversely, RUNX3 promotes the proliferation and migration capabilities of HSCs, thereby facilitating the activation of HSC.

Objective:To evaluate the efficacy and safety of nebulized inhalation of recombinant human interferon (IFN) α1b injection in the treatment of respiratory syncytial virus (RSV) associated lower respiratory tract infections (pneumonia and bronchiolitis) in children. Methods:A randomized, double-blind, parallel, placebo-controlled add-on design was used.Children with pneumonia or bronchiolitis aged 2 months to 5 years who tested positive for RSV antigen within 72 hours of onset from 30 clinical trial sites including Beijing Children′s Hospital, Capital Medical University between February 2021 and December 2022 were included in this study and randomly divided into 2 groups at a ratio of 1∶1 based on a stratified-block method. Both groups received basic treatments such as cough control, asthma relieving, expectorant treatment, fever reduction, oxygen therapy, etc.The experimental group received additional nebulized inhalation of IFN α1b injection at a dose of 2.0 μg/(kg·time), twice a day.The control group received nebulized inhalation of placebo twice a day.Clinical efficacy was evaluated based on indicators such as the duration of clinical symptoms and signs, and the Kaplan-Meier method was used to calculate the median and 95% CI of the duration of clinical symptoms and signs. The Log-rank test was used to compared data between groups.Safety was assessed through the incidence of adverse reactions and laboratory tests, and the Chi-square test was used to analyze the difference between groups. Results:There were 123 children in the experimental group and 122 children in the control group. The median durations of all the 5 clinical symptoms and signs [including shortness of breath, wheezing, dyspnea (visible retractions), decreased transcutaneous oxygen saturation, and abnormal mental state] in the experimental group after treatment were slightly shortened than those in the control group [2.7 d(95% CI: 1.9-3.0 d)] vs.[2.9 d(95% CI: 2.6-3.6 d), P=0.027].The improvement in dyspnea (retractions) was especially pronounced in the experimental group, with a relief rate of 50.0% (0, 100%) on the first day of administration[compared with 0 (0, 50.0%) in the control group ( Z=2.002, P=0.025)].The median duration of dyspnea in the experimental group was nearly 1 day shorter than that in the control group [1.0 d(95% CI: 0.7-1.7 d) vs.1.8 d(95% CI: 1.0-2.5 d), P=0.046].There were no significant difference in hospital stay [6.0(5.0, 8.0) d vs.6.5(5.0, 8.0) d, Z=0.675, P=0.500], oxygen therapy duration [32.0(14.0, 96.3) h vs.39.0 (24.0, 83.2) h, Z=0.094, P=0.925], the recovery rate from clinical symptoms during treatment [(105/106, 99.1%) vs.(96/101, 95.0%)], and recurrence rate [(0/106, 0) vs.(2/101, 2.0%)] between the 2 groups (all P>0.05).However, the above-mentioned four indicators in the experimental group showed a trend of clinical benefits. The quantitative virus detection results showed that the RSV viral load in both groups decreased after treatment compared to before treatment. After 2 days of treatment, the decline rate of RSV viral load from the baseline was 0.90 lg copies/(mL·d) in the experimental group and 0.25 lg copies/(mL·d)in the control group, with a statistically significant difference ( P<0.05).Furthermore, there was no statistically significant difference in the incidence of adverse reactions between the 2 groups ( P>0.05).Importantly, no drug-related serious adverse reactions occurred in both groups. Conclusions:The nebulized inhalation therapy of IFN α1b demonstrates efficacy and safety in treating pediatric RSV associated lower respiratory tract infections. It particularly offers outstanding clinical therapeutic value for severe children.

Objective To compare the accuracy differences of three-dimensional reconstruction of zygomatic-maxillary complex using cone beam computed tomography(CBCT)and multi-slice spiral computed tomography(MSCT)in the parameters related to zygomatic implantation. Methods Five adult skull specimens(10 zygomatic-maxillary complexes in total)were selected.According to the clinical application characteristics of zygomatic implantation, the parameter location for each skull specimen was conducted, and then CBCT and MSCT were used for scanning and three-dimensional reconstruction.Four parameters were measured on both skull specimens and reconstructed models:the thickness of the maxillary zygomatic process at the intersection of the zygomatic implant path and zygomaticomaxil-lary suture(LB-B1),the thickness of the zygomatic bone at the intersection of the zygomatic implant path and the midpoint of the zygomatic bone surface(LC-C1),the width of the alveolar ridge between the second premolar and the first molar(LAR),and the buccolingual width of the first molar(LM).The specimen group was measured by electronic vernier caliper, and the CBCT group and MSCT group were measured based on CBCT and MSCT.The absolute and relative errors of these four parameters were calculated, and the accuracy of the CBCT and MSCT three-dimensional reconstruction models were analyzed. Results All parameters demonstrated excellent measurement reliability and repeatability, with intra-class correlation coefficient(ICC)≥0.90.The measurement results of each parameter in the CBCT group and the MSCT group showed statistically significant differences compared with those in the specimen group(P<0.05).The measurement result of LM in the CBCT group was smaller than that in the MSCT group, and the difference was statistically significant(P<0.05).However,the measurement results of LB-B1,LC-C1 and LAR in the CBCT group showed no statistically significant difference compared with those in the MSCT group(P>0.05).The mean absolute error and relative error of the LM measurement results to the specimen group in the CBCT group were both smaller than those in the MSCT group, and the differences were statistically significant(P<0.05).There was no statistically significant difference in the mean absolute error or relative error of the LB-B1,LC-C1 and LAR measurement results to the specimen group between the CBCT group and the MSCT group(P>0.05).Conclusion CBCT and MSCT demonstrat comparable accuracy in three-dimensional reconstruction of the zygomatic-maxillary complex. However,CBCT exhibits superior accuracy for fine structures such as dental tissues, which is recommended as the primary choice for imaging data acquisition in zygomatic implantation.
