Abstract:
Objective To search for the optimal fermentation conditions for producing recombinant Ganoderma lucidum manganese peroxidase (rGlMnP) by Pichia pastoris. Methods Based on the single factorial experiment, an optimum fermentation condition for rGlMnP production by recombinant Pichia pastoris was obtained through a central composite design (CCD) and response surface methodology (RSM) in the present work. The effects of initial pH, temperature, concentrations of methanol and heme on the enzyme activity of rGlMnP produced by Pichia pastoris were explored. The interactive effects and the optimal range levels of the 4 factors were studied and the validity of the model was also verified. Results The results showed that initial pH, temperature, the concentrations of methanol and heme had significant effects on the enzymic activity. The optimum conditions for enzymic activity of rGlMnP produced by Pichia pastoris were as follows; initial pH 5. 5, temperature 30 °C, concentration of methanol 1. 5% and concentration of heme 1. 0 mmol/L. Conclusion This study has laid a foundation for producing rGlMnP by Pichia pastoris in industrial scale.
