The Relationship of Expression of CD_(44)V_6, PCNA to Lymph Node Metastasis in Colorectal Carcinoma

To explore the relationship of expression of metastasis associated protein-CD 44 V 6 and proliferating cell nuclear antigen(PCNA) to lymph node metastasis in colorectal carcinoma,randomly selected patients with colorectal carcinoma who underwent coloproctectomy plus D 2～D 3 lymph node dissection were studied.The 104 patients were divided into:A group (n=48) without lymph node metastasis and B group(n=56) with metastasis.The expressions of CD 44 V 6 and PCNA were assayed by immunohistochemical technique.The results indicated that the 56 cases(75%) of B group were positive for expressions of CD 44 V 6 and PCNA,there was obvious correlation between both the expressions and lymph node metastasis(P

The Malignant Obstruction of the Left Hemicolonic Cancer And its Resection and Anastomosis in I Stage

In this paper,the feasibility and clinical significance of left hemicolectomy colocolostomy in treating the obstruction of the left colonic cancer were discussed.Preoperative preparation,intraoperative and postoperative treatments in 12 patients with the obstruction of the left colonic cancer who received left hemicolectomy colocolostomy were studied retrospectively.Radical resection was performed in all 12 patients.There was only 1 case of incision infection.No fistula of the anastomotic stoma was seen.From the results,left hemicolectomy colocolostomy was feasible in those with permitted situation,enough blood supply of colon without obvious colonic swelling and inflammation.The fistula of the anastomotic stoma was avoidable when the ways of preoperative preparation,intraoperative and postoperative treatments were emphasized.For reducing the economic burden of the patients,duration of hospitalization,avoiding suffering operation twice,especially improving the prognosis,it may be the best choice to resect the left colonic cancer in I stage.

The Protective and Therapeutic Effects of Anisodamine on Acute Lung Injure and Its Molecular Mechanism

To examine the protective action and therapeutic effect of anisodamine on acute lung injure and its molecular mechanism,63 rats were randomly divided into three groups :normal control group(NS),lipopolysaccharide (LPS) group and anisodamine (ADM) group.Rat model of acute lung injure(ALI) was produced by injecting LPS(5mg/kg) into the tail vein.The lung index(LI),WBC count,the percentage of polymorphonuclear leukocyte(PMN) in the bronchoalveolar lavage fluid(BALF) were observed at 0,1,3,5 hours(T 0,T 1,T 2,T 3).The concentration of TNF-? were measured both in BALF and plasma at four time points.The expression of P65 was analyzed by immunohistochmical method five hours after injecting LPS.The results showed that LI of ADM group were significantly lower than that of LPS group at T 2 after injecting LPS(P

Action of Triptolide in the Expressions of IL-5,GM-CSF mRNA in Pulmonary Tissue of Asthmatic Guinea Pig

To research the expressions of IL-5,granulocyte macrophage colony stimulating factor(GM-CSF) in the asthmatic guinea pig pulmonary tissue and influence of triptolide on the expression,30 guinea pigs were divided at random into three groups:the asthma group(n=10),treated group(n=10) and control group(n=10).The expression levels of the IL-5,GM-CSF mRNA were detected by using in situ hybridization method.The results showed that the expressive intensities of IL-5,GM-CSF mRNA in asthma group were 36 38?7 65 and 32 78?9 43 respectively;the treated group were 17 46?7 32 and 18 92?6 38 respectively;control group were 13 82?5 43 and 18 27?7 45 respectively.The expressive intensities of IL-5,GM-CFS mRNA in asthmatic group were obviously higther than those of both treated and control groups,there was significantly difference between two groups(P

Autosecretion of the Fresh Leukemic Cells from Patients with Acute Myeloid Leukemia in Vitro and Apoptosis in the Cells

To investigate the effects of granulocyte macrophage colony stimulating factor (GM-CSF) which was secreted from the fresh leukemic cells of patients with acute myeloid leukemia (AML) and recombinant human GM-CSF(rhGM-CSF) on the apoptosis of the primary incubative AML cells,the level of GM-CSF was measured by means of radioimmunoassay,the DNA ladder was detected by using DNA electrophoresis,and the percentage of apoptotic cells,percentage of expression and the mean fluorescence intensity(MFI) of bcl-2 were respectively assayed by means of flow cytomtric analysis.After the fresh leukemic cells from 5 cases were incubated for 24 hours,GM-CSF in 3 of 5 cases were seen,their percentage of apoptotic cells was less than the other 2 cases,and the MFI of bcl-2 in the 3 of 5 cases was obviously enhanced.After the fresh leukemic cells were treated with cytarabine (Ara-C) 5?g/ml in combination with rhGM-CSF 5?g/ml for 24 hours,the percentage of apoptotic cells was more than the cells treated with Ara-C alone and it upward to (56 4?7 4)%(p

Detection of Telomerase Activity in Gastric Mucosa by Improved TRAP Assay

Telomerase activity in 43 biopsy of gastric mucosa was detected with the modified telomeric repeat amplification protocol(TRAP) assay to elucidate pathogenesis of gastric cancer.The detected positive rates were as follows:87%(20/23) in gastric cancer,28 6%(2/7) in moderate and non-typical gastritis ,0%(0/10 in atrophic and superficial gastritis,respectively.These results suggest that the detection of telomerase activity may play a significant role in the diagnosis of gastric cancer.

Assay of Correlative Oncogene of Testicular Tissues in Cryptorchids

The P53,Rb,nm23,C-berB-2 genes of testicular tissues were assayed with immunohistochemical method in 30 hereditary cryptorchids.The results indicate that nm23 of 4 cases and C-berB-2 of 2 cases were positively stained in 30 cases,and one of the 6 cases was both positively stained in an abdominal undescended testis.The findings suggest that aberrantion of nm23,C-berB-2 genes is associated with testicular neoplasms of retained testis,specifically abdominal undescended testis has trend to variation of correlative oncogenes of testicular tissues.

Assay of the Activities of Human Tumor Necrosis Factor and Natural Killer Cell in Patients with Lung Cancer

To study the changes of activities of tumor necrosis factor(TNF),natural killer cell (NKC) in patients with lung cancer and clarify the influence of clinical factors on the activities,we observed the level of TNF from peripheral blood cells by detective method of whole blood cell-derived TNF,and NKC activity was also observed with DNA-fluorometric assay for cytotoxic effect in thirty-five patients with lung cancer.The results showed that the NK activity in lung cancer patient was seriously inhibited and got worse with disease phase progress,tumor size and the extent of lymph node metastasis,but it was not associated with the pathological type.The extent and time of reduction of NK activity were related to the quality of primary lesion.As for the changes of TNF activity,PBMC (peripheral blood mononuclear cells) were activated under the presence of tumor to secrete a high level of TNF,which was not related to the stage of disease,the tumor diameter,the extent of lymph node metastasis and type of pathology.These results indicate that the cellular immune function in lung cancer patient is seriously impaired,and the function of immunoregulation is in a complicated chaotic status.We consider that the determination of the activities of TNF and NKC may provide a theoretical evidence for comprehensive treatment of patients with lung cancer.

Relationship between Angiotensin Converting Enzyme Gene Polymorphism and Serum Angiotensin Converting Enzyme Activity

To study the relationship between angiotensin converting enzyme(ACE) gene polymorphism and angiotensin converting enzyme activity,the polymerase chain reaction(PCR) was used to detect an insertion/deletion(I/D) polymorphism of ACE gene in its sixteenth intron.The genotype,enzyme coupling spectrophotometry were used to measure serum ACE activity among the three genotypes.Results showed that the Serum ACE activities were (337 6?92 0),(292 7?76 5 )and(243 5?58 9)u/L for genotypes DD,ID and ⅠⅠ respectively.The findings indicate that the ACE gene polymorphism closely associated with serum ACE activity.

Study on the Effect of Residual Uracil DNA Glycosylase upon HCV cDNA

To explore the destructive effect of residual uracil DNA glycosylase on dUTP incorporation to the cDNA PCR products,PAGE and DNA-EIA hybridization technique were applied to detect the efficiency of anti-contamination and the rate of DNA hybridization.In this study,it was found that:⑴Good effect could be achieved with the reaction of 2u of uracil DNA glycosylase(UDG)for 20min at 37℃.⑵UDG could be inactivated at 94℃ for 10min.The hybridization rate of control group of PCR products was 94 18% at room temperature for 65 hours.The hybridization rates of test groups at-20℃ for 65 hours were 87 69%,77 24%,76 83%,respectively.And the inactivation times of anti-contamination groups at 94℃ were“0”min,“10”min and “20”min,respectively;at room temperature for 65 hours,these hybridization rates were 74 77%,72 50%,70 29%,respectively.There were significant differences between control and rest groups(P0 05).Our study suggested that the hybridization rate of test groups kept at-20℃ decreased 19 41%,after 65 hours,the rate decreased 27 45%,but in control group only decreased 5 82% for 65h at room temperature.So that,we conclude that the residual UDP can cause above results.
