Objective:
To understand the current situation and regulatory effectiveness of daylighting and artificial lighting in primary and secondary school classrooms in Shanghai, so as to provide a basis for enhancing the visual environment of school classrooms.
Methods:
From April 2021 to December 2023, the daylighting and artificial lighting conditions of classrooms in 1 735 regular primary and secondary schools currently in operation in Shanghai were monitored, and the qualified rate of each indicator was calculated. The Chisquare test or Fisher exact probability method were used to compare the differences in qualification rates across different educational stages, regions, school type, both before and after the implementation of regulatory measures. The regulatory measures included convene interview, propaganda and education, supervision order, supervisory opinion paper, rectification requests and offenses and punishment.
Results:
The qualified rate of daylighting and artificial lighting in primary and secondary school classrooms was 30.1%, with a qualified daylighting rate of 85.6% and a qualified artificial lighting rate of 32.9%. There was no statistically significant differences in the qualified rate of daylighting and artificial lighting in primary and secondary schools (32.4%,28.1%;χ2=3.76,P>0.05). However, statistically significant differences were found in the qualified rate of daylighting and artificial lighting in urban and rural school classrooms (32.6%,26.7%), as well as in public and private schools (31.4%, 20.6%) (χ2=6.99,9.92,P<0.05). Following the implementation of regulatory measures, the qualified rate of classroom daylighting and artificial lighting improved from 30.1% to 83.2%, while the respective qualified rates of daylighting and artificial lighting increased from 85.6% to 91.1% and 32.9% to 90.5%. Compared to the preimplementation period, the qualified rate of classroom daylighting and artificial lighting, as well as the respective rates of daylighting and artificial lighting, all showed statistically significant differences after the implementation of regulatory measures (χ2=995.29,25.34,1 219.87,P<0.01).
Conclusions
Effective regulatory measures can promote enhanced classroom daylighting and artificial lighting. Attention should be paid to improving classroom artificial lighting, in order to provide students with enhanced visual environment.

Recent Advances in Ophthalmology 2024;44(8):613-618
Objective To explore the effect of targeted inhibition of miR-628-5p by long non-coding RNA(lncRNA)KCNQ1 overlapping transcript 1(KCNQ1OT1)on high glucose(HG)-induced retinal pigment epithelial cell injury.Meth-ods The real-time fluorescence quantitative polymerase chain reaction(RT-qPCR)was performed to detect KCNQ1OT1 expression in serum and aqueous humor of patients with diabetic retinopathy(DR),type 2 diabetes mellitus(T2DM)and uveitis.Human retinal pigment epithelial cells(ARPE-19)were treated with different concentrations of glucose, and their KCNQ1OT1 expression was detected to determine the optimal glucose concentration.ARPE-19 cells were divided into the control(CON)group,HG group,HG+si-NC group,HG+si-KCNQ1OT1 group,HG+miR-NC group,HG+miR-628-5p group,HG+si-KCNQ1OT1+anti-miR-NC group, and HG+si-KCNQ1OT1+anti-miR-628-5p group. The expression levels of KCNQ1OT1 and miR-628-5p in cells were measured by RT-qPCR.The cell apoptosis was detected by flow cytometry. The malondialdehyde(MDA)level and superoxide dismutase(SOD)activity were detected by enzyme-linked immunosorbent assay. The expression level of cleaved Caspase-3 protein was measured by Western blot. The targeting relationship between KCNQ1OT1 and miR-628-5p was detected by dual-luciferase reporter assay. Results The expression levels of KCNQ1OT1 in serum and aqueous humor of DR patients were higher than those of T2DM patients and uveitis patients, and those of uve-itis patients were the lowest(all P<0.05).Compared with the 0 mmol·L-1 glucose group, the expression levels of KC-NQ1OT1 in ARPE-19 cells in the 5 mmol·L-1 glucose group,15 mmol·L-1 glucose group, and 30 mmol·L-1 glucose group significantly increased, and those in the 30 mmol·L-1 glucose group were the highest(all P<0.05);thus,30 mmol·L-1 glucose was selected for subsequent experiments.Compared with the CON group, the KCNQ1OT1 expres-sion,apoptosis rate, cleaved Caspase-3 protein expression, and MDA level in the HG group significantly increased, while the SOD activity and miR-628-5p significantly decreased(all P<0.05).Compared with the HG+si-NC group, the KC-NQ1OT1 expression, apoptosis rate, cleaved Caspase-3 protein expression, and MDA level in the HG+si-KCNQ1OTl group significantly decreased, while the SOD activity significantly increased(all P<0.05).Compared with the HG+miR-NC group,miR-628-5p and SOD activity in the HG+miR-628-5p group significantly increased, while the apoptosis rate, cleaved Caspase-3 protein expression, and MDA level significantly decreased(all P<0.05).Compared with the HG+si-KCNQ1OT1+anti-miR-NC group,miR-628-5p and SOD activity in the HG+si-KCNQ1OT1+anti-miR-628-5p group significantly decreased, while the apoptosis rate, cleaved Caspase-3 protein expression, and MDA level significantly increased(all P<0.05).Conclusion lncRNA KCNQ1OT1 can reduce the HG-induced apoptosis and oxidative damage of ARPE-19 cells by targeted inhibition of miR-628-5p.

Aim To investigate the role of cholesterol in the regulation of lipid raft and the function of platelets. Methods Using in vitro incubation of methyl β-cyclodextrin(MβCD)and in vivo elevation of peripheral blood total cho-lesterol levels to remove and load platelet cholesterol, respectively.Cholera toxin B staining combined with flow cytometry was used to detect platelet lipid raft content, fluorescence antibody staining combined with flow cytometry was used to detect the expression levels of P-selectin and activated integrin α Ⅱ bβ3,annexin Ⅴ labeling combined with flow cytometry was used to detect the level of phospholipid efflux,in vitro experimental system and rat tail bleeding experiment were used to detect platelet aggregation ability. Results The content of lipid raft on B lymphocytes decreased with the removal of cholesterol, while in vitro incubation of MβCD to remove platelet cholesterol significantly increased its lipid raft level(P<0.05).Consistent with this,in vivo cholesterol loading increased the lipid raft content of B lymphocytes but decreased the lipid raft content of platelets(P<0.05).The increase in lipid raft after removing cholesterol was not conducive to platelet activation and aggregation function. In vivo cholesterol loading downregulated platelet lipid raft content(P<0.05),enhanced its ability to respond to low concentration stimulant for activation aggregation and coagulation, and this enhancing effect disappeared after cholesterol removal. Conclusion Platelet cholesterol is a key regulator of platelet lipid raft content and platelet function, which can negatively regulate lipid raft, promote platelet activation, and enhance their coagulation function.

Background and Aims:Cholangiocarcinoma,a rare malignant tumor,is difficult to diagnose and often detected at an advanced stage, limiting treatment options to palliative care.Conventional chemotherapy drugs have poor efficacy against cholangiocarcinoma, making the search for new treatments critical. This study was conducted to investigate the effects of Huai'erjunzhi on the malignant biological behavior of human cholangiocarcinoma cells and its relationship with the TGF-β/Smad pathway, aiming to provide a theoretical basis for the use of Huai'erjunzhi in cholangiocarcinoma treatment. Methods:Human normal liver cells(L-02)and human cholangiocarcinoma cells(HuCCT1)were incubated with different concentrations of Huai'erjunzhi for various durations.Cell proliferation was assessed, and the half-maximal inhibitory concentration(IC50)was calculated.HuCCT1 cells were divided into a negative control group(no intervention),a positive control group(15 mg/L cisplatin),and different Huai'erjunzhi intervention groups(1/5 IC50,2/5 IC50,and IC50 based on preliminary experimental results).Scratch and Transwell assays were used to measure cell migration and invasion, while Western blot was employed to detect the expression of proteins related to the TGF-β/Smad pathway in HuCCT1 cells. Results:Only high concentrations of Huai'erjunzhi(>312.5 mg/L)significantly inhibited the proliferation of L-02 cells.Huai'erjunzhi significantly inhibited the proliferation of HuCCT1 cells at concentrations above 5 mg/L in a concentration-dependent manner(all P<0.05),with IC50 values of 138.52 mg/L at 24 h,99.41 mg/L at 48 h,and 113.52 mg/L at 72 h.Compared with the negative control group, the positive control group and the three Huai'erjunzhi intervention groups(20,40,and 100 mg/L)exhibited reduced migration distance, decreased invasive cell numbers, lower expression of TGF-β1,Smad2,Smad3,Smad4,N-cadherin,Snail,and Slug, and increased expression of E-cadherin(all P<0.05).Compared with the positive control group, these changes in the Huai'erjunzhi groups were less pronounced but showed a clear concentration-dependent relationship(all P<0.05). Conclusion:Huai'erjunzhi can potentially inhibit the malignant biological behavior of HuCCT1 cells by inhibiting the TGF-β/Smad pathway.

ObjectiveTo observe the regulatory effect of Yuyetang on Ghrelin level in rats with type 2 diabetes mellitus (T2DM)-induced cognitive impairment (DCI) and explore the pathway in the prevention and treatment of DCI. MethodThe T2DM model was induced in rats by intraperitoneal injection of streptozotocin (STZ) combined with the high-fat and high-sugar diet (STZ). The model rats were divided into model group, metformin group (200 mg·kg-1), and low-,medium-, and high-dose Yuyetang groups(4.575,9.15, 18.3 g·kg-1)according to the blood glucose, with 10 rats in each group. A normal group was also set up. The rats were administered with corresponding drugs by gavage for 30 days, and the body weight and blood glucose of the rats in each group were observed and recorded. After drug intervention, the learning and memory abilities of rats were tested by the Morris water maze. After the test, the whole brains of rats were sampled for hematoxylin-eosin (HE) staining to observe the pathological changes in the hippocampal CA1 region, and the expression of Ghrelin in gastric tissues and hippocampal CA1 region was detected by immunohistochemistry. ResultCompared with the normal group , the model group showed increased blood glucose(P<0.01),reduced body weight(P<0.01),prolonged escape latency(P<0.05,P<0.01), shortened retention time and movement distance in the target area, decreased number of platform crossings(P<0.01), abnormal morphology and structure of cells with disordered arrangement and reduced number in the hippocampal CA1 region, and decreased expression of Ghrelin in the serum, hippocampal CA1 region, and gastric tissues(P<0.05, P<0.01). Compared with the model group, the medium- and high-dose Yuyetang groups showed increased body weight, while all Yuyetang groups showed reduced blood glucose(P<0.01), shortened escape latency (P<0.05), prolonged retention time and movement distance in the target area, increased platform crossings (P<0.05, P<0.01), improved morphology and structure of cells, increased number of normal cell in the hippocampal CA1 region, and elevated Ghrelin levels in the serum, gastric tissues, and hippocampal CA1 region(P<0.05, P<0.01). ConclusionYuyetang can effectively improve the cognitive ability of DCI rats, and its mechanism may be related to the regulation of Ghrelin levels in the serum, hippocampal CA1 region, and gastric tissues.
