A CHEMICAL METHOD FOR THE DETERMINATION OF VITAMIN D IN FORTIFIED FOODS

Abstract
The colorimetric method for the determination of vitamin D in fortified multivitamin milk powder containing 200-3000 IU vitamin D/100g, and oil concentrate and pellet containing 200-5000 IU vitamin D/g is described. The fortified samples are extracted by ethyl ether petroleum ether at ratio of 1:1, then the solvent is evaporated with the rotatory concentrator. The residues are saponified with KOH (50%). The cocentrated samples are saponified directly with KOH. The cholesterol in the saponified samples are eliminated by digitonin precipitation. Other interferences from other fat-soluble vitamins, additives in the formulas and biologically inactive isomers of vitamin D are eliminated by column chromatography on phosphate treated alumina, polyethylene-glycol-600-chromosorb W and fuller's earth and a maleic anhydride addition reaction. After color development, determine the absor-bance at both 500 and 550nm wave lengths. The mean recoveries of vitamin D added to fortified food, milk powder and concentrates, expressed as percentage of the known amount of vitamin D added were 106%; 85.8%; 97.2%-101% respectively.
