Abstract:
Objective: To study the chemical constituents from the herb of Dendrobium aurantiacum var. denneanum. Methods: The chemical constituents were purified by column chromatography over silica gel, macroporus resin AB-8, Sephadex LH-20, and preparative HPLC, etc. Their structures were identified by spectroscopic data analysis, and their inhibitory activity on tumor cells growth was investigated by MTT method. Results: Nine compounds were isolated from D. aurantiacum var. denneanum and identified as (-)-(7R,7'R,8R,8'S)-4,4'-dihydroxy-3,3',5,5'-tetramethoxy-7,9'-epoxylignan-7',9-lactone (1), (-)-syringaresinol (2), (-)-secoisolariciresinol-4-O-β-D-glucopyranoside (3), moscatin (4), gigantol (5), moscatilin (6), (+)-naringenin (7), melilotoside methyl ester (8), and vanillic acid (9). Among them, compound 1 was a new compound and showed the different extent of cytotoxic activities against A549, HepG2, and A2780 tumor cells, with IC50 values of 5.49, 51.2, and 119.3 μmol/L, respectively. Conclusion: Compound 1 is a new lignane with the evident cytotoxic activity and named dendroresinol.
