Abstract:
The conventional treatment strategies offer limited efficacy in advanced or recurrent cases of urothelial carcinoma.Antibody-drug conjugates(ADCs), as a novel type of targeted therapy, deliver cytotoxic agents directly to tumor cells with both high potency and specificity, making them an important tool in precision oncology.This review systematically summarizes the structural components of ADCs and their antitumor mechanisms, including targeted antigen internalization and bystander effect.It also highlights key clinical research findings of representative ADCs, such as nectin cell adhesion molecule 4, trophoblast cell surface antigen 2, and human epidermal growth factor receptor 2.Furthermore, the article addresses the current challenges in the application of ADCs, such as drug resistance, toxicity management, and high cost, while exploring the development of next-generation ADCs and their potential in combination with immunotherapy.Based on current evidence, this review aims to offer valuable insights to support the clinical application and future research of ADCs in the treatment of urothelial carcinoma.
