1.The clinical research of IFNGR1 proximal promoter polymorphism in susceptibility and prognosis of breast cancer

This study investigated the association between a proximal promoter polymorphism of IFNGR1 (interferon-γ receptor α chain, IFNGR-α) and breast cancer susceptibility, as well as the prognostic value of its expression variation in breast cancer patients. A case-control study was conducted at the Sixth Medical Center of PLA General Hospital from June 2020 to June 2022. The study included 182 pathologically confirmed breast cancer patients as the breast cancer group, 177 non-tumor patients with benign breast lesions as the benign breast lesions group, and 229 healthy individuals as the normal control group. 2-3 ml EDTA anticoagulant whole blood samples were collected from all participants, and genomic DNA was extracted and stored for further analysis. Basic patient information was retrieved from the hospital′s electronic medical records by patients′ ID number. The proximal promoter sequence of IFNGR1 was obtained from NCBI, and sequencing primers were designed using Primer Premier 6.0. Sanger sequencing was employed to analyze the IFNGR1 promoter sequence in the three groups, and the results were compared with the Eukaryotic Promoter Database (EPD) sequence using Bioedit software. Statistical analysis was performed on single nucleotide polymorphisms (SNPs) in the IFNGR1 promoter. The TCGA database was utilized to assess the relationship between IFNGR1 expression levels and breast cancer patient survival. The findings revealed that the -56 TG genotype of the IFNGR1 promoter was significantly associated with increased breast cancer risk ( Z=2.73, P<0.05). Notably, IFNGR1 expression was lower in breast cancer group compared to normal control group ( P<0.05). Analysis of the TCGA database indicated that patients with high IFNGR1 expression had longer survival times than those with low expression ( HR=0.87, 95% CI:0.77-0.98, P<0.05). In summary, the IFNGR1 -56 TG genotype is associated with an increased risk of breast cancer, and there is a positive correlation between IFNGR1 expression levels and the survival of breast cancer patients.
2.Expression level of miR-196a in patients with HPV16 and HPV18 subtypes infections and bioinformatics analysis of its association with survival of cervical cancer

OBJECTIVE To explore the expression level of miR-196a in cervical cells infected with high-risk human papillomavirus(HPV)16 and 18.METHODS The Gene Expression Omnibus(GEO)was used to screen for dif-ferentially expressed miRNAs between HPV 16 or 18-positive cervical cancer cells and normal cervical cells.On-line biological software https://kmplot.com/analysis/was utilized to analyze the relationship between the most differentially expressed miRNA and the overall survival of cervical cancer patients.Cervical swab samples positive for HPV 16 or HPV 18,detected by real-time fluorescent quantitative polymerase chain reaction(qPCR)genoty-ping,were collected as the study subjects.Cervical swab samples from the same period of physical examination population that were negative for HPV 16 or HPV 18 by qPCR genotyping served as negative controls.The qRT-PCR method was employed to detect the level of miR-196a in cervical cells,with data processed via the 2-△△Ctmethod.RESULTS Differential analysis of the GSE86100 data revealed that miR-196a expression de-creased in HPV 16 or HPV 18-positive cervical cells(log2FC=-6.60,P<0.001),while miR-3188 expression significantly increased(log2FC=6.22,P<0.001).Using online analysis tools https://kmplot.com/analysis,it was found that cervical cancer patients with high miR-196a expression had a shorter overall survival compared to those with low m iR-196a expression(HR=1.87,95%CI:1.17-3.00,P=0.008).H owever,there was no cor-relation between miR-3188 and the overall survival of cervical cancer patients(HR=1.47,95%CI:0.92-2.37,P=0.110).The results of specific qRT-PCR testing showed that the expression levels of miR-196a in cervical cells positive for HPV 16 and HPV 18 were 0.93±0.09 and 0.51±0.07,respectively,which were lower than those in the normal control group(1.89±0.13)(P<0.05),consistent with the sequencing analysis results CONCLUSIONS Infection of cervical cells with HPV 16 or HPV 18 can lead to decreased expression of miR-196a,and the expres-sion level of miR-196a is negatively correlated with the overall survival of cervical cancer patients.
3.Construction and validation of clinical prediction model of somatic symptom disorder in epilepsy patients

Objective To explore the influencing factors of somatic symptom disorder(SSD)in patients with epilepsy,and to construct a nomogram prediction model.Methods Using structured interviews,according to the diagnostic criteria of Diagnostic and Statistical Manual of Mental Disorders(Fourth Edition),206 patients with epilepsy included in this study were divided into SSD group and non-SSD(n-SSD)group.The clinical data of the two groups were compared.The quality of life in epilepsy inventory(QOLIE-31),general anxiety disorder-7(GAD-7),neurological disorders depression inventory for epilepsy(NDDI-E)and Pittsburgh sleep quality index(PSQI)were used to evaluate the anxiety,depression and sleep of patients.LASSO regression was used for variable screening,and Logistic regression model was used to explore the risk factors of SSD in patients with epilepsy.Based on these factors,a nomogram was constructed and the area under the ROC curve(AUC)was calculated and verified internally.Calibration curve and decision curve analysis were used to evaluate the calibration and clinical utility of the nomogram,respectively.Results Compared with those in the n-SSD group,there were significant differences in age,age of onset,educational background,place of residence,number of physical diseases and negative life events in the SSD group(all P<0.05).Compared with those in the n-SSD group,GAD-7 score,NDDI-E score,PSQI score,total score of QOLIE-31 and the scores of seizure worry,drug influence,energy/fatigue,life satisfaction,social function and emotion in the SSD group were significantly lower(all P<0.05).Multivariate Logistic regression analysis showed that age(OR=1.076,95%CI:1.015-1.141),negative life events(OR=6.624,95%CI:2.130-20.606),seizure worry(OR=0.945,95%CI:0.895-0.999),energy/fatigue(OR=0.923,95%CI:0.872-0.977),GAD-7 score(OR=1.274,95%CI:1.037-1.565),NDDI-E score(OR=1.233,95%CI:1.038-1.442),PSQI score(OR=1.375,95%CI:1.097-1.723)were independent predictors of SSD.According to the variables in the results of multivariate Logistic regression analysis and their corresponding regression coefficients,the nomogram of SSD in patients with epilepsy was established.The AUC of the nomogram was 0.939(95%CI:0.904-0.975),the best cut-off value was 0.200,the sensitivity was 0.847,the specificity was 0.953,and the discrimination was good.The decision curve risk threshold showed that the model provides significant clinical net benefits.Conclusions Age,negative life events,seizure concerns,energy/fatigue,GAD-7 score,NDDI-E score and PSQI score are risk factors for epileptic SSD.The columniogram model constructed based on the above factors can effectively predict the risk of epileptic SSD.

7.A case report of glans penis hemangioma

Hemangioma of the penile head is rare. This paper reported a patient, 16 years old, who was admitted to hospital due to the discovery of multiple masses on the head of the penis for more than two years. Physical examination showed that three vascular mass-like masses were distributed along the coronal sulcus at the 3, 9, and 12 points of the penile head, and the larger one was about 10 mm×5 mm size, blue-purple, soft, and painless. Ultrasound examination suggested that the patient had a penile head hemangioma. Surgical resection was performed, and the postoperative pathological diagnosis was penile head hemangioma.The follow-up of 3 months showed that the wound healed well without recurrence, and the penile head appearance was not obviously deform.
8.Effects of salvia tetramethylpyrazine on hemorheology in vertebrobasilar transient ischemia attack

Objective To analyze the effect of salvia tetramethylpyrazine on hemorheology in patients with vertebrobasilar transient ischemia attack (VB-TIA). Methods 84 patients with VB-TIA were divided into observation group and control group, 42 cases in each group. They both received the treatment of Ginkgo-diyidamolum injection. The observation group were treated by salvia tetramethylpyrazine, and the control group were treated by compound Danshen injection. The therapeutic effect of the two groups and the changes of blood rheology were compared. Results The clinical remission rate of the observation group was significantly higher than that of the control group, and the time of onset and symptoms of the drug were shorter than those of the control group (P < 0.05). The blood flow velocity of vertebral basilar artery in the two groups were significantly higher than those in the observation group (P < 0.05). After treatment, the hemorheology of the two groups were significantly improved, the high shear viscosity and low shear viscosity, platelet aggregation rate, erythrocyte deformation index, erythrocyte aggregation index and blood haematocrit of observation group improved more obviously (P < 0.05). The incidence of adverse reactions in the two groups was not statistically significant. Conclusion Salvia tetramethylpyrazine could effectively shorten the drug onset time, improve the blood velocity of vertebrobasilar artery, recover blood rheology and stability on the condition of not increasing the adverse reactions, which has a positive effect on the clinical remission rate.

Objective: To investigate the effective of Sanhua decotion(三化汤) on early brain water content,penetration of blood brain-barrier and the expression of aquaporin4(AQP4) in rats with acute cerebral ischemia/reperfusion(I/R).Methods: The string inserting method was employed to reproduce the rat model of focal cerebral I/R SD rats were randomly divided into sham operation group,model group and Sanhua decotion group.After I/R,each group was then divided into 6 hours,12 hours,24 hours,48 hours and 72 hours five subgroups.The brain water content was detected by the ratio of wet/dry weight to evaluate cerebral edema and the blood-brain barrier(BBB) damage was observed by Evan′s blue(EB) staining.The AQP4 mRNA and protein expressions were measured by reverse transcription-polymerase chain reaction(RTPCR) and immunohistochemistry respectively.Results: Compared with sham operation group,the brain water content,EB content,AQP4 protein and AQP4 mRNA expressions were increased at every time point after I/R in the model group(P
