Abstract:
SAMHD1 protein was first identified as a crucial intrinsic antiviral factor in 2011 and is also associated with autoimmune diseases such as Aicardi-Goutières syndrome(AGS) and familial chilblain lupus.In recent years, it has become a focal point of research in the field of molecular biology.SAMHD1 is primarily expressed in cells such as dendritic cells and macrophages, which prevents retroviral replication by decreasing intracellular deoxyribonucleoside triphosphate(dNTP)levels.The Vpx protein of human immunodeficiency virus(HIV)-2/simian immunodeficiency virus(SIV) antagonizes SAMHD1 function.Recent studies have also indicated that the activity of SAMHD1 is influenced by multiple factors and can regulate LINE-1(long interspersed element 1, L1) activity in tumor cells.This paper comprehensively analyzes the complex antiviral mechanisms of the SAMHD1 protein, as well as its mechanism against LINE-1, and also covers the latest understanding of its biological functions and structural aspects.
