Abstract:
Diabetic cardiomyopathy(DCM) is a critical phenotype of diabetic mellitus(DM)-specific cardiovascular complications, and mitochondrial dysfunction plays a central regulatory role in the pathological progression of DCM.Among the key factors, mitochondrial transcription factor A(TFAM) is essential for maintaining the stability and integrity of mitochondrial DNA(mtDNA),thereby playing a pivotal role in myocardial energy metabolic homeostasis.Notably, nuclear respiratory factor1(NRF1),as an upstream regulator of TFAM,modulates the expression of genes involved in mitochondrial biogenesis and oxidative phosphorylation, forming the NRF1-TFAM cascade signaling axis. This signaling axis is the core regulatory pathway to maintain mtDNA stability and energy metabolism, and its dysfunction is closely related to the occurrence and development of mitochondrial dysfunction. This article systematically reviews the research progress of the regulation mechanisms of NRF1-TFAM signaling axis on mtDNA stability and its potential role in DCM,with the aim of providing a theoretical basis for the identification of early biomarkers and the development of targeted therapeutic strategies for DCM.
