Relationship between serum amyloid A and Insulin resistance in patients with metabolic syndrome

Objective To probe the relationship between serum amyloid A and insulin resistance in patient with metabolic syndrome.Methods Parameters of body height,body weight,waistline,SBP,DBP,TG,TC,FPG,and HDL were measured in the group of 40 patients with metabolic syndrome while 30 healthy people were referred as control group.Results The level of serum SAA,HOMA,and IR were significantly higher than those of the control group(P<0.01),and the other parameters were also indicated significantly difference(except the parameters of age,height,TC,and LDL-C).In patients of metabolic syndrome,the SSA had a positive correlation with body weight,waistline systolic pressure LDL-C,FINS,HOMA and IR,while the SSA had a negative correlation with HDL-C.Conclusion SAA is closely associated with insulin resistance,and it may serve as a marker in patients with metabolic syndrome

Wet Smear Compared With Gram Stain Diagnosis of Bacterial Vaginosis in Asymptomatic Pregnant Women

Objective To compare wet smear and Gram stain diagnoses of bacterial vaginosis among asymptomatic pregnant women.Methods Between September 1,2006 and January 31,2008,asymptomatic women who initiated prenatal care in our obstetric clinics were invited to participate.Clinical diagnosis that required two of three positive criteria for bacterial vaginosis(vaginal pH,whiff test,and clue cells on wet smear)was compared with Gram stain diagnosis(Bacterial vaginosis score 7-10 by Nugent criteria).Results Population characterstics included an average matemal age of(27.3±6.6)years,26 nulliparas.The mean gestational age at entry was(15.6±7.6)weeks.Twenty-seven percent(18/67)of the study population was diagnosed with bacterial vaginosis by definitive Gram stain.Using Gram stain diagnosis of bacterial vaginosis as the standard,clinical diagnosis had sensitivity of 56%(95% confidence interval[CI]32%,78%),aspecificity of 96%(95% contidence interval[CI]90%,100%),a positive predictive value of 83%,and a negative predictive value of 85%.Conclusion In asymptomatic pregnant women,bacterial vaginosis can be diagnosed reliably by Gram stain.

Investigated the serum α1-antitrypsin concentrations in 1352 petrochemical workers

Objective To observe the effect of atmospheric pollution on workers'α1-antitrypsin concentrationb in petrochemical enterprises.Methods During health examination,the workers were indivded into 3 groups according tO the length of job experience.Ⅰ group(<10 years)included 291 cases,aged 22-35 years old,Ⅱ group(10-20 years)included 634 cases,aged 33-45 years old,and Ⅲgroup(>20 years)included 427 cases,aged 44-55 years old.The normal control were individed int different group according to the status of petrochemical workers.,all the serum samples were detected on automatic biochemical analyzer,α1-antitrypsin levels were analyzed between the different groups.Results In group Ⅰ,the α1-AT concentration was 2.58±0.38 g/L(P>0.05)without significant difference compared with control.In group Ⅱ and Ⅲ,the concentration α1-AT were 2.02±0.43 g/L and 1.72±0.41 g/L respectively with significant differences compared with control.And the significant difference existed among the three groups of petrochemical workers..But the sigficant difference did not exist between different group of control people.Conclusion During the process of oil refining,the plants exhausted the polluted gas of nitrogen dioxide,olefins,alkanes,alcohols,carbonyl and other harmful substances,it influenced theα-AT levels of body.It is a good suggestion to decreas the petrochemical pollution,and to monitor the serum concentration of α-AT content among petrochemical workers,it might to reduce the incidence of lung disease of great significance.

Clinical significance of the detection of serum IL-8 and peripheral blood white blood cell counts in elderly coronary artery disease patients

Objective To explore the relationship between serum IL-8 and peripheral blood white blood cell(WBC)counts and coronary artery disease(CHD)and the pathological changes degree of coronary artery.Methods 296 CHD patients were divided into acute myocardial infarction(AMI)(n=100),unstable angina(UA)(n=100)and stable angina pectoris(SAP)(n=96)groups,the other 100 healthy controls were selected as normal control group.Serum IL-8 level was detected by ELISA.The correlation between Deripheral blood WBC and serum IL-8 level was analyzed.Results Serum IL-8 level in AMI,UAP and SAP groups was higher than of control group(P<0.05);that of AMI and UAP groups was higher than that of SAP group;serum IL-8 level in CHD patients was positively related to peripheral WBC.Conclusion Serum IL-8 level might be the marker of coronary atherosclerosis in the elderly,is closely related to the pathological changes degree of coronary artery,participating in the development of CHD.

Mycoplasma culture and results analysis on drug resistance of infertility patients in Lanzhon

Objective To understand ureaplasma urealyticum(Uu)and mycoplasma hominis(Mh)infection and drug resistance of mycoplasma from infertility patients in Lanzhou,to direct the clinical use of antibacterial drugs.Methods Uu and Mh from 1 278 cases were cultered and tested as well as their drug resistance were analyzed.Results 42.9%infertility patients showed Uu positive,7.0% showed Mh positive,and 9.0% infertility patients were mixed infection of both Uu and Mh.The sensitive antibacterial drugs were josamycin,doxycycline,minocycline hydrochloride,and clarithromycin,which the sensitive rates of both Uu and Mh were>80%,however,the sensitive rates of Uu and Mh against ciprofloxacin were 11.3% and 16.0%,respectively.Conclusion The mycoplasma infection rate was high in infertility patients,and the problem of drug resistance was serious.The results of drug sensitivity analysis should be recognized as a guide for rational drug use.

Bacteria colonized and antibiotic resistance analysis on nongonococcal urethritis in men

Objective To investigate the composition and drug resistance of pathogenic bacteria of nongonococcal urethritis in men,in order to give some references for clinical treatment.Methods The pathogens were cultured by Urethral secretions of nongonococcal urethritis in men and drug susceptibility test followed.Results Coagulase negative staphylococcus and mycoplasma urealytium were major pathogens of nongonococcal urethritis in men;The constituent ratio of meticillin resistance staphylococcus was above 40%.There was significant difference that the constituent ratio of meticillin resistance coagulase negative staphylococcus compared with specimen in the same period.Meticillin susceptible coagulase negative staphylococcus and meticillin susceptible Staphylococcus aureus were sensitive to cephalosporins and quinolones,excluding penicillins.The antimicrobial susceptibility of meticillin resistance staphylococcus was just opposite completely;Mycoplasma urealytium and Mycoplasma hominis were high sensitivity to josamycin、doxycyline and minocycline.Conclusion The colonization bacterium of nongonococcal urethritis in men are primarily consist of coagulase negative staphylococcus and mycoplasma urealytium.It is worthy of further researching correlation between coagulase negative staphylococcus and nongonococcal urethritis,it also suggested that antibiotics on should be rationally choosed on therapy of nongonococcal urethritis.

Active Pseudomonas aeruginosa OprM efflux phenotype and genetic testing

Objective To take the initiative in Huaibei region of Pseudomonas aeruginosa OprM efflux phenotype and the presence of genetic analysis to explore the multi-drug resistant Pseudomonas aeruginosa and the active efflux mechanism.Methods assignment pump inhibitor carbonyl cyano-right-chlorophenyl hydrazone(CCCP)on Pseudomonas aeruginosa ciprofloxacin(CIP) reversal of the sensitivity tests to screen active efflux of Pseudomonas aeruginosa phenotype-positive bacteria;using PCR amplified active efflux phenotype-positive bacteria OprM genes.Results CCCP under the action of 36 Pseudomonas aeruginosa,24 strains of the CIP to improve the sensitivity of four times more active efflux phenotype positive rate was 66.7%(24/36);in OprM gene PCR extension by experiment,there are 16(44.4%,16/36)was amplified 848bp fragment of OprM.Conclusion active efflux phenotype in clinical Pseudomonas aeruginosa in widespread;OprM of Pseudomonas aeruginosa genes in active efflux of the most common bacteria.

The dynamic expression profile of forkhead transcription factor genes in normal Balb/c mice and after bile duct ligation

Objective The present study was initiated to determine the expression profiles of the Fox genes in normal Balb/c mouse liver and their dynamic expression changes during fibrogenesis induced by experimental bile duct ligation(BDL).Methods RT-PCR was employed to detect 18 Fox family members including Foxo1,Foxo3,Foxm1,and Foxl1 in normal mouse liver.After mice were bile-duct-ligated,real time fluorescence quantitative PCR(FQ-PCR)was performed to ananlyze the dynamic mRNA expression changes of 9 inflammation-or proliferation-related Fox family genes.Results All the 18 Fox genes were found to be exDressed in the normal mouse liver and bile duct ligation profoundly influenced the expression of Fox transcriptional factor family genes.The expression of Foxol was significantly reduced by BDL,while FoxoL1 and Foxom1 expression were enhanced in this roodel.Moreover,the expression of Foxo1 and Foxo3 were the highest among the Fox members.Conclusion The Fox family genes-related to inflammation and proliferation were dynamically changed during BDL-induced liver injury,it indicates these genes were involved in the pathogenesis of liver fibrosis.

Establishment and Evaluation the Review Criteria for Sysmex KX21 Blood Cell analyzer

Objective To find a proper way for accurate results of blood counts,and provide reference for the user of 3 cluster blood cell analyzer.Methods Based on the results of blood cell analyzer KX21 and International Consensus Review Rules,set up the criteria for blood cell microscopic examination,1700 blood specimen were detected and the results were analyzed according to the criteria..Statistics analysis were made to evaluate the accordance between warnings of analyzer and manual examination,likewise the reliability of the criteria.Results According to the review rules and slide riview positive criteria[1],the true positive ratio,the false positive ratio,the true negative ratio and the false negative ratio were 11.88%(202/1 700),21.88%(372/1 700),64.29% (1 093/1 700) and 1.94% (33/1 700) respectively..No blast cell and immature cell was missed.Conclusion The smear review criteria suggested by the International Consensus Group could facilitate to establish the review criteria for 3 cluster blood cell analyzer..The review rules used on KX21 blood cell analyzer in this study are suitable for our laboratory.

Identification and molecular analysis on an individual with para-Bombay Phenotype

Objective To identify a case of Para-Bombay phenotype and analyze it's molecular background.Methods Routine serological methods were used to identify the antigens on red cells and antibodies in serum,the weak antigens on red cells were detected by absorption and elution test.ABO substances were determined by agglutination inhibition test in saliva.FUT1 and FUT2 gene were sequenced with molecular methods.Results The para-Bombay was confirmed as Ah-secretion.Two of the three T repeats located at the nucleotides 880-882 were deleted.This deletion causes the shifting of the open-reading frame.The FUT2 genotype was Se357Se357.Conclusion Para-Bombay phenotype is rare.Homozygous genotype of FUT1 allele,hh(nt880-882de1TT)might be the cause of H-antigens deficiency..The wild type of FUT2 allele was consistent with the secretor status.
