1.Correlation between serum thymosin α1 and cardiac function in patients with acute anterior wall ST-segment elevation myocardial infarction

Objective To explore the correlation between serum thymosin α1 level and left ventricular ejection fraction(LVEF)in patients with acute anterior wall ST-segment elevation myocardial infarction(STEMI)after receiving percutaneous coronary intervention(PCI).Methods A total of 74 patients with acute anterior wall STEMI(acute anterior wall STEMI group)and 38 patients having no myocardial infarction(control group),who were admitted to the Shanghai Sixth People's Hospital of China from December 2019 to February 2022,were enrolled in this study.According to the LVEF value after the recanalization of anterior descending coronary artery with PCI during hospitalization period,the patients of acute anterior wall STEMI group were divided into LVEF<50%subgroup(n=33)and LVEF≥50%subgroup(n=41).Serum thymosin α1 level was determined by enzyme linked immunosorbent assay(ELISA),the results were compared between the groups.Logistic regression analysis was used to analyze the correlation between thymosin α1 level and LVEF.The receiver operating characteristic(ROC)curve of serum thymosin α1 level for predicting cardiac function in patients with acute anterior wall STEMI after receiving PCI was drawn.Results The serum thymosin α1 level in LVEF≥50%subgroup was significantly higher than that in the LVEF<50%subgroup(P=0.032).During the post-PCI hospitalization period,the serum thymosin α1 level was positively correlated with LVEF.Logistic regression analysis revealed that serum thymosin α1 level was an independent predictor for LVEF<50%in patients with acute anterior wall STEMI after receiving PCI.The area under ROC of serum thymosin α1 level for predicting LVEF≥50%in patients with acute anterior wall STEMI during hospitalization was 0.644(P=0.034).The area under ROC of serum thymosin α1 level combined with peak troponin I level and with peak NT-proBNP level for predicting LVEF<50%in patients with acute anterior wall STEMI during hospitalization was 0.780(P<0.01)and 0.702(P=0.003)respectively.When taking the median serum thymosin α1 level as the cut-off value,the proportion of LVEF≥50%patients was higher among the patients having the post-PCI serum thymosin α1 level>2,890 ng/L.Conclusion In patients with acute anterior wall STEMI,the serum thymosin α1 level is closely related to the LVEF value during the post-PCI hospitalization period,it is an independent predictor for cardiac function improvement after PCI.It is expected that the serum thymosin α1 level may become a new indicator for predicting the improvement of cardiac function in patients with STEMI after recanalization of anterior descending coronary artery with PCI.
2.The influencing factors of microcirculation dysfunction in patients with anterior wall acute myocardial infarction and the establishment of prediction model

Objective To explore the influencing factors of microcirculation dysfunction in patients with anterior wall acute myocardial infarction(AMI)and to establish a relevant prediction model.Methods A total of 130 patients with anterior wall AMI,whose microcirculation function was assessed by caIMR after receiving percutaneous coronary intervention(PCI)at Shanghai Sixth People's Hospital of China from January 2017 to September 2020,were enrolled in this study.The patients were divided into abnormal microcirculation resistance group(n=52)and normal microcirculation resistance group(n=78).The clinical data were compared between the two groups.The regression analysis was used to analyze the influencing factors of microcirculation dysfunction.Results In the abnormal microcirculation resistance group the contrast agent consumption,the onset-to-operation time,the Gensini total score and the LAD Gensini score were(121.92±31.37)mL,(10.51±5.12)min,(97.91±31.77)points and(69.36±13.15)points respectively,which were significantly higher than(109.03±28.2)mL,(4.94±2.94)min,(81.05±35.22)points and(54.45±23.48)points respectively in the normal microcirculation resistance group,the differences in the above indexes between the two groups were statistically significant(all P<0.05).A prediction model covering interventional strategies was established,and its accuracy was higher than that of a conventional model,its AUC compared with the conventional model was 0.91 to 0.87,indicating that this model could well predict the risk of microcirculation dysfunction in patients with AMI after receiving PCI.Conclusion This prediction model can promptly identify high-risk microcirculation dysfunction patients with anterior wall AMI after receiving PCI.
3.Development and application of a rapid scheme for detection of respiratory virus nucleic acid.

This study aimed to develop a portable, accurate and easy-to-operate scheme for rapid detection of respiratory virus nucleic acid. Quantitative real-time polymerase chain reaction (qRT-PCR) was used to verify the effect of extraction-free respiratory virus treatment reagent (RTU) on viral nucleic acid treatment and the effect of ultra-fast fluorescence quantitative PCR instrument (FQ-8A) on nucleic acid amplification, respectively. RTU and FQ-8A were combined to develop a rapid detection scheme for respiratory virus nucleic acid, and the positive detection rate was judged by Ct value using a fluorescence quantitative PCR instrument, and the accuracy of the scheme in clinical samples detection was investigated. The results showed that RTU had comparable sensitivity to the automatic nucleic acid extraction instrument, its extraction efficiency was comparable to the other 3 extraction methods when extracting samples of different virus types, but the extraction time of RTU was less than 5 min. FQ-8A had good consistency in detection respiratory syncytial virus (RSV) and adenovirus (ADV) compared with the control instrument ABI-7500, with kappa coefficients of 0.938 (P < 0.001) and 0.887 (P < 0.001), respectively, but the amplification time was only about 0.5 h. The RTU and FQ-8A combined rapid detection scheme had a highly consistent detection rate with the conventional detection scheme, with a sensitivity of 91.70% and specificity of 100%, and a kappa coefficient was 0.944 (P < 0.001). In conclusion, by combining RTU with FQ-8A, a rapid respiratory virus nucleic acid detection scheme was developed, the whole process could be completed in 35 min. The scheme is accurate and easy-to-operate, and can provide important support for the rapid diagnosis and treatment of respiratory virus.

4.Viscoelastic properties of embryonic neuroepithelial cells in neural tube defects mice

Objective To explore the role of mechanical properties of embryonic neuroepithelial cells in the process of neural tube closure.Methods Neural tube defects (NTDs) mouse model was established by intragastric administration with all-trans retinoic acid at embryo 7.25 day.All the pregnant mice were sacrificed at embryo 9.5 day and 11.5 day,respectively,and the primary neuroepithelial cells were isolated from neural tube tissue of normal and NTDs mice,respectively.The mechanical characteristics of neuroepithelial cells were analyzed using micropipette aspiration technique combined with a standard solid viscoelastic mechanical model.Results The mechanical characteristics of mouse embryonic neuroepithelial cells showed typical viscoelastic solid characteristics.Compared with the control group,the three viscoelastic parameters,i.e.equilibrium modulus,transient modulus and apparent viscosity coefficient,of the neuroepithelial cells in the NTDs group were significantly increased,and the differences were statistically significant (all P<0.05).However,there was no significant difference in the viscoelastic parameters of the same group between embryo 9.5 d and 11.5 d (all P>0.05).Conclusion The decrease in the deformability of embryonic neuroepithelial cells may be one of the factors responsible for neural tube closure disorders.
5.Effect of Coronary Artery Tortuosity on Coronary Hemodynamics Based on Case Study

Objective To investigate the hemodynamic changes in a tortuous coronary to elucidate the effects of tortuosity on coronary perfusion and wall shear stress (WSS). Methods A single tortuous and non-tortuous patient-specific left anterior descending (LAD) coronary artery cases were selected. Two LAD models with and without coronary tortuosity were reconstructed in Mimics software and then transferred to the ANSYS Fluent software for performing computational fluid dynamics (CFD) simulation. The hemodynamic characteristics of both the LAD models were compared. Results The vessel WSS of the tortuous coronary artery clearly decreased in the bend section where the maximum curvature was larger than 1 mm-1.Such a scenario could led to an inadequate blood supply in the downstream vessels. A low WSS (0-26 Pa) acted on the outer wall of the bend, whereas the inner wall of the bend had a high WSS (>100 Pa). The mean WSS of the non-tortuous and tortuous models was 10.79 Pa and 36.12 Pa, respectively. The overall WSS of the tortuous model was larger compared with that of the non-tortuous model. Conclusions Coronary tortuosity increased the overall WSS, which could delay the progress of coronary atherosclerosis.
6.Discussion on Consistency of TCM Yin-Yang Balance and Microecological Balance

Yin-yang theory is the core theory in TCM. In order to extend the guidance effects of theory of the balance of yin and yang in the clinic, and help TCM theory and curative effect be further recognized by the world, this article analyzed the consistency of different systems by comparing TCM balance theory and microecology, with a purpose to explore the closed link between TCM balance theory and microecology and microscopic nature of TCM balance theory.

Objective To evaluate the necessity of construction of hospital information system integration platform for county Hospital as the urban and rural medical hub in the context of the development of modern medical information.Methods By referring to the development of the domestic and international integration platform,and comprehensively considered the development status of the county hospital,and analyzed based on the perspective of sustainable development.Results The hospital information integration platform was formed to meet the needs of the county hospital,and it platform was used to open up the internal and external medical information resources,realize the integration of business resources and process optimization..Conclusion The county hospital building information system integration platform is the inevitable trend of the current construction of information technology,and its comple-ted construction can enhance province -city-county tertiary care facility data communication,reduce the coupling within the hospital business systems,and make the county hospital more flexible in the future construction and devel-opment.
8.Growth Difference between CNE-2 and S-18 Cell Lines after Subcutaneous Xenograft

Objective To study the growth difference and possible mechanism between nasopharyngeal carcinoma (NPC) cell line CNE-2 and its subclone S-18.Methods CNE-2 and S-18 cells were cultured in vitro.6 x 105 cells/mouse were xenografted subcutaneously in the back of nude mice.The volumes of rumors were measured on the 3 rd,7 th,10 th,14 th day after grafting.Mice were sacrificed on the 14 th day and tumors were isolated and weighed.RNA from tumor tissues were extracted and transcriptional levels of HSP27 and NF-K B were detected.Results (1) S-18,instead of CNE-2,grew to form tumor mass 7 days after xenografting subcutaneously;both cell lines formed tumor mass 10 days after xenografting,however,the volumes of S-18 tumors [(223.13 ± 21.32) mm3,10 th day;(420.25 ± 24.52) mm3,14 th day] were significant bigger than CNE-2tumors [(113.70±11.70) mm3,10thday;(279.86±25.78) mm3,14thday];The weights of S-18 umors were significantly higher than CNE-2 tumors on the 14 th day after xenografting;(2) The transcriptional levels of HSP27 and NF-KB in S-18 tumor were significantly higher than in CNE-2 tumor.Conclusion Xenografted S-18 NPC grows faster than Xenografted CNE-2 NPC.HSP27 and NF-κ B are probably involved in the regulation of growth in NPC.
9.Architecture design of county hospital information system integration platform

Based on the current county hospital information construction in the problem of integration,pro-posed a scheme based on a unified architecture application integration platform.The original business system interface standardization of the construction of hospital information systems analysis and integration platform architecture and integration framework,the framework can be achieved through integrated hospital between heterogeneous systems.A unified standardized interfaces can achieve a good cross -system data integration.The construction of county hospitals information system integration platform,data sharing and interaction is conducive to solve the problem of data islands facing the county hospital,hospital to enhance business development capabilities and scale.
10.Association of gankyrin protein expression in human colorectal cancer with postoperative prognosis.

OBJECTIVETo investigate the association of gankyrin protein expression in colorectal cancer (CRC) with its prognosis.
METHODSClinical data and resection samples of 100 colorectal cancer patients identified by pathology undergoing resection in our department from June 2008 to June 2009 were collected. The gankyrin expression in CRC tissues and matched adjacent noncancerous tissues collected during the operation of 100 CRC cases was detected by immunohistochemical staining and Western blotting. The associations of gankyrin expression level with overall survival, clinicopathologic features were analyzed by Chi square test, Cox regression analysis, Kaplan-Meier analysis and log rank test.
RESULTSImmunohistochemical staining showed that the positive brown granules were mainly distributed in the cytoplasm, and nuclear immunostaining was observed in tissue samples of 29 cases, of whom 16 cases had distal metastasis [55.2% (16/29)]. The positive rate of gankyrin and the relative gray value of Western blotting in CRC tissues were 67% (67/100) and 0.69±0.23, respectively, which were significantly higher than those of 2 cm adjacent noncancerous tissues [6% (6/100) and 0.31±0.16] and 10 cm adjacent noncancerous tissues [1%(1/100) and 0.16±0.11] (all P<0.001). Patients with positive expression of gankyrin had worse survival than those with negative ones (41.8% vs. 72.7%, P=0.008). The gankyrin expression was associated to lymph node metastasis (P=0.005), tumor stage (P=0.001) and distal metastasis (P=0.002). Cox regression analysis showed that distal metastasis (P=0.004) and high expression of gankyrin (P=0.038) were independent risk factors for poor prognosis of patients with CRC.
CONCLUSIONUp-regulated expression of gankyrin is related to invasion and metastasis of human CRC, and gankyrin may be valuable in predicting prognosis.
