1.Non small cell lung cancer with SMARCA4 deficiency harboring rare EGFR mutations exhibited significant tumor response when treated with afatinib: a case report.

SMARCA4-deficient non small cell lung cancer (SMARCA4-dNSCLC) has recently garnered increasing attention due to its high malignancy and poor prognosis. The literature suggests that in non small cell lung cancer (NSCLC), the loss of SMARCA4 frequently co-occurs with mutations in KRAS, KEAP1, and STK11 rather than in EGFR, ALK, and ROS1. Herein, we present the first documented case of SMARCA4-dNSCLC accompanied with rare mutations of EGFR exon 20 S768I and exon 18 G719X. The patient achieved partial response with afatinib for 17 months. Our case highlights the importance of EGFR mutations in the precision targeted treatment of SMARCA4-dNSCLC.

2.Effects of freeze-drying and dry heat virus inactivation processes on the biological activities of human plasma derived α2-macroglobulin

Objective To assess the impact of freeze-drying and dry heat virus inactivation processes on the activity ofα2-macroglobulin(A2M)derived from human plasma Cohn fraction Ⅳ.Methods A2M derived from human plasma Cohn fraction Ⅳ was prepared and subjected to programmed freeze-drying with dry heat virus inactivation.The lyophilized products were evaluated for their appearance,water content,and validation of the viral inactivation process.The bioactivity of the products before and after lyophilization as well as before and after dry heat inactivation was determined via trypsin inhibition,and the comparisons were studied.Results The appearance of the lyophilized product was fluffy,and the water content was(5.83±0.45)%.The specific activities of the samples before and after lyophilization were(10.199±0.137)and(10.033±0.201)μg/mg,respectively,with no statistically significantdifference between the two groups(P>0.05).The viral inactivation of the samples was carried out by using dry heat inactivation conditions at 100 ℃ for 30 min.After inactivation,the reduction was ≥5.125 LgTCID50/0.1 mL in Pseudorabies virus(PRV)titers,≥4.500 LgTCID50/0.1 mL in Sindbis virus(SinV)titers,≥6.375 LgTCID50/0.1 mL in encephalomyocarditis virus(EMCV)titers,and≥4.500 LgTCID50/0.1 mL in porcine parvovirus(PPV)titers.The specific activities of the samples before and after dry heat were(9.921±0.292)and(10.091±0.278)μ g/mg,respectively,with no statistically significant difference between the two groups.Conclusion A2M derived from human plasma Cohn fraction Ⅳ,when subjected to freeze-drying followed by dry heat inactivation at 100 ℃ for 30 minutes,can effectively inactivate viruses without altering the biological activity of the product.
3.The expression of circular RNA hsa_circ_0046701 in glioma tissues and its effect on the proliferation,migration and invasion of glioma U251 cells

Objective:To investigate the expression of circular RNA hsa_circ_0046701 in glioma tissues and its effect on the proliferation,migration and invasion of glioma U251 cells.Methods:Tumor tissue specimens and clinical data of fifty-two glioma patients treated surgically in Putuo People's Hospital Affiliated to Tongji University between June 2022 and March 2023 were collected,and another 30 normal brain tissue specimens were collected as controls.The expression level of hsa_circ_0046701 in glioma tissues was detected by qPCR to analyze the relationship between the level and clinical characteristics of the patient.Kaplan-Meier method was used to analyze the relationship between hsa_circ_0046701 level and survival prognosis.Using RNA interference technology,circ_0046701 overexpression and empty vector,siRNA-circ_0046701 and negative control(si-NC)plasmid were transfected into glioma U251 cells,respectively.The experiment cells were divided into the circ 0046701 OE group,Vector group,si-circ_0046701 group and si-NC group.CCK-8 method and Transwell assay were applied to detect the proliferation,migration and invasion abilities of cells in each group.Western blotting was applied to detect the expressions of vimentin,Snail,E-cadherin and cyclin D1 proteins in cells of each group.Results:The expression level of hsa_circ_0046701 in glioma tissues was significantly higher than that in normal brain tissues(P<0.01).The percentage of patients with WHO glioma grading(grades Ⅲ~Ⅳ)in the hsa_circ_0046701 high-expression group was significantly higher than that of the hsa_circ_0046701 low expression group(P<0.01).The OS of patients in the high-expression group was significantly shorter than that of the low-expression group.Compared with those in the si-NC group,the proliferation ability of U251 cells in the si-circ_0046701 group was significantly reduced(P<0.05 or P<0.01),and the numbers of migrated and invaded cells decreased significantly(both P<0.01).The expressions of vimentin,Snail,and cyclin D1 proteins in the cells was significantly reduced(all P<0.01),while the expression of E-cadherin protein increased significantly(P<0.01).Compared with those in the Vector group,the proliferation ability of U251cells in the circ_0046701 OE group increased significantly(P<0.01);the numbers of migrated and invaded cells increased significantly(both P<0.01);the expressions of vimentin,Snail,and cyclin D1 proteins in the cells increaseds significantly(all P<0.01),and the expression of E-cadherin protein decreased significantly(P<0.01).Conclusion:Circular RNA hsa_circ_0046701 is highly expressed in glioma tissues,and is correlated with poor patient prognosis.Knockdown of hsa_circ_0046701 expression could inhibit the proliferation,migration and invasion of glioma U251cells.
4.Study on long noncoding RNA LBX2-AS1 regulates glioma cell proliferation, migration and apoptosis through epidermal growth factor receptor signaling pathway

Objective:To investigate the mechanism of long noncoding RNA (lncRNA) LBX2-AS1 regulating glioma cell proliferation, migration and apoptosis through epidermal growth factor receptor (EGFR) signaling pathway.Methods:From April 2018 to August 2021, glioma U251 cells (U251 cells for short) were divided into control group and observation group, with 4 strains in each group. The control group was routinely cultured, and the observation group was transfected with specific small interfering RNA (siRNA) targeting LBX2-AS1. The proliferation ability of U251 cells was detected by methyl thiazol tetrazolium method, the metastasis rate of U251 cells was detected by scratch test, the apoptosis rate of U251 cells was detected by flow cytometry, and the expression of total protein and vascular endothelial growth factor (VEGF), phosphorylated inositol 3 kinase (p-PI3K), phosphorylated protein kinase B (p-Akt), phosphorylated Ras (p-Ras) and phosphorylated Raf (p-Raf) protein were detected by Western blot.Results:The proliferation ability and metastasis rate of U251 cells in observation group were significantly lower than those in control group: (27.15 ± 1.38)% vs. (63.54 ± 2.47)% and (37.09 ± 3.74)% vs. (82.17 ± 9.24)%, the apoptosis rate of U251 cells was significantly higher than that in control group: (69.17 ± 5.83)% vs. (17.58 ± 1.22)%, and there were statistical differences ( P<0.01). The expression of total protein and VEGF, p-PI3K, p-Akt, p-Ras, p-Raf protein of U251 cells in observation group were significantly lower than those in control group (1.52 ± 0.23 vs. 2.39 ± 0.31, 0.73 ± 0.08 vs. 1.68 ± 0.45, 0.57 ± 0.11 vs. 1.89 ± 0.31, 0.68 ± 0.06 vs. 1.74 ± 0.51, 0.84 ± 0.12 vs. 1.99 ± 0.63 and 0.71 ± 0.08 vs. 1.52 ± 0.37), and there were statistical differences ( P<0.01). Conclusions:The lncRNA LBX2-AS1 is highly expressed in glioma cells. Silencing the expression of lncRNA LBX2-AS1 inhibits the proliferation and metastasis of glioma cells through EGFR pathway.
5.Effect of telmisartan on expression of metadherin in the kidney of mice with unilateral ureter obstruction.

OBJECTIVE:
To explore the effect of telmisartan on the expression of metadherin in the kidney of mice with unilateral ureter obstruction.
METHODS:
Eighteen male C57 mice were randomized into sham-operated group, model group and telmisartan treatment group. In the latter two groups, renal interstitial fibrosis as the result of unilateral ureter obstruction (UUO) was induced by unilateral ureteral ligation with or without telmisartan intervention. Renal pathological changes of the mice were assessed using Masson staining, and immunohistochemistry and Western blotting were used to detect the expression of extracellular matrix proteins and metadherin in the kidney of the mice. In the experiment, cultured mouse renal tubular epithelial cells (mTECs) were stimulated with transforming growth factor-β1 (TGF-β1) and transfected with a siRNA targeting metadherin, and the changes in the expressions of extracellular matrix proteins and metadherin were detected using Western blotting.
RESULTS:
The expressions of extracellular matrix proteins and metadherin increased significantly in the kidney of mice with UUO ( < 0.05). Intervention with telmisartan significantly lowered the expressions of extracellular matrix proteins and metadherin and alleviated the pathology of renal fibrosis in mice with UUO ( < 0.05). In cultured mTECs, siRNA-mediated knockdown of metadherin obviously reversed TGF-β1-induced increase in the expressions of extracellular matrix proteins and metadherin.
CONCLUSIONS
Telmisartan can suppress the production of extracellular matrix proteins and the expression of metadhein to attenuate UUO-induced renal fibrosis in mice.

6.Epidemiological analysis of acute carbon monoxide poisoning in children induced by coal burning for home-heating in Tianjin area from 2012 to 2017

Objective To analyze the epidemiological features of acute carbon monoxide poisoning (ACOP) in children induced by coal burning for home-heating, so as to provide evidence for ACOP prevention and control.Methods A retrospective analysis was made on the medical data of gender , age, dwelling place, poisoning time, intoxication of family members , implementation time of hyperbaric oxygen ( HBO) therapy in child patients with ACOP admitted to the Department of HBO of Tianjin Children's Hospital from June 1, 2012 to May 31, 2017 due to coal burning for home-heating in Tianjin area from 2012 to 2017.Results In the past 5 years, a total of 381 patients with ACOP were admitted for treatment , including 322 patients intoxicated (84.51%)owing to burning coal for home-heating.Of the 322 patients, 181 were boys (56.21%) and 141 were girls (43.79%).Age ranged from 22 days to 15 years old, with an average age of 4.2 ±3.3 years.And of the 322 patients, 73 were younger than 1 year old (22.67%), 85 were in the age of 1-3 years (26.40%), 91 were in the age of 3-6 years (28.26%), and 73 were over 6 years old (22.67%).As to the area of distribution of ACOP, 37 cases were in 6 main municipal districts of Tianjin , accounting for (11.49%), 166 cases were in the suburban , accounting for 51.55%and 119 cases were in exurban of the city , accounting for 36.96%, with an annual average incidence of 64.4 cases.Most cases ( 281, 87.27%) occurred in the centralized heating period, i.e.from Nov 15 to Mar 15 nest year, and relatively few cases (41, 12.73%) occurred beyond the heating period .Beyond the heating period , the percentage of ACOP patients younger than 1 year was 29.27%(12/41), and statistical significance could be noted when comparisons were made (P=0.004).There were 280 ACOP patients (86.96%) who had ACOP at night (86.96%), and there were 272 patients (84.47%) whose family members also had ACOP , with a total 489 family members having ACOP . One hundred and forty-one patients (43.79%) received HBO therapy , 6 hours after the onset of symptoms of carbon monoxide poisoning , and on the average , they had 6.1 sessions of HBO therapy .Conclusion In the highly prevalent city areas , during the winter season and among special population of the city , propaganda and early warning concerning knowledge of carbon monoxide poisoning should be made , so as reduce the rate of ACOP.

Since the concept of molecular imaging was put forward in 1999,optical molecular imaging techniques have been widely applied in the field of biomedical and clinical research.Its unique application value is especially shown in hepatobiliary surgery,such as in liver tumor imaging,anatomical liver resection,liver transplant angiography,cholangiography,and bile or pancreatic leakage prevention.Optical molecular imaging technique "lights up" targeted areas in surgical operations and provides convenience in carrying out precision operation.This paper reviewed the advantages of optical molecular imaging technology in clinical research and discussed its limitations in translational surgery,and put forward possible directions in improvement for the future.
9.Effect and safety of ultrasound-guided injection of poly-alcohol on patients with scar pregnancy

Objective To investigate the efficacy and safety of ultrasound-guided polyglycol in patients with scar pregnancy. Methods Thirty-six patients with scarring treated in Third People's Hospital of Hangzhou from November 2014 to November 2016 were selected as subjects. All patients underwent ultrasound-guided polyglycol administration. The results were analyzed and analyzed. The results were analyzed. Gonadotropin (β-HCG) returned to normal time and the occurrence of adverse symptoms. Results The average recovery time, treatment cost, average recovery time and adverse reaction rate of blood β-HCG in the study group were lower than those in the control group, the difference was statistically significant (P<0.05). Conclusion The effect of ultrasound on the injection of polyphenols in patients with scar pregnancy is safe, feasible and easy to be accepted by patients and their families. It has clinical significance.
10.Comprehensive rehabilitation is superior to surgical intervention for many patients with recurrent lumbar disc herniation

Objective To compare the effectiveness of comprehensive rehabilitation therapy with that of surgical intervention for patients with recurrent lumbar disc herniation (LDH).Methods A total of 124 LDH patients were recruited antd randomly divided into an experimental group and a control group,each of 62.The experimental group was given comprehensive rehabilitation consisting of medicine,physical therapy,and spinal stabilization exercises,while the control group received lumbar spine surgery.The patients' back and leg pain scores and their Oswestry Disability Index (ODI) scores were compared at admission and discharge,as well as 3 and 12 months after discharge.Anxiety and depression were also assessed at admission and 3 months after discharge.Results At discharge and 3 and 12 months later the average back and leg pain scores and ODI ratings were significantly lower than that at admisshin for both groups.There were no significant inter-group differences.In terms of anxiety and depression,however,these had decreased signifieantly in both groups,but the experimental group's average rating was significantly better at diseharge.Conclusions Comprehensive rehabilitation is superior to surgical intervention in relieving the depression of patients with recurrent LDH.Comprehensive rehabilitation should be considered as the first ehoiee for patients with rccurrent LDH,and only supplemented by surgical intervention if it is not effective.
