Correlation between polymorphisms in TNF-α and IL-10 genes and plasma cincentration with diffuse large B-cell lymphoma

Purpose To explore the association between single nucleotide polymorphisms (SNPs) of TNF-αand IL-10 genes as well as their plasma levels and pathogenesis of diffuse large B-cell lymphoma (DLBCL).Methods Snapshot SNP genotype technique was used to assess genetic variation in 6 SNPs for TNF-α and IL-10 in 38 DLBCL cases and 77 healthy controls,and enzyme-linked immunosorbent assay (ELISA) was used to measure the plasma concentrations of TNF-α and IL-10 in the above population.Results Plasma IL-10 level in DLBCL cases was higher than that in controls (P < 0.05),but there was no significant difference in plasma TNF-αlevel between the two groups (P > 0.05).Of all the candidate SNPs,the geneotype distribution for TNF-α-863C/A showed significant differences between the case and control groups (P < 0.05),and carriers with CA/AA genotypes had more two-fold risks of DLBCL than those with the CC genotype (95% CI =1.091-4.765,P =0.028).However several other candidate SNPs showed no significant difference in the geneotype distributions between the cases and controls (P > 0.05).After excluding the effects of disease status on plasma TNF-alpha and IL-10 levels,the polymorphism of each gene had no significant effect on the corresponding plasma concentration (P > 0.05).Conclusion TNF-α-863C/A polymorphism may be associated with the risk of DLBCL,and IL-10 play an important role in occurrence and development of DLBCL.

EGFR mutations detection in non-small cell lung cancer liquid based cytology samples by ARMS method

Purpose To explore the application and characteristics of liquid-based cytology samples of non-small cell lung cancer for detection of EGFR mutations by ARMS method.Methods The positive samples of liquid-based cytology were collected and the DNA of samples was extracted to detect the EGFR mutations by ARMS method and the analyze the association with clinical features,types of samples,pathological types and the contents of tumor cells,etc.Results There were 117 genetic mutations detected in 279 liquid-based cytology specimens,with the mutation rate of 41.9%.The mutation rate of adencarcinoma was 44.7% and the other was 11.3%.When the tumor ceils in cytology samples were abundant,medium,small clusters and few,EGFR gene mutation rate were 53%,44%,45% and 44% respectively.19Del was 51.9%.Exon 21L858R missense mutation occurred at 39.4% of EGFR mutations.Conclusion All liquid-based cytology of non-small cell lung cancer samples are adequate for EGFR mutation analysis.In the tumor cell-rich samples EGFR gene mutation rate is higher than that of the less tumor cells samples.19Del is the most common type of EGFR mutations.

Expression of Uba-2 in hepatic carcinoma and its effect on the proliferation, invasion and metastasis of hepatoma cells

Purpose To investigate the effect of differential expression,over expression and low expression of Uba-2 in hepatic carcinoma on the proliferation and invasion of hepatoma cells.Methods The expression of Uba-2 mRNA in clinical specimens was detected by qRT-PCR.Mter transfection of Uba2 over expression vector and siRNA expression vector into hepatoma cell HepG2,the effect of Uba-2 on proliferation of hepatoma cells was detected by MTS assay while the effect on the invasion and metastasis of hepatoma cells was detected by cell migration/invasion assay.The expression levels of TGF-β1-3,VEGF and Snail proteins in hepatoma cell HepG2 were detected by Western blot.Results The expression of Uba-2 was high in hepatic carcinoma.Compared with that in adjacent liver tissues,there were significant differences (P < 0.01).qRT-PCR detected over expression or low expression of Uba-2 in HepG2 cells.Cell proliferation data showed that the expression level of Uba-2 was related to the proliferation ability of hepatoma cell HepG2,and siRNA interferring the expression of Uba-2 in silent HepG2 cells could inhibit its growth.The data of cell migration/invasion showed that over expression of Uba-2 could promote the invasion and metastasis of hepatoma cell HepG2.Compared with cells in the control group,there were statistically significant differences.The results of Western blot indicated that the abnormal expression of Uba-2 affected the expression levels of TGF-β2,VEGF and Snail proteins.Conclusion The high expression of Uba-2 gene in hepatic carcinoma may promote the proliferation,invasion and metastasis of hepatoma cells.

Relevant research of PD-1, PD-L1 protein expression and K-RAS gene mutations in lung adenocarcinoma

Purpose To investigate the association of the PD-1 and PD-L1 protein expression with K-RAS gene mutations in lung adenocarcinoma.Methods The protein expression of PD-1 and PD-L1 was detected by immunohistochemical EnVision two-step staining,the K-RAS mutation was examined by realtime fluorescent quantitative PCR.Results The positive rate of PD-1 and PD-L1 was higher in lung adenocarcinoma than benign lung disease (P < 0.01).There was no relationship between PD-1 and PD-L1 protein expression with the gender,age,smoking condition,differentiation,lymph node metastasis and TNM stages (P > 0.05).The K-RAS gene mutations were detectable in 8 patients (22.2%) among 36 lung adenocarcinoma,there was no association between K-RAS gene mutation with the gender,age,smoking condition,differentiation,lymph node metastasis and TNM stages (P > 0.05).The correction analysis showed that there was no relationship between PD-1 and PD-L1 protein expression with K-RAS mutation (P > 0.05).Conclusion The positive rate of PD-1 and PD-L1 is higher in lung adenocarcinoma than benign lung disease,but there is no relationship among PD-1 and PD-L1 protein expression with its clinal pathological characteristics and K-RAS mutation in lung adenocarcinoma.

Children peripheral neuroblastic tumor: pathology, biology and its clinical significance

Purpose To summarize the histological,immunophenotypic feature and MYCN (MYC gene) amplification results of peripheral neuroblastic tumours in children,and to predict its biological behavior and prognosis.Methods The histology and immunophenotype of 100 cases of neuroblastoma (NB) and ganglioneuroblastoma (GNB) were retrospectively analysed,MYCN status was detected in 60 cases.Results Among The average age of 100 cases of GN and GNB was 2.7 years old,and that of males was more than that of females.NB could be divided into three subtypes:undifferentiated,poorly differentiated and differentiating.GNB could be divided into two subtypes:intermixed (iGNB) and nodular (nGNB).Immunohistochemical staining showed neuroblastoma cells were positive for NSE,NF,PGP9.5,Syn,CgA in varying degree.Schwann cells were positive for S-100 and GFAP.MYCN amplification was detected in 1 1.67% of the cases,and no MYCN amplification was seen in iGNB patients.Conclusion The diagnosis of peripheral neuroblastic tumours is mainly based on histological morphology,special tests (immunohistochemistry,electron microscope and cytogenetics) can helpful for identifing undifferentiated neuroblastoma cells.The prognosis of neuroblastoma derived tumors is evaluated according to the age of patients,the classification and staging of tumors,and molecular genetic alterations.

Influence of long chain non-coding RNA (lncRNA)-HOTAIR on migration and invasion ability of endometrial cancer

Purpose To investigate the influence of long chain non-coding RNA (lncRNA)-HOTAIR on endometrial cancer cell proliferation,invasion,metastasis and other biological behaviour.Methods 20 cases of endometrial carcinoma tissue specimen,20 cases of hyperplasia tissue sample and 10 cases of normal tissue specimen were collected.Difference expression of lncRNA-HOTAIR in normal endometrium,hyperplasia endometrium tissue and endometrial carcinoma tissue at all periods were detected with RT-PCR assay.HOTAIR-siRNA transfection into Ishikawa cells was utilized with Lipofectamine 2000.MTT experiment was used to detected the proliferation ability of cells in all groups.Transwell chamber experiment was used to test the migration and invasion ability of cells in all groups.Results The gene expression level of of lncRNA-HOTAIR in endometrial carcinoma group at all stages was prominently increased compared with normal endometrium group (P < 0.05).The expression level of lncRNA-HOTAIR in simple hyperplasia endometrium group and atypical hyperplasia endometrial group was not significantly different (P > 0.05).Cell proliferation,invasion ability and migration ability of HOTAIR-siRNA targeting suppression group were lower than the blank control group and the negative control group significantly (P < 0.05).Conclusion lncRNA-HOTAIR may involved in occurrence and development of endometrial cancer,which may play an important role in the aggression and metastasis of endometrial cancer.

Expression and clinical significance of autophagy gene LC3 in endometriosis

Purpose To study the relationship between autophagy and endometriosis,and its significance.Methods The eutopic and ectopic endometrial stromal cells from patients with endometriosis (n =31) and normal endometrial stromal cells (n =31) were cultured primarily and identified by immunocytochemistry.The ultrastructural changes of autophagic vacuoles were observed by transmission electron microscopy (TEM).RT-PCR and Western blot methods were used to detect LC3 mRNA and protein expression in endometrial cells of endometriosis patients.Results TEM analysis showed the number of autophagic vacuoles of eutopic and ectopic endometrial cells was fewer.RT-PCR and Western blot analysis showed that the expression of LC3 mRNA and protein in eutopic and ectopic endometrium of patients with endometriosis was significandy lower than that of the normal endometrial stromal cells (P < 0.05).There was no significant difference in the expression of LC3 mRNA between eutopic and ectopic endometrium of patients with endometriosis (P > 0.05),but the expression of LC3 protein in ectopic endometrium was higher than that in eutopic endometrium of patients with endometriosis (P < 0.05).Conclusion Autophagy activity is downregulated in eutopic and ectopic endometrial stromal cells with endometrosis.Autophagy may be involved in the pathogenesis of endometriosis.

Expression of HIF-1α and BCL-2 in salivary adenoid cystic carcinoma and its clinical significance

Purpose To investigate the relationship of HIF-1α,BCL-2 and clinicopathological features in salivary adenoid cystic carcinoma by detecting the levels of two proteins.Methods Immunohistochemical of SP staining assay was used to detect the level of HIF-1α and BCL-2 expression in fifty-six section from primary resection of salivary adenoid cystic carcinoma.Furthermore,the correlation between the expressions of two proteins with clinical data was evaluated through differently statistical analysis.Results Thirty-six samples (64.3%) were found to express HIF-1α.The level of HIF-1α was significantly correlated with TNM stage of tumors and the primary site of salivary adenoid cystic carcinoma (P < 0.05).Positive BCL-2 expression was detected in fifty-five cases (98.2%).The level of HIF-1 α was positively correlated with that of BCL-2 (P < 0.05)and the significant correlation between HIF-1α and TNM stage was dependent upon the strong expression of BCL-2 (P < 0.05).Conclusion It is therefore indicated that the expression of HIF1α and BCL-2 may influence the clinical stage of salivary adenoid cystic carcinoma.

Primary pulmonary lymphoepithelioma-like carcinoma: a clinicopathological analysis of twenty cases

Purpose To study the clinicopathological characteristics,treatments and prognosis of primary pulmonary lymphoepithelioma-like carcinoma (LELC).Methods Twenty cases of LELC were collected and the clinicopathological characteristics,immunohistochemical expression,survival data and clinical treatments were analyzed,with review of the literatures.Resuits Histopathologically,it was characterized by the syncytial pattern of growing with large vesicular nuclei,conspicuous nucleoli,and a marked lymphocytic infiltrate in the stroma that was similar to undifferentiated nasopharyngeal carcinoma.The immunohistochemical staining of CKpan showed positive expression in all cases (100%).Also the positive expression of in situ hybridization of Epstein-Bar virus-encoded RNA (EBER) were in all cases (100%) while the positive cases of CK5/6,p63 and p40 were 19,18,and 16 respectively.But gene mutations such as EGFR,K-RAS were negative.All the LELC patients in this cohort were survived until the deadline of follow-up.Comprehensive treatments were done such as surgical resection,radiotherapy and chemotherapy.Conclusion Primary pulmonary LELC is a rare disease that histomorphological analysis and immunohistochemistry are the main methods of diagnosis for it.It must be excluded the metastasis carcinoma from other locations.While both EGFR mutation and anaplastic lymphoma kinase (ALK)mutation could be commonly detected in non-small cell lung cancer,but seldom in LELC of lung.The high expression of PD1/PD-L1 may provide a rationale for immunotherapy in this subtype of lung cancer.Currently,experimental therapy is still taken in the pulmonary LELC.

Cribriform adenocarcinoma of lung:a clinicopathological analysis of twenty cases

Purpose To investigate the clinicopathological features,diagnosis and differential diagnosis,and genetic alteration of lung cribriform adenocarcinoma (LCA).Methods Clinicopathological features and immunophenotype were retrospectively evaluated in twenty LCA cases collected from Fujian Provincial Hospital,combined with genetic mutation analysis of EGFR.Results The tumors occurred in 5 women and 15 men aged from 39 to 74 years (median =59.25).Diameter of masses ranged from 1.5-5.5 cm (median =3.265).Histologically,the tumors were unencapsulated and were divided by fibrous septa into lobules.Major parts of the lesions were composed of areas with solid and microcystic growth patterns.The most striking cytological feature was that the tumor cell nuclei were pale staining with a ground glass feature,and they often appeared to overlap.Immunephenotypically,the tumor cells were positive for TTF-1(20/20),Napsin A (18/20),SPB(19/20),CK7(20/20),CD56 (2/20),EGFR (7/20),β-catenin (4/20),ALK (2/20).Tg,CDX-2,Syn,CgA,S-100 and SMA were negative.Gene mutation detection confirmed that EGFR genes were mutating in nine cases,and eleven cases were wild type.The twenty cases were all performed for the completely surgical resection.20 cases accepted chemotherapy,and 4 cases unaccepted chemotherapy.Conclusion LCA is a rare tumor with distinct morphologic feature.Clinically and pathologically,it needs to differentiate from alveolus adenocarcinoma,adenoid cystic carcinoma and papillary carcinoma of the thyroid,and so on.The primary treatment for LCA is completely surgical excision and chemotherapy.Otherwise its prognosis is poor.
