Objective To explore the effect and preliminary mechanism of the plant-derived immunostimulatory molecule,ophiopogonin,on enhancing the immune response of a subunit vaccine with the receptor-binding domain(RBD)of coronavirus spike protein as the antigen.Methods CCK-8 assay was used to determine the cytotoxicity of ophiopogonin D'(OPD')on bone marrow-derived dendritic cells(BMDCs).Female Balb/c mice were randomly divided into RBD,RBD/OPD',RBD/Alum,and control groups.The immunization dose was 5 μg of antigen per mouse and 100 μg of adjuvant per mouse,and immunization was carried out according to the intramuscular injection immunization procedure on days 0,21,and 42.The titers of specific IgG and its subtype antibodies were detected by ELISA.The cytokine levels in the supernatant of splenocytes were detected using ELISA.The number of splenocytes secreting IFN-γ was detected by ELISpot.Laser confocal microscopy was employed to observe the uptake of antigen by BMDCs.The phagocytic ability of BMDCs for antigen was quantitatively analyzed by flow cytometry.The mechanism of its enhanced immune effect was preliminarily explored using transcriptomics technology combined with bioinformatics research.Results When the concentration of OPD'was less than 5 μg/mL,the survival rate of BMDCs was 100%.After a single intramuscular injection in mice,except for a slight decrease in body weight,the other biochemical indicators were within corresponding normal ranges.After intramuscular injection immunization of the vaccine,the titers of serum-specific IgG,IgG1,and IgG2a in the RBD/OPD'group were significantly higher than those in the RBD group(P<0.05).Compared with the RBD group,the RBD/OPD'group induced a high-level Th1 cell immune response of IL-1β,TNF-α,and IFN-γ(P<0.01)and had more lymphocytes secreting IFN-γ(P<0.001).Laser confocal microscopy displayed that BMDCs took up more antigens after OPD'treatment,which was further confirmed with flow cytometry in quantitative analysis on antigen uptake rate(P<0.01).Transcriptomics results indicated that there was more significant enrichment of the PPAR signaling pathway in the RBD/OPD'group than the RBD group,suggesting that OPD'may activate the PPAR signaling pathway to exert its adjuvant effect.Conclusion OPD'effectively enhances the immune response of the RBD subunit vaccine,and its action mechanism may be related to the activation of the PPAR signaling pathway.

Objective To analyze the characteristics of resilience among empty-nest elderly in community with exploratory latent class analysis method and provide a basis for making targeted intervention measures according to different characteristics.Methods This study enrolled 203 empty-nest elderly people as the study respondents.The general information,resilience of empty-nest elderly were investigated using the general information questionnaire,Connor-Davidson resilience scale in Chinese version.Latent class analyze (Mplus 7.0) was used to discuss the characteristics of resilience among empty-nest elderly.Results The results showed that the resilience of empty-nest elderly had obvious group characteristics and the statistical indicators(AIC,BIC,Entropy and so on) can support the three categories of potential model(P=0.006).Based on the conditional probability on the various items of the questionnaire in each category,they were respectively named high resilience group (54 cases (26.6％)) and low pressure resilience group (82 cases (40.4％)) and low resilience group(67 cases (33.0％)).Further study showed that high resilience and low resilience group had a significant effect of age (P＜0.05),low pressure resilience and low resilience group had a significant effect of gender(P＜0.05).Conclusion The resilience of empty-nest elderly in communities can be divided into three categories according to its characteristics.Each category has different proportion of gender and age.

Objective To analyze the status quo and hot spots in studies on family caregivers by bibliometric analy-sis. Methods PubMed-covered papers on family caregivers from 2011-01-01 to 2016-07-01 were retrieved and analyzed by BICOMS and gCLUTO to generate subject heading clustering curve, subject heading clustering matrix and subject heading tree. Results Six hot spots were detected in studies on family caregivers from 4945 papers from which 82 subject headings were extracted with a frequency≥36 , including social and psychological intervention of family caregivers for ischemic stroke patients, intervention of family caregivers for mental disease patients, mental health of family caregivers, mental health intervention of family caregivers for PD patients, mental health of family caregivers for AIDS patients, and mental health of family caregivers for tumor patients. Conclusion Importance is attached to the studies on family caregivers by foreign and domestic scholars with their studies focused on the inves-tigation and intervention of mental health in family caregivers.

Objective To evaluate the advantages of nasal septum deviation with three lines reduce tension septorhinoplasty under endoscopy.Methods Three lines reduce tension septorhinoplasty of 48 cases were compared with nasal septum submucous resection(Killian) of 43 cases.Results Followed-up for 3 months to 1 year postoperation,study group were remedied and no clinical symptoms,no complications,control group had septal perforation of 1 case,nasal synechia of 2 cases,saddle nose of 1 case.The incidence of two groups postoperative complications had statistically significant difference.Conclusion Compared with Killian,three lines reduce tension septorhinoplasty was easy to stuty and have the advantages of micro-invasion,safety and efficiency,and was favorable recovery nasal cavitysinuses physiological functions.
