Abstract:
Streptococcus pneumoniaeis a major pathogen causing pneumococcal infections, and pneumococcal diseases remain one of the persistent challenges in global public health.Vaccination plays a critical role in the prevention of such infections.Measurement of vaccine-induced serum pathogen-specific immunoglobulin G(IgG) levels serves as a key approach to evaluate vaccine immunogenicity and efficacy. As an advanced technical platform, the Luminex-based direct immunoassay(dLIA) exhibits unique advantages including high efficiency and rapidity in the quantitative detection of serum-specific IgG antibodies against pneumococcal vaccines. This review systematically describes the principle of dLIA technology and its application in the detection of serum-specific IgG antibodies induced by pneumococcal vaccines. The advantages of this technique compared with conventional enzyme-linked immunosorbent assay(ELISA) are also compared and analyzed. In addition, the technical optimization strategies, current challenges, and future development prospects are discussed,in order to provide comprehensive reference for the research and improvement of quality assessment techniques for the clinical efficacy of pneumococcal vaccines.
