The number and significance of CD+68 positive cells in hepatocellular carcinoma and cirrhosis tissue

Objective:To investigate the number of CD+68 cells in hepatocellular carcinoma (HCC),paraneoplastic tissues,cirrhosis tissues and normal liver tissues and their clinical significance.Methods:Surgical specimens from 60 cases of HCC,62 cases of cirrhosis and 23 cases of normal liver were investigated by immunohistochemical staining of CD+68with streptavidin-horseradish peroxidase detective system.Results:(1)The order of the mean number of CD+68cells in the tissues from the highest to the lowest was:paraneoplastic,cirrhosis,normal liver and HCC tissues(P〈0.01);(2)The number of CD+68cells in the tissues decreased successively with the decrease of HCC differentiation (P〈0.05);(3)There was no relationship between the number of CD+68cells in HCC and its clinical TNM stage;(4)The number of CD+68cells in the cases with metastasis within 15 months was significantly lower than that without metastasis (P〈0.05).Conclusion:The number of CD+68cells might be an important marker to estimate the immune status and a useful factor to predict the prognosis of HCC patients.

Expression of CEA,ICAM-1 on lymphatic endothelial cells in rectum cancer of human

Objeetive:To study the expression of cell adhesion molecules on the lymphatic endothelial cells In the rectum cancer and peritumoral lymph nodes as well as their relationship with the diffusion of cancer cellS. Methods: In this Study the immunhistochemistry method was employed to observe the expression of CEA, ICAM-1 and CD31 on rectum cancer lymphatic endo-thelial cells. Results :The results demonstrated that CEA and lCAM-1 were highly expressed on the lynphatio endothelial cells in the peritumoral rectum tissues and lymph nodes. It was, howev-er,negative in normal lymphatic endothelial cells. CD31 was expressed on both lymphatic endothe-Ilal cells in the periturmral rectum tissues and lymph nodes and vessel endothelial cells of the normal human. Conelusion: The study suggests that there are relationships between the dis-semination of cancer cells and excession of CEA, lCAM-1 and CD31 in lymphatic endothelial cells.The CAE and ICAM-1 seem to induce stable adhesion between oancer cells and endothelial cells.

Clinical significance of the expression rates of cytokeratin5/6/18 positive cells in peripheral blood cells from patients with breast carcinoma

Objectives:To investigate the clinical significance of the expression level of cytokeratin5/6/18 positive cells in peripheral blood cells from patients with breast carcinoma.Methods:The expression rates of ck5/6/18 positive cells in peripheral blood cells from 56 patients with breast carcinoma were measured and analyzed by flow cytometry.Results:The expression rates of ck5/6/18 positive cells in peripheral blood cells from 56 patients with breast carcinoma was 46.4% and there was significant differences compared with healthy volunteers and the patients with breast benign diseases.The positive rates increased significantly with the pathological staging and show no significant relationship with the ER and PR.Conclusion:The expression of ck5/6/18 positive cells in peripheral blood cells correlates significantly with the pathological staging of breast carcinoma and it could be an effective signal to clinical monitor and prognosis.

Investigation of inhibition of liver metastasis of colon carcinoma on the nude mice models with the system of TK/GCV and CD/5-Fc

Objectives:To explore the inhibition effect of TK/GCV and CD/5-FC system on liver metastasis of colon carcinoma.Methods:Forty nude mice were divided into 4 groups randomly,which were the control group,TK group,CD group and TK-CD group with 10 nude mice in each group.The mice in the control group were injected SW480 cells in the spleen and NS into the peritoneal cavity.The mice in other groups were all injected SW480/TK-CD cells in the spleen.And GVC,5-Fc,GVC+5F-c were injected into the peritoneal cavity of the mice in the TK group,CD group and TK-CD group respectively.Liver metastasis rate,the number of liver metastasis,the pathological changes of the tumor tissues under the electronic microscope,tumor cell's apoptotic index and life span and other indexes of nude mice in four groups were analyzed.Results:Liver metastasis rate of nude mice in every treatment group was lower than that in control group.Compared with control group,average number of liver metastasis decreased,life span of nude mice was prolonged,and the apoptosis rate of cancer cell in liver metastasis increased.The combined genes in the TK-CD group worked more markedly,and there was mutual effect between TK/GCV and CD/5-Fc.Conclusions:Synergistic effect was acquired combining TK/GCV with CD/5-FC system and it inhibited the formation of liver metastasis of colon carcinoma efficiently.

Comparative study of isoflurane and sevoflurane on the effects of anesthesias in abdominal surgeries with the close-loop anesthesia system

Objectives: To study the differences between sevoflurane and isoflurane in abdomen surgery anesthetized with the target-controlled close-loop anesthesia system.Methods: Forty patients (ASA I-II) were divided into sevoflurane group (n=25) and isoflurane group (n= 15).BIS value of 55 was the control variable index.The target-controlled infusion system of sevoflurane was administered for the maintenance of anesthesia in both groups after intravenous nduction. The BIS values,blood pressures,heart rates and doses of assistant drugs were recorded and analyzed.Results: No statistical difference was observed in BIS、HR and pulse pressure in two groups. Study of Diallyl trisulfide affection on human colonic carcinoma HT29 cells proliferation and its mechanisms

Objective: To study the affection of Diallyl trisulfide on cultured human colon carcinoma cell line HT-29 and its mechanism. Methods: The viability of cells after various treatments were determined using the method of MTT. Flow cytometry were used to analyze the change of cell cycle and Bcl-2, Bax levels. Results: The proliferation of human colon carcinoma HT-29 cells were inhibited by Diallyl trisulfide. The inhibition was associated with dose and time dependence. Flow cytometry results showed that: 1) When cells were treated with Diallyl trisulfide at the concentration of 10 ?g/mL for 12 h and 24 h, the percentage of G0/G1 phase cells was decreased and that of G2/M phase cells was significantly increased compared with those in the control group.2) Diallyl trisulfide down regulated Bcl expression, while up-regulated bax expression. Conclusions: It was suggested that Diallyl trisulfide inhibited HT29 cell proliferation. And these were associated with arrest of cell cycles and down regulation of Bcl2/Bax ratio.

Study on endostatin inhibitation of the growth of transplanted hepatic cancer in nude mice

Objective: To explore the effect of endostatin on the growth of human liver carcinoma in vivo. Methods: Mice endostatin gene was transferred into SMMC 7721 cancer established in nude mice. Immunohistochemistry and Western blot analysis were applied to examine the transfection and expression. Effect of vascular endothelial growth factor-C on invasive capability and proliferation of human cholangiocarcinoma cells

Objective: To investigate the effect of vascular endothelial growth factor-C (VEGF-C) on invasive capability and proliferation of human cholangiocarcinoma cells. Methods: The effect of VEGF-C on FRH0201 was assayed by MTT. The cycle pattern and apoptosis was assayed using flow cytometry. The effect of VEGF-C on homotypic adhesion and metastasis in FRH0201 was examined with 3H-TdR infiltration and Boyden chamber. Results: VEGF-C could enhance the proliferation of FRH0201 in a dose and time dependent manner. And VEGF-C could inhibit cell apoptosis significantly. After cultured for 2 hours with 1, 5, 10ng/ml of VEGF-C, there were more cells in the lower chamber than the control group. After 60, 90, 120 minutes induction by 1ng/ml、5ng/ml、10ng/ml, the cells showed significantly lower homotypic adhesion than that of the control group. Conclusions: VEGF-C could enhance the proliferation of FRH0201 and inhibit cell apoptosis. VEGF-C could decrease homotypic adhesion of FRH0201 and might be a cause of the metastasis.
