Abstract:
Influenza poses a significant global health threat due to its high morbidity and mortality rates.Given the rapid mutation rate of influenza viruses, the WHO recommends annual vaccination for optimal protection.Egg-based vaccine production has long been the predominant method for influenza vaccines. However, traditional egg-derived vaccines have limitations, including potential reductions in vaccine efficacy caused by egg-adaptation mutations affecting viral antigenicity.Consequently, the WHO advises considering appropriate cell-based influenza vaccines as alternatives or supplements to eggbased vaccines.Among available cell substrates, MDCK cells are the most widely utilized for influenza vaccine production.MDCK cells support high-yield propagation of diverse influenza viruses, including avian, swine, and equine strains. This paper reviews the research progress and challenges associated with MDCK cell-based influenza vaccines, encompassing the discovery and application of MDCK cells, cell culture platforms, the immunogenicity and clinical safety of cell-based influenza vaccines.
