The effects of c-fos antisence olioneucleotide on the intimal proliferation of autogrfted veins

Objective:To investigate the effects of c-fos antisence olioneucleotide on the intimal proliferation of autogrfted veins with antisence technology.Methods:The external jugule veins were grafted into common carotid arteries in 20 rabbits and were divided into test group and control group randomly.The anastomosis and transplanted vein were coated with c-fos antisence olioneucleotide glue gel in the test group,while the contral group were merely coated with glue gel.The autografted veins were removed and measured by means of pathology and immunohistochemistry two weeks later.Results:The results show that the thickness of the venous intima,the degree of the vascular stricture,the expression of PCNA and the numbers of vascular smooth muscle cell were decreased in test group.Conclusion:The results suggest the c-fos antisence olioneucleotide can inhibit the intimal proliferation of the autografted veins.It is a prospective and idea genetic prophylactic therapy to intimal hyperplasia.

Expression of 5-LOX gene and its inhibitory effect in human pancreatic cancer cell

Objective:To detect the expression of 5-LOX and observe its inhibitory effect in humane pancreatic cancer cell.Methods:5-LOX mRNA and protein expression were analyzed by RT-PCR and immunocytochemical staining as well as images analyzing system,and the effect of MK886 was tested.Results:MK886 effectively down-regulated the expression of 5-LOX mRNA.Immunocytochemical staining and images analysis confirmed that the expression 5-LOX protein in cell treated with MK886 was lower than untreated control cell.Conclusion:Inhibitor of 5-LOX can effectively prevent the target mRNA into protein product,thus block or weaken the expression of 5-LOX.

The relationship between TGF-?_1 mRNA and acute rejection of small intestinal allografts in rats

Objective:To investigate the relationship between TGF-? 1mRNA and acute rejection of small intestinal allografts in rats.Methods:Rats were divided into two groups.Group Ⅰ:Wistar→SD Small intestinal transplantation,Group Ⅱ:Wistar→SD Small intestinal transplantation+CsA.The pathologic changes of allografts at 3 rdday,5 thday,7 thday after operation investigated and the transcription of IL-2,IFN- ?,and TGF-? 1mRNA was detected by semi-quantity RT-PCR.Results:Pathologic change:There was acute rejection at 3 rdday,and became severe at 7 thday in group Ⅰ,and there was only slight rejection at 7 thday in groupⅡ.TIFN-?、IL-2mRNA obviously increased after operation in groupⅠ.But they increased only a little in group Ⅱ.It had statistical significance between the two groups(P

Detection of alpha-fetoprotein messenger RNA in peripheral blood of patients with hepatocellular carcinoma

Objective:To determine whether alpha-fetoprotein(AFP) messenger RNA(mRNA),a specifically hepatocyte-expressed gene,could be a marker of metastasis in hepatocellular carcinoma. Methods:Peripheral blood samples were obtained from 30 patients with hepatocellular carcinoma (HCC).A sensitive reverse transcription polymerase chain reaction( RT-PCR) technique was used to detect the human AFP mRNA in the blood.Results:Of the 30 HCC patients,14(46.7%) patients had positive AFP mRNA.The presence of AFP mRNA correlated with the TNM clinical stages,the size of tumor,extrahepatic metastasis or with portal vein carcinothrombosis(P0.05).Conclusion:The positive of AFP mRNA in the peripheral blood can indicate the presence of hematogenous metastasis in patients with HCC.

Relationship between apoptosis index,expression of P53 and radiosensitivity of rectal carcinoma

Objective:To investigate the correlation between the levels of the spontaneous apoptosis index(SAI),expressions of P53 and the radiosensitivity of rectal carcinoma.Methods:Thirty-six patients with rectal carcinoma who received preoperative radiotherapy were registered in this study.Specimens were obtained by biopsy before irradiation and after operation.The apoptotic index(AI) of each tissue specimen was evaluated by terminal deoxynucleotidyl transferase-mediated deoxyuridine triphophate nick-labling (TUNEL) assay and expressed as a percentage,Immunohistochemical evaluation for P53 and the radiosensitivity of the tumor was also peformed.Results:Consistent low levels of spontaneous apoptosis were observed in all specimens evaluated,the mean AI of spontaneous apoptosis was 0.98%+/-0.31%;There was an inverse correlation between SAI and the expression of P53 protein(r=0.72,P0.98% was significantly higher than that of SAI

A three-dimensional model of angiogenesis in vitro

Objective:To develop an experimental three-dimensional model by ECV304 cells(human umbilical vein endothelial cell line) for investigating the mechanisms of angiogenesis in vitro.Methods:ECV304 cells were seeded onto three-dimensional collagen gels made of rat-tail collagen.When the endothelial cells were cultured and grown to near confluence,treated with bFGF for 3 to 12 days,and then assessed with inverted phase contrast microscope.Results:The endothelial cells migrated into the gels,formed complex networks by cell cords at different levels through the bottom view,and sprouted capillary-like structures through the side view.Conclusion:ECV304 cells are capable of expressing some early events of angiogenesis in the three-dimensional collagen gels:proliferating,migrating and sprouting and so on.It should be useful for studying angiogenesis in vitro.

The expression of NF-?Bp65 and bcl-x_ L gene in pancreatic carcinoma and their significance

Objective:To study the expression of NF-?Bp65 and bcl-x L gene in pancreatic carcinoma and their clinical significance.Methods:The expressions of NF-?Bp65 and bcl-x L protein in 52 pancreatic carcinoma were studied by immunohistochemistry,and their relationship to clinical pathology were analyzed.Results:The positive rates of NF-?Bp65 protein and bcl-x L protein in pancreatic carcinoma were 63.5% and 73.1% respectively,and which were higher than those of normal pancreatic tissues.The expression of NF-?Bp65 was related to tumor diameter,TNM stage and lymph node metastasis,and had no relation with the extent of differentiation and type of pathology.There was positive correlation between NF-? Bp65 and bcl-x L expression.Conclusion:NF-? Bp65 may be an important oncoprotein,and may have positive regulation on bcl-x L,then promote tumorgenesis.

Continuous observation to rejection following small bowel transplantation in inbred rats

Objective: To explore the values for rejection research of the models through observing rejection rulers following small bowel transplantation(SBT)in Inbred strain rats.Mothods: Heterotopic SBT models were established with Inbred strain F344/RT11 and BN/RT1n by microsurgical technique according to Monchik method,including Isotransplantation(F344→F344,n=8)and allotransplantation(BN→F344, n=8).Results: 1.The mean living time was over 30 days in IT x group ,which was 12 days in AT x group.2.The general conditions have no significant difference on various time points of postoperative day(POD)in IT x group,while pathological features of grafts were concided with diagnosis of nonspecific slight inflammation on POD3 and were the same as the normal grafts on POD 0?5?7 and 9.3.Significant difference of IL-2R? chain and ?-INF mRNA transcription was earlier than pathological feature appearance of rejection.4.The general conditions and pathological features of grafts were concided with diagnosis of mild,moderate and severe actute rejection in AT x group on POD 5?7and 9 respectively.Conclusion:Inbred strain BN to F344 combination is fitter for research on rejection mechanism than F344 to Wistar combination and Outbred strain Wistar to SD combination.

Study on the expression of neuron-specific enolase,chromogranin and synaptophysin in functional islet cell tumors

Objective:To study the expression and clinical significance of neuron-specific enolase,chromogranin and synaptophysin in islet cell tumors.Methods:Twenty-one paraffin-embedded specimens with islet tumors and 3 cases cholangiocarcinoma and tissue nearby the tumor were recruited from Jan 1988 to Jun 2001.The S-P method was used for the detection of chromogranin,synaptophysin and neuron-specific enolase.Results:There were positive expressions of neuron-specific enolase,chromogranin and synaptophysin in 19,15 and 18 cases respectively,while negative results were observed in 3 cases of cholangiocarcinoma and the para-cancer tissue.Conclusion:CG,SYN and NSE may be specific tumor markers of islet cell tumors and combined detection can help to enhance the sensitivity.

Analysis of diagnosis and treatment about tuberculosis of thyroid

Objective:To expore early diagnosis and treatment of tuberculosis of thyroid.Methods:One patient with tuberculosis of thyroid who was misdiagnosed for 7 months was reported,a retrospective analysis of 87 patients with tuberculosis of thyroid gland diagnosed by needle biopsy and postoperative pathologic examination.Results:15 cases were diagnosed by needle blopsy,others by postoperative pathologic examination,most of them were misdiagnosed as thyroid carcinoma,thyroid adenoma,the fewer were misdiagnosed as endemic goiter,nodular goiter,hyperthyroidism.Conclusion:Thyroid node accompanied by fever and pulmonary tuberculosis must be considered of tuberculosis of thyroid,needle biopsy is a convenient and effective method for diagnosis.
