Study on the expression and significance of nucleolin in osteosarcoma

ObjectiveTo investigate the expression of nucleolin in osteosarcoma and its clinicalsignificance.MethodsThe expression of nucleolin was evaluated by immunohistochemistry in paraffin-em bedded tissues from 35 cases of osteosarcoma and 15 cases of osteochondroma.ResultsAmong the osteo sarcoma cases, 8.6％ were negative for nucleolin, 20％ weakly positive, 34.3％ moderate positive, 37.1％ strong positive.The positive rate of nucleolin in osteochondroma was 40％, 46.7％, 13.3％, 0％, respectively.The nucleolin expression showed significant difference in the osteosarcoma and osteochondroma(P ＜0.01).Among the conventional ostcosarcoma cases, 6.9％ were negative for nucleolin, 13.8％ weakly positive, 34.5％ moderate positive, 44.8％ strong positive.The positive rate of nucleolin in paros teal osteochondroma was 16.7％, 50％, 33.3％, 0％, respectively.The nucleolin expression showed re markable correlation with pathological and genetical classification (P ＜ 0.05).The positive rate of nucleo 1in in osteoblastic osteochondroma was 5.9％, 5.9％, 29.4％, 58.8％, while in chondroblastic osteochon droma 0％, 33.3％, 33.3％, 33.3％ and 16.7％, 16.7％, 50％, 16.7％ in fibroblastic osteochondro ma, respectively.The positive expressions of nucleolin were almost the same in each subtype of osteosarco ma (P ＞ 0.05).ConclusionsThe nucleolin expression had correlation with property of tumors in bone, and the positive expressions of nucleolin had correlation with pathological and genetic classification of osteo

Generation of human induced pluripotent stem cells from human skin fibroblasts

ObjectiveGeneration of human induced pluripotent stem cells from human skin fibroblasts.MethodsSox2, Klf4, Oct4, c-Myc were transfected into HSF cells with retrovirus, and then HSF cells was reprogrammed to iPS cells.Detecting cells endogenous and exogenous gene, analyzing karyotype,cells alkaline phosphatase staining and immunofluorescence staining, differentiating into teratomas in vivo and embryoid bodies in vitro were performed.ResultsiPS cell morphology was similar to embryonic stem cells (ES).The expression of Nanog, Oct4, Rex1, Sox2 in iPS cells were higher than HSF cells, and Sox2, Klf4, Oct4, c-Myc were silenced for the iPS cells.Exogenous genes were inserted into the nucleus of iPS cells, which was confirmed by 1％ agarose gel electrophoresis.iPS cell karyotype was normal, alkaline phosphatase activity increased, ES cell-specific protein expressed.iPS cells were differentiated into a teratoma in vivo and embryoid bodies in vitro.ConclusionsiPS cells were similar to ES cells, which have pluripotency.

Association of SNP276 in adiponectin gene with complicating different component numbers of metabolic syndrome in population with impaired glucose regulation

ObjectiveTo study the association of SNP276 in adiponectin gene with impaired glucose regulation (IGR) complicating with the different component numbers of metabolism syndrome (MS) in population of IGR in Han people of Shanxi region.MethodsThe study population consisted of 256 subjects with IGR which was composed of 123 subjects with component numbers of MS ＜2 (IGRA) and 133subjects with component numbers of MS≥2 (IGRB),and 128 subjects with normal healthy (Normal) who were Chinese Hans residents and in Shanxi province.Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was applied to test the adiponectin SNP276G/T polymorphism.ResultsThe distributions of genotypes and alleles of SNP276 both displayed significant difference among the three groups ( x2 =16.893,P =0.002 ; x2 =18.149,P =0.000).In the IGRA,the SNP276 non-TT ( GT + GG) genotype was no difference which increased risk of complicating with the different component numbers of MS( P =0.781,P =0.809).In IGRB,the SNP276 non-TT ( GT + GG) genotype was associated with increased risk of complicating with the different component numbers of MS,and after age and sex-adjusted,there was significant difference ( P =0.007,P =0.007).ConclusionsSNP276 in APM1 increased the risk of complicating with the components of MS in population with impaired glucose regulation in Han people of Shanxi region.

Effects of berberine on mRNA expression of 11β-Hydroxysteroid dehydrogenase type Ⅰ in insulin-resistant HepG2 cells

Objective To investigate the effect of berberine on glucose absorption and mRNA expression of 11 β-hydroxysteroid dehydrogenase type 1 ( 11β-HSD1 ) in insulin-resistant HepG2 cell model.Methods HepG2 cells were incubated with high-concentration insulin for 24 hours to build insulin-resistant cell model.In order to evaluate the cells for insulin resistance,the cells were cultured with different concentrations of insulin for 24 hours.The insulin-resistant cells were treated with different concentrations of berberine and insulin for 24 hours and the non insulin-resistant cells were used as a control.The glucose concentration in culture medium was detected by the method of glucose oxidase-peroxidase (GOD-POD).According to the glucose concentrations in blank medium and those in the medium of culturing cells 24 hours later,the rate of absorpting glucose by the cells was calculated.The mRNA expression of 11β-HSD1 in the insulin-resistant cells was detected by the reverse transcription polymerase chain reaction (RT-PCR).Results Incubated with 10-7mol/L insulin for 24 h,the insulin-resistant cell model had been built.The rate of glucose absorption of the model cell treated with high concentration berberine ( 10 μmol/L) was significantly improved [(42.53 ±1.99)％ vs (28.16±1.99)％,t =12.9457,P ＜0.01].High-concentration berberine showed a strong synergy with insulin on glucose absorption of the model cells.As the cells became resistant to insulin,the mRNA expression of 11β-HSD1 increased significantly compared to non insulin-resistant cells( relative expression quantity was (4.60 ±0.96 vs 0.67 ±0.42,t =4.9476,P ＜0.05 ).While the mRNA expression of 11β-HSD1 reduced in the insulin-resistant cells after treated with high-concentration berberine,and the relative expression quantity was not significantly different with non insulin-resistant cells ( 1.12 ±0.35 vs 0.67 ±0.42,P ＞0.05).However,low-concentration ( 1 μmol/L) of berberine had not the same role.Conclusions It is concluded that one of the acting mechanism of berberine improving the insulin sensitivity may be that the mRNA expression of 11β-HSD1 is downregulated in the insulin-resist-ant liver cell model depending on concentration.

Comparison of sufentanil and fentanyl in total intravenous anesthesia for liver resection: A double blind randomized clinical trial

Objective To compare fentanyl and sufentanil in total intravenous anesthesia for liver resection. Methods 100 patients undergoing liver resection were randomly received either sufentanil (group S, n = 50) anesthesia or fentanyl (group C, n = 50). Hemodynamic variables, time to spontaneous eye opening and extubation were recorded. VAS-scale was used to assess postoperative pain. The incidence of nausea & vomiting, agitation and respiratory depression was recorded and compared. Results Patients in group S had a more stable hemodynamic variables, and the incidence of postoperative pain (VAS: group S 4. 2± 1.1, group C 6. 1 ± 1.2), agitation (group S 2 cases, group C 11 cases) and respiratory depression (group S 1 case, group C 5 cases)was lower. Conclusions Both sufentanil and fentanyl were safe and efficacious in total intravenous anesthesia for liver resection. Compared with fentanyl, Sufentanil had higher hemodynamic stability, and lower incidence of postoperative pain, respiratory depression and agitation.

Relationship between the angiotensin converting enzyme 2 gene polymorphism and antihypertensive effect of valsartan

Objective To study the relationship of single nucleotide polymorphism of angiotensin converting enzyme 2 ( ACE2 ) with essential hypertension(EH) and antihypertensive response of valsartan.Methods A total of 120 case EH patients was received valsartan once daily for four weeks.Direct DNA sequencing was performed to detect the signgle nucleotide polymorphisms of ACE2 in 120 EH patients and 60 controls with normal blood pressure.ResultsThe genotyping data indicated that there were significant differences of G allele frequency between male or female EH group and controls( x2 =5.310,4.423,P ＜0.05 ).Treated with valsartan,patients with.G allele were not found to be associated with reduction in blood pressure( P ＞0.05).Conclusions Our data suggest that the ACE2 G8790A signgle nucleotide polymorphisms might be involved in the development of EH.The risk developing hypertension in the people carrying G allele is higher than that in those carrying other allele.Therefore,ACE2 gene might be a sensitive gene associated with EH.

Effects of valsartan on transforming growth factor-β1 ,Smad in an acute lung injury rat model

Objective To observe the effect of Valsartan on a rat model of acute lung injury and the expression of transforming growth factor-β,TGF-β) ,Smad2/3, Smad7. Methods Twenty-four male adult Sprague-Dawley rats were random divided into three groups : The bleomycin (BLM) group, the control group, and the Valsartan group. Each group contained eight rats. The Valsartan group was treated with Valsartan everyday at a dose of 20 mg/kg after a single intratracheal instillation of bleomycin at a dose of 5mg/kg. BLM group was treated with saline instead of Valsartan after an instillation of bleomycin. The control group was treated with saline instead of Valsartan and bleomycin. Each group was killed at the 7th day after instillation. The lung tissues were harvested for H. E. stain, the immunohistochemistry was used to detect the expressions of TGF-β1, Smad2/3 ,and Smad7. Results The degree of alveolitis in the Valsartan group was ameliorated, compared with those in BLM group (P ＜0. 01). The expressions of TGF-β1 and Smad2/3 in lung tissue of the Valsartan group were significantly lower than that of BLM group(P ＜0. 01). The expressions of Smad7 in lung tissue of the Valsartan group were significantly higher than that of BLM group (0.23 ±0. 02 vs0. 36 ±0.03, P ＜0.01). Conclusions Valsartan could alleviate acute lung injury in rats, which probably be due to the expression decrease of TGF-β1 and Smad2/3 and the expression increase of Smad7 in lung tissues.

Association of EPHX2 polymorphism G860A with hypertension in middle ages

Objective To investigate the association between the EPHX2 polymorphism G860A and the risk of hypertension in mild ages. Methods In a hospital based case-control study, one common polymorphism C860A in EPHX2 gene in a case-control study of 100 hypertension and 300 age- and sex frequency - matched disease-free controls were genotyped in a Southern Chinese population. SAS 9. 13 was used toanalysis the polymorphism and hypertension risk. Results Genotype frequencies of EPHX2 G860Alocus between the cases and the controls were significantly different (P =0. 01). Compared with the most common 860GG genotype, the 860GA heterozygote had an increased risk of hypertension (adjusted OR =1.98; 95%CI = 1.19 -3.51), the AA homozygote had a further increased risk of hypertension (adjusted OR =2. 84; 95%CI = 1.01 -6. 11). There was a significant trend for an allele dose effect on risk of hypertension (P trend = 0. 03). Conclusions EPHX2 polymorphism G860A is associated with an increased risk of hypertension, and the 860A variant may be a marker for susceptibility to hypertension.

Evaluation on adequacy of hemodialysis by on-line clearance monitoring and Kt/V of Correlation Analysis

ObjectiveTo evaluate the adequacy of hemodialysis by online clearance monitoring (OCM) and Kt/V of correlation analysis.MethodsThe Kt/V values of 48 maintenance hemodialysis patients in the Department of Nephrology of General Hospital of PLA were accessed by both OCM of Germany BeiLang Dialog + hemodialysis machine and single-pool urea kinetic model.The Kt/V calculation value was also checked by the urea dynamics - room variable volume model.The results generated at different time point were compared,including at the real time,before and after appearing through blood by On - line Clearance Monitoring,and correlation analysis of Kt/V value was performed.ResultsThere was no significant difference on evaluating the level of Kt/v by these two methods.The relationship between the results of the two methods was significantly positive related.(r=0.539,P ＜ 0.01).ConclusionsDuring hemodialysis,the OCM option provides an accurate tool for continuous on-line monitoring of urea clearance.

A study on change of serum levels of HMGB1 and its significance in patients with acute pancreatitis

ObjectiveThe study was performed to observe the change of serum levels of high mobility group protein box 1 (HMGB1) in patients with acute pancreatitis (A.P) and to determine its relationship to the severity of AP in order to reveal its clinical significance.MethodsThe serum levels of HMGB1were measured in 50 patients with AP including 24 cases of severe acute pancreatitis (SAP) and 26 cases of mild acute pancreatitis (MAP) in fnst three days after admission.The serum levels of HMGB1 were also detected in 20 healthy volunteers.Furthermore,clinical data of patients was collected and APACHE Ⅱscores were calculated at the time when serum HMGB1 levels were detected to assess the severity of the disease.The 28 day mortality of AP patients was also observed.Then,the relationship between serum HMGB1levels and the severity of AP was analyzed.ConclusionsSerum levels of HMGB1 in patients with AP were significantly positively correlated with APACHE Ⅱ scores.Serum levels of HMGB1 in patients with AP were significantly positively correlated with the 28 day mortality.HMGB1 could be used as an effective indicator to evaluate severity of AP.
