Effect of 1,6-Fructose diphosphate capules on animal cerebral is chemia

OBJECTIVE:To inves tigate the therapeutic effects of 1,6-fructose diphosphate(FDP) in mice and rat s which were in the induced states of anoxia and acute cerebral ischemia.M ETHODS:Anoxia was induced in mice with a closed normobaric hypoxia chamber .The clinical condition of acute cerebral ischemia was simulated in mice and rat s by decollating and ligating both carotid arteries.FDP was given intragastrical ly in all tests.RESULTS:Intragastric FDP markedly prolonged the surv ival time of mice which were in the induced states of anoxia and acute cerebral ischemia. Intragastric FDP markedly decreased the WBR(wet wt of brain/body wt×1 00) and water weight of brain and inhibited the rise of serum CK in rats with ac ute cerebral ischemia.Intragastric FDP also improved the capacity of blood passi ng through the blood vessel. There was a change in serum LDH,but it was not sign ificant.CONCLUSION:FDP has therapeutic effects in mice and rats to c ounteract the tissue damage caused by the experimentally induced states of anoxi a and cerebral ischemia.

The effects of amiodarone on QT dispersion in patients with atrial fibrillation

OBJECTIVE:This study evaluated the eff ects of amiodarone on the myocardial repolarization in the patients with atrial fibrillation.METHODS:QT dispersions measured from 12-lead standard surface ECG were retrospectively assessed in 48 patients treated with Amiodarone ,and compared with that of paroxysmal atrial fibrillation and health groups.RESULTS:No disparity in QT interval and QT dispersion was found between paroxysmal atrial fibrillation and health group(QT:405±44vs397+30ms.QTc:448±47 vs424±26ms,QTD 29±17vs27±16ms,QTcD:28±19vs26±18ms,P>0.05). However,compared with these two group,we found that both of the QT interval and QT dispersion significantly increased in Amiodarone group(QT:465±53vs405±44 or 397±30ms,QTc:449±54vs448±47 or 424±26ms,QTD:47±20vs29±19 or 27±16ms,QTcD :49±21vs28±19 or 26±18ms,P<0.01).CONCLUSION:Amiodaro ne not only effectively control atrial fibrillation with QT interval prolongatio n but also increased QT dispersion.

Efficacy observations of repaglinide in the treatment of type 2 diabetes among 68 cases

OBJECTIVE:To evaluate the therapeu tic effectiveness of repaglinide in lowering the FBG and PPBG in type 2 diabetes among 68 patients.METHODS:68 cases were divided at random into two groups:Repaglinide group(34 patients, 17female, 17male) and Placebo-controlled gr oup(34 patients,16female and 18male).0.5mg repaglinide was taken orally before e ach meal in therapeutic group for 4 weeks,while the cases in control were treate d by placebo.RESULTS:Repaglinide could effectively decrease the leve ls of FBG,PPBG and HbA1C with significant difference when compare with the place bo(P<0.01).At the meantime,it could also lower the Cholesterol and triglyceride levels when compared with the control group(P<0.01).CO NCLUSION:Repaglinide was effective in lowering the levels of FBG,PPBG and HbA1C in type 2 diabetes patients without obvious side effects.

Inhibition of lmmediate and delayed type hypersensitivity by the 70% ethanolic extract from Fructus Kochiae Scopariae

OBJECTIVE:To study the inhibitory activities of the 70% ethanolic extract from Fructus Kochiae Scopariae(FKS) on the immediate and the early and late phases of delayed type hypersensitivity(DTH),and explore its mechanisms of action.METHOD:In vitro,FKS was pre-incubated with rat peritoneal mast cells and isolated guinea pig ileum,respectivvly.Its effects on histamine release elicited by compound 48/80 and ileum constriction by 5-HT were observed.In mouse DTH models upon challenge with SRBC or PC,the effects of FKS and total saponin isolated from it on the early and late phases were studied by oral administration after antigen challege.RESULTS:FKS produced a concentration-dependent reduction of histamine release from rat peritoneal mast cells and showed an inhibitory tendency on the guinea pig ileum constriction.Furthermore,FKS and its saponin significantly suppressed the two phases of DTH in mice,which were stronger on the early phase than on the late one.CONCLUSION:These findings indicated that FKS inhibited immediate hypersensitivity and the early phase of DTH mainly due to its stabilization of mast cell membrane and reduction of release of histamine,5-HT and other anaphylactic mediators,and inhibited the late phase of DTH maybe due to its anti-inflammatory activity.Saponin was the active component of FKS responsible for its inhibition on DTH.

Effects of Naokangtai capsule on the ability of learning and memory of rats with Alzheimer′s disease induced by quinolinic acid

OBJECTIVE:To investigate the effect of memory and learning of Naokangtai capsule on AD rats.METHOD:Experimental model of Alzheimer′s disease was made with rats induced by intracranial injection of quinolinic Acid.RESULTS:Naokangtai capsule could increase the ability of memory and learning of AD rats,adjust the level of monoamine transmitters in the brain and hormones in the blood and improved electro-encephalogram of AD rats significantly.CONCLUSION:Naokangtai capsule possesses obvious effects of improving memory and learning of AD rats.

Protective effect of gypenosides on brain ischemia induced by photochemical occlusion of middle cerebral artery in rats

OBJECTIVE:To study the protective effects of gypenosides(Gyp) on brain ischemia.METHOD:The middle cerebral arteries occlusion(MCAO) induced by photochemical reaction in rats were used.The changes of the contents of Na+、Ca2+、H2O and TBARS,SOD activity and the infarct areas were monitored.RESULTS:Gyp reduced the infarct areas induced by photochemical reaction and decreased the contents of Na+、Ca2+、H2O and TBARS significantly,but the content of K+ and SOD activity were increased.CONCLUSION:Gyp inhibited MCAO-induced brain ischemia and brain edema.

Effect of Rannasangpei on blood rheology in increased blood viscosity rats

OBJECTIVE:To obeserve the effect of RNSP on blood rheology in increased blood viscosity rat and analysis its machanism that is related to cerebral vascular.METHOD:The animal model of acute blood stagnation was formed quickly in rats by injected Adrenaline and cold water stimulation,the parameter of blood rheology were measured.RESULTS:RNSP 0.5g/kg and 1g/kg markedly decreased whole blood viscosity(ηb) and whole blood reduced viscosity(ηr) at shear rates of 10s-1,50s-1 and 120s-1,specially at low shear rate,deduced the aggregation inder of RBC(AI) and hemotocrit(Hct).At the same time,no effects on bleeding time and clotting time were observed.The effect of RNSP 1g/kg is similar to that of injectio salvia miltiorrhiza(SM).CONCLUSION:RNSP had the inhibition effect on the aggregate and coagulate function of Red blood cells,and would decrease the blood viscosities.It promoted the blood circulation to remove blood stasis,but had no effect on bleeding time and clotting,in normal animals.

The antitumor effects and toxicity of carboplatin and Xiaozhiling administered intratumorally in mice with tumor

OBJECTIVE:To observe the antitumor effects and toxic reaction of carboplatin and Xiaozhiling after intratumor administration in mice with solid S180 tumor.METHODS:30 and 60 mg/kg carboplatin to 10 ml/kg xiaozhiling were administered intratumorally and 30mg/kg carboplatin intravenously every three day for one week in mice with transplanted tumor.The weights of tumor and body,the index of spleen and thymusgland were measured.RESULTS:Compared with normal saline group,the growth of tumor in 30,60 mg/kg carboplatin-treated and 10 ml/kg xiaozhiling groups was all inhibited,with inhibitory rates of 27.4%,41.9% and 25.6%,respectively.The indexes of spleen were decreased by 38.6% and 49.8%,and thymusgland 4.9% and 14.2% in mice treated with 30 and 60 mg/kg carboplatin,while the indexes of spleen and thymusgland in mice treated with Xiaozhiling were increased by 13.2% and 33.5%,respectively.The average mouse weights had no significant difference in every group.CONCLUSION:Carboplatin and xiaozhiling showed significant antitumor effects when injected intratumorally.The indexes of immune organs was decreased by carboplatin,while increased by Xiaozhiling.

Effect of captopril on plasma activity of t-PA and PAI-1 in rat

OBJECTIVE:To assesss the effect of captopril on the plasma activity of t-PA and PAI-1 in rat.METHODS:Captopril was administrated at 3 different doses,to observe effects on the plasma activity of t-PA and PAI-1 in rat.RESULTS:After the treatment captopril,the plasma activity of t-PA increased significantly(P<0.01),the plasma activity of PAI-1 and the activity ratio of PAI-1/t-PA decreased significantly(P<0.01).This effect was in a dose-dependent manner to some extent.CONCLUSION:Captopril can activate the activity of fibrinolytic system.This effect is in a dose-dependent manner to some extent.

Study on the technology of Shaping in Gongyujing capsule

OBJECTIVE:To choose the best process to clear impurity.To fix the type and filling capacity of capsule.To provide the relative humidity of workshop.METHODS:The nature of extract prepared with different methods were studied.The pile-density and critical relative humidity were determined.RESULTS:The extract that was prepared by high-speed centrifuge was easier to dry.The fluidity of the powder was better.The hygroscopicity of the powder was lower.CONCLUSION:The high-speed centrifugal method was chosen.The type of capsule was defined as 0#.The package was 0.45 gram for each capsule.The relative humidity of workshop was below 54%.
