Abstract:
ObjectiveTo investigate the effects of overexpression of insulin-like growth factor 2 mRNA-binding protein 1(IGF2BP1) on the malignant biological characteristics of diffuse large B-cell lymphoma(DLBCL) SU-DHL-8 cells, in order to provide experimental basis for finding new prognostic markers and therapeutic targets for DLBCL.MethodsUsing the Gene Expre-ssion Profiling Interactive Analysis 2.0(GEPIA2) platform, the transcriptomic data from DLBCL tumor samples in The Cancer Genome Atlas(TCGA) and normal lymphoid tissue samples from the Genotype-Tissue Expression(GTEx) database were integrated.After standardizing gene expression levels, the expression differences of IGF2BP1 between DLBCL tumor tissues and normal lymphoid tissues were compared.Additionally, leveraging TCGA-DLBC cohort survival data from GEPIA2,high-and low-expression groups were defined based on the median IGF2BP1 expression level to analyze its correlation with DLBCL patient prognosis.In experiments in vitro, a DLBCL cell model overexpressing IGF2BP1 was constructed, and the overex-pression efficiency was verified via real-time quantitative PCR(RT-qPCR) and Western blot.The effects of IGF2BP1 overex-pression on cell proliferation, cell invasion capacity and apoptosis levels were detected by CCK-8 assay, Transwell chamber test and flow cytometry, separately.ResultsAnalysis using the GEPIA2 platform revealed that IGF2BP1 was significantly overexpressed in DLBCL tumor tissues(P=2.22 × 10~(-16)).Survival analysis based on the TCGA-DLBC cohort data demon-strated no significant correlation between IGF2BP1 expression levels and overall survival in DLBCL patients(P=0.58).In vitro experimental results showed that compared with the control group, the overexpressed group exhibited significantly higher IGF2BP1 mRNA levels(t=13.29, P<0.01) and IGF2BP1 protein levels(t=5.700, P<0.01).Overexpression of IGF2BP1 markedly enhanced the proliferative activity(t=7.269, P<0.01) and invasive capacity(t=5.623, P<0.05)of SU-DHL-8 cells, and simultaneously inhibited apoptosis in the cells(t=7.799, P<0.05).ConclusionOverexpression of IGF2BP1 can enhance the proliferation and invasion capacity of DLBCL cells while inhibiting their apoptosis, suggesting that IGF2BP1 may play a oncogene role in the malignant progression of DLBCL and could serve as a potential therapeutic target.
