Mechanistic analysis of Astragali Radix in treatment of nephrotic syndrome using network pharmacology

Abstract:
Objective: To explore the molecular mechanism of Astragali Radix in the treatment of nephrotic syndrome (NS) by the network pharmacology. Methods: The main active ingredients of Astragali Radix were obtained through TCMSP and literature search. PharmMapper, Gene Cards and CoolGeN were used to predict and screen the targets of Astragali Radix. The protein interaction network was constructed using the String database and Cytoscape software. The relation between the main active ingredients and targets was validated by Systems Dock Web Site. The GO and KEGG pathways involved in the targets were analyzed by DAVID. The ingredients-targets-pathways-diseases network was constructed by Cytoscape software. Results: The results showed that 25 active components and 47 targets of Astragali Radix were involved. The network results showed that the process of oxidative stress, podocyte apoptosis, lipid metabolism were mainly involved, which played a role in the treatment of nephrotic syndrome by adjusting the Ras, Rap1, MAPK, PI3K-Akt, AGE-RAGE and other signal pathways. Conclusion: This research method initially revealed that the active ingredients of flavonoids and saponins in Astragali Radix are the material basis for the treatment of NS. Its mechanism of action involved anti-oxidative stress, anti-apoptosis and regulation of lipid metabolism.
