Effect of subinhibitory concentrations of antibiotics on Staphylococcus aureus producing panton-valentine leukocidin

Objective To explore the effect of subinhibitory concentrations (sub-MICs) of four antibiotics on pan-ton-valentine leucocidin ( PVL ) expression by Staphylococcus aureus . Methods Eight strains of PVL-positive Staphylococcus aureus were isolated by PCR. The MICs of these antibiotics were determined by constant dilution. then, PVL expression of Staphylococcus aureus was measured at different MICs of these antibiotics by ELISA. Re-sults Clindamycin, linezolid and tigecycline all could reduce the expression of PVL at 1 / 8 MIC ~ 1 / 2 MIC. after 4 and 6 hours of culturing, the relative PVL mRNA expression was reduced about 17% ~ 82% , 8% ~ 85% and 11% ~ 78% by clindamycin, linezolid and tigecycline, respectively. The protein expression of PVL was reduced by 65% and 83% under 1 / 4 and 1 / 2 MIC of clindamycin, and reduced by 40% and 61% under 1 / 4 and 1 / 2 MIC of linezolid, and reduced by 61% only at 1 / 2 MIC of tigecycline. There had no effect of vancomycin on PVL expres-sion. Conclusion The effects of four antibiotics on PVL expression at sub-MICs were different. Clindamycin and linezolid could significantly reduced PVL expression. Tigecycline colud reduced PVL expression only at high sub-MICs. Vancomycin had no effect.

Experimental study of Apobec-1 recombinant adenovirus in treatment of renal hyperlipemia

Objective To explore the effect of the liver acquired expression of apolipoprotein B editing catalytic polypeptide-1 (Apobec-1) on hyperlipidemia of renal disease. Methods Thirty healthy ordinary level male New Zealand rabbits were randomly divided into three groups: sham operation group, nephropathy group, and Apobec-1 treatment group (Each group has 10 rabbits). Adapt feeding for one week, nephropathy group and Apobec-1 treat-ment group underwent left nephrectomy, and one week later, adriamycin (4 mg / kg) was used to construct the ne-phropathy model by ear vein injection. The eleventh week after operation, apobec-1 recombinant adenovirus (1 × 1013 Virus/ kg) was injected by ear vein in apobec-1 treatment group. The twelfth week after operation, all rabbits were sacrificed. Right kidney, liver, blood and 24h urine were left. In three groups, 24 hour urinary protein (24UPr), albumin (Alb), blood urea nitrogen (BUN), creatinine (Cr), blood lipid were detected. Renal pa-thology was observed by HE staining. Expressions of liver apobec-1, apolipoprotein B48(ApoB48)were observed by Western blot. Results ① Compared with the sham operation group, nephropathy group showed that 24UPr, BUN, Cr, total cholesterol (TC), total triglyceride (TG), very low density lipoprotein (VLDL-C), low density lipoprotein (LDL-C), apolipoprotein B100 (ApoB100) were increased(P < 0. 05), but Alb was decreased(P <0. 05). ② Compared with the nephropathy group, Apobec-1 treatment group showed that TC, TG, VLDL-C, LDL-C, ApoB100, ApoB48 were decreased(P < 0. 05). ③ Compared with the sham operation group, Apobec-1 treat-ment group showed that 24UPr, BUN, Cr were increased( P < 0. 05), but Alb, ApoB48 were decreased( P <0. 05). ④ Compared with the sham operation group and nephropathy group, Apobec-1 treatment group showed that the expression of Apobec-1 and ApoB48 were up-regulated (P < 0. 01). Conclusion When liver aquires expres-sion of Apobec-1 in hyperlipidemia of renal disease, it can reconstruct ApoB mRNA, increase the synthesis of ApoB48-lipoprotein, and play a certain lipid-lowering effect.

Cerebral protective effect of isoflurance preconditioning for adult rat model with local cerebral ischemia

Objective To investigate the brain protective effects of isoflurane preconditioning for local cerebral is-chemia adult rat model by detecting HIF-1α, HO-1 and Bcl-2 expression level. Methods 84 two-month old Spra-gue-Dawley male rats were randomly divided into sham group, ischemia-reperfusion group ( MCAO group) and isoflurance preconditioning group (ISO group). ISO group was exposed to isoflurane for 30 min and then underwent a 90 min middle cerebral arterial occlusion (MCAO), while MCAO group underwent MCAO only. At 24 h after reperfusion all the groups were investigated by modified neurological severity score for neurological deficits, TTC staining for infarct percentage. Cerebral cortices were harvested for HIF-1α, HO-1 and Bcl-2 protein expression level detection at 6 h, 24 h and 72 h after reperfusion using RT-PCR and western blot. Results The mNSS score and infarct percentage in ISO group decreased significantly compared with those of MCAO group at 24 h after reper-fusion(P < 0. 05). Compared with the MCAO group, ISO group had significantly up-regulated protein levels of HIF-1α, HO-1 and Bcl-2 at 6 h and 24 h after reperfusion(P < 0. 05). There was no difference between ISO group and MCAO group for 3 gene expression at 72 h(P > 0. 05). Conclusion Isoflurane preconditioning plays short-term cerebral protective effect through the up regulation of HIF-1α, HO-1 and Bcl-2 gene expression.

Effect of long non-coding RNA HOTAIR on the proliferation and apoptosis of renal cancer cells

Objective To investigate the effect of long non-coding RNA (LncRNA) Hox transcript antisense inter-genic RNA ( HOTAIR ) on the cell proliferation and apoptosis of renal cancer cell lines 786-O and ACHN. Methods Small interfere RNA ( siRNA ) that aims to down-regulate HOTAIR expression was transfected into two renal cell lines respectively, and the transfection efficiency was evaluated by qRT-PCR. Then the MTT assay,Ho-chest staining assay and enzyme-linked immunosorbnent assay(ELISA) were used to detect cell proliferation and apoptosis. Results The expression of HOTAIR could be down-regulated effectively by the siRNA (P < 0. 05). Down-regulation of HOTAIR could inhibit the proliferation(P < 0. 05) and increase apoptosis(P < 0. 05) of two re-nal cancer cells. Conclusion HOTAIR plays a role in promoting cell growth of renal cancer.

Establishment of irradiation induced heart injury in C57 mice model and identification of serum marker

Objective To establish and investigate C57 BL/6 mice model of radiation induced heart injury and ser-um marker. Methods Twenty eight female C57BL/6 mice were randomly divided into 3 groups:normal group (n=4 ) , 18 Gy radiation therapy group ( n=12 ) , 25 Gy radiation therapy group ( n=12 ) . The mice were weighed every week. Respectively, 0, 8 and 16 weeks after irradiation, the pathological changes of myocardial tissue were observed by hemetoxylin-eosin( HE) staining and made pathological score. Inferior vena cava blood was collected to take cardiac troponin I ( cTnI) test. Results Compared with the normal group,the weight of radiation group went down first then went up slowly. The myocardial tissue from mice had obvious histopathological changes. Acute in-flammation was the main change in the early days. At the late stage, progressive fibrosis was the main characteris-tic. With the increase of the dose, the inflammatory reaction and fibrosis degree had aggravated, pathological chan-ges had occurred earlier. cTnI showed a trend of higher performance over time, and had a correlation with patholo-gy. Conclusion We successfully established radiation induced myocaridal injury model. As a noninvasive serum marker, cTnI can be used as the evaluation standard of radiation induced myocardial injury animal model.

JNK contributes to the proliferation inhibition of the rat fetal neural stem cells triggered by EI

Objective To explore the general anesthetic EI on the rat fetal neural stem cell proliferation(NSCs) and the role played by JNK in the influence. Methods The cultured rat fetal NSCs were randomly divided into 6 groups (n=8 each):normal group (group N);fat milk group (group F);EI groups(including 8. 12,9. 80,12. 04 mmol/L EI) , 9. 80 mmol/L EI group+20 μmol/L SP600125 group ( group EISP) . After incubated for 12 hours, the cellular effects of EI and cell viability were evaluated by MTT reduction assay. The apoptotic rate of the rat NSCs were determined by flow cytometry, and the expression of protein Caspase-3 was observed by Western blot. Results There was no statistical difference in cell viability, apoptotic rate and protein expression of Caspase-3 in group N and group F. EI group had higher cell apoptotic rate(P<0. 01), protein expression of Caspase-3 (P<0. 05 ) , but lower cell viability than group N ( P<0. 01 ) . And significant differences were found between three do-ses of EI groups (P<0. 05). Compared with EI group, lower cell apoptotic rate (P<0. 01), protein expression of Caspase-3 but higher cell viability were observed in group EISP ( P<0. 05 ) . Conclusion JNK plays an important role in EI-induced cytotoxicity possibly in a dose-dependent manner.

Effect and mechanism of miRNA-4465 overexpression in the migration and invasion of breast cancer MDA-MB-231 cells

Objective To investigate the effect of overexpression of miRNA-4465 on invasion and metastasis of breast cancer MDA-MB-231 cells and its mechanism.Methods MiRNA-4465 minics was transfected into the MDA-MB-231 cells,and real-time PCR was used to detect the expression of miRNA-4465.Transwell assay was used to investigate the migration and invasion capability of cells after being transfected.Bioinformatics analysis were performed to predict the potential targets of miRNA-4465,and finally,luciferase reporter plasmids assay and west-ern blot was used to confirm the potential target of miRNA-4465.Results Comparing with negative control group, the expression of miRNA-4465 in miRNA-4465 minics group was increased significantly.Transwell migration assay showed that migration capability of cells in miRNA-4465 minics group was decreased(P =0.001);transwell inva-sion assay showed that invasion capability of cells in miRNA-4465 minics group was decreased(P =0.010).Lucif-erase reporter plasmids assay showed that comparing with the negative control +psiCHECK2-EZH2 3’UTR(wild type),the fluorescence activity of miRNA-4465 minics +psiCHECK2-EZH2 3’UTR(wild type)was decreased by 66%(P =0.001).Moreover,comparing with the negative control group,the expression of EZH2 protein was de-creased after transfecting miRNA-4465 minics into MDA-MB-231 cells.Conclusion MiRNA-4465 can suppress the invasion and metastasis of breast cancer MDA-MB-231 cells,which may be related to the regulation of EZH2.

The study of correlative affecting factors on re-establishing stereopsis of surgically aligned adult concomitant strabismus

To analyze the effect of pre-surgical status on the post-surgical stereopsis in surgically aligned adult con-comitant strabismus.By using Titmus and Randot stereo tests,we measured near stereo acuity of 99 adults who had constant exotropia and were successfully aligned by surgery.A covariance analysis showed that the post-surgical stereo acuity was significantly determined by the type of strabismus (i.e.,whether that's intermittent or not;P =0.001),the duration of pre-surgical exotropia (P =0.002)and the year on-set (P =0.048)but not by the pre-surgical deviation (P =0.133).We conclude that the stereopsis of adult concomitant strabismus can be reestab-lished by surgery,while the extent of such an improvement is determined by the type of strabismus,the duration of pre-surgical exotropia and the year on-set of patients.

The value of ultrasonography in the differential diagnosis for TTTS and sIUGR

12 patients with inconsistent growth of twins were involved.The acoustic features were analyzed,inclu-ding 2D and Doppler images.The differential diagnosis was verified during follow-up exam in some patients.8 cases were diagnosed with sIUGR by ultrasonography,meanwhile 3 were diagnosed with TTTS.The rest 1 case could not be etiologically diagnosed due to the bad image conditions.We can differentiate the two diseases by evaluating the umbilical cord insertion,the amiotic fluid and the hemodynamics of the other twin.Ultrasonography is helpful in the differential diagnosis for TTTS and sIUGR.

Correlation between peri-procedural changes of serum inflammatory cytokines and early prognosis in patients with acute myocardial infarction undergoing percutaneous coronary intervention

Objective To investigate the variation and clinical significance of serum levels of inflammatory cyto-kines after percutaneous coronary intervention (PCI)treatment in acute myocardial infarction (AMI)patients. Methods 118 incipient AMI patients with successful underwent PCI (Defined as treatment group,blood samples were collected from pre-operation,12 h after operation,24 h after operation,48 h after operation and 90 d postop-erative follow-up period)and 52 AMI patients with diagnostic coronary angiography (CAG)(Defined as control group,blood samples were collected prior to CAG,12 h after CAG,24 h after CAG,48 h after CAG and 90 d fol-low-up period)were enrolled in this study.Serum levels of IL-6,IL-18,hs-CRP,TNF-αand MMP-9 were detec-ted in all the subjects by enzyme-linked immune sorbent assay(ELISA)and major adverse cardiac events(MACE) occurrence rate was analyzed in 90 days followed-up cases.Results No significant differences in baseline levels of IL-6,IL-18,hs-CRP,TNF-αand MMP-9 were found in the two study groups(P >0.05).No significant differences of levels of IL-6,IL-18,hs-CRP,TNF-αand MMP-9 were found after CAG in control group (P >0.05).The serum levels of IL-6,IL-18,hs-CRP and TNF-αafter PCI were significantly increased (P <0.01 )while no significant differences were found in level of MMP-9 (P >0.05)in PCI group.There were significant differences of levels of IL-6,IL-18,hs-CRP and TNF-αbetween MACE group and without MACE group after PCI.The multivariable lo-gistic analysis showed that IL-6,IL-18,hs-CRP and TNF-αwere risk factors of MACE after 90 days follow-up. Conclusion The concentrations of serum IL-6,IL-18,hs-CRP and TNF-αare significantly increased in AMI pa-tients treated with PCI.PCI operation may induce inflammatory reaction.High serum levels of peripheral inflamma-tory cytokines IL-6,IL-18,hs-CRP and TNF-αhave an important role in major adverse cardiac events(MACE)and short-term prognosis in the first AMI patients treated with successful primary PCI.
