An experimental study of retinoblastoma planting in the rabbit eyes

Abstract:
Objective To establish the animal model of retinoblastoma in the rabbit eye, in order to extend the experimental clinical ophthalmic therapy on the tumor of animal model. Design Experimental study. Participants 16 eyes from 8 rabbits, the tumor cell line is HXO-Rb44. Method 0.1ml of the cell suspension with the concentration of (1.17?07)/ml were injected into the choroid of rabbits after general anesthesia; exam and record the ocular fundus of the rabbits after 1, 3, 7, 14 days of implantations, and enucleate the rabbits eyeballs for pathologic examination and tumor cell identification at 14th day. Main Outcome Measures The quantity and the type of neoplasm, verified by pathologic examination with H-E stain and immune chemical stain. Results Neoplasm was seen growing in the rabbit eyes one week later after implantation, in shape of lump or widespread; the survival rate is 7/16. The tumor cells grow actively with evident interval regions of growing and necrosis. The tumor cells have undifferentiating features: bigger and deeply stained nucleus, less cell plasma, disorder nucleus and plasma ratio, dense chromatin, morphology very different and many mitosis. In rabbit Rb model it is testified that the growth mode, differentiation type and cell characters are as same as those in human Rb with H-E and immune chemical stain. Conclusion Cell line HXO-Rb44 can be alive and form tumor in the rabbit eyes. Rabbit may be an ideal animal for Rb model.
