Abstract:
Objective To systematically evaluate the protective effects of different viral protectants on the titers of respiratory syncytial virus expressing enhanced green fluorescent protein(RSV-EGFP) under various stress conditions,in order to screen for the optimal preservation protocol. Methods RSV-EGFP samples were prepared with 10% trehalose,15% sucrose,0.1%human serum albumin(HSA),10% fetal bovine serum(FBS),10% DMSO,and 2% FBS,respectively.Their stability was assessed under repeated freeze-thaw cycles(1-7 times),short-term storage(7 and 14 days) at different temperatures(37 ℃,4 ℃),and long-term storage(6 months) at-80 ℃.Viral titers were determined using an EGFP-based reporter gene assay and calculated by the Reed-Muench method. Results During repeated freeze-thaw cycles and short-term storage at4 ℃,15% sucrose exhibited the best protective effect, with a titer drop of < 1.00 lg after freeze-thaw and only a decrease of0.74 lg after 14 days of storage at 4 ℃(P = 0.887 8),demonstrating significantly superior stability compared to 10% trehalose and 10% DMSO.When stored at 37 ℃,15% sucrose had a certain delaying effect on the decline rate of virus titer, but none of the three protective agents showed significant protective effect on RSV-EGFP virus within 7 days(P < 0.000 1);For longterm storage at-80 ℃ for 6 months, the 10% DMSO group showed no significant fluctuation in viral titer within 0-3 months(P > 0.999 9),with only a 0.91 lg reduction at 6 months, which was lower than the 2.38 lg reduction for 10% trehalose and 2.05 lg reduction for 15% sucrose.Protectants containing FBS or HSA showed unsatisfactory protective effects under all tested conditions. Conclusion The selection of RSV preservation agents should be tailored to storage conditions:15%sucrose is suitable for repeated freeze-thaw cycles and short-term storage at 4 ℃,whereas 10% DMSO is the optimal choice for long-term cryopreservation at-80 ℃.This study provides key data support and practical guidance for the stable preservation of RSV in related experiments.
