Abstract:
ObjectiveTo investigate the immunomodulatory effect of mung bean extract(MBE) on macrophage RAW264.7 and its possible mechanism, so as to provide a basis for elucidating the mechanism of MBE in improving immunity and its application.MethodsMBE was extracted by ethanol, and the composition of MBE was analyzed by ultra-performance liquid chromatography tandem quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS). RAW264.7 cells were treated with different concentrations(15. 62, 31. 25, 62. 5, 125, 250, 500, and 1 000 ??g/mL) of MBE, and the cell viability was detected by CCK-8 method. The blank control group, MBE group(62. 5, 125, 250, and 500 ??g/mL) and LPS positive drug group were set up respectively. Real time quantitative polymerase chain reaction(RT-qPCR) was used to determine the mRNA expression levels of interleukin-1β(IL-1β) and tumor necrosis factor-α(TNF-α). The protein expression levels of p65, p38, p-p38,extracellular signal-regulated kinase(ERK), p-ERK, c-Jun N-terminal kinase(JNK) and p-JNK were detected by Western blot. The 250 ??g/mL MBE group was added with inhibitors of p38, JNK, ERK and nuclear factor kappa-B(NF-κB) for NF-κBmitogen-activated protein kinases(NF-κB/MAPKs) signaling pathway verification experiment.ResultsWhen the concentration of MBE was 62. 5, 125, 250, and 500 ??g/mL, it was not toxic to cells. Different concentrations of MBE could significantly up-regulate the mRNA levels of IL-1β and TNF-α in RAW264.7 cells, and increase the phagocytosis rate of macrophages. The expression of p65, p-p38, p-ERK and p-JNK proteins was promoted by MBE at 125, 250, and 500 ??g/mL. After the addition of p38, JNK, ERK and NF-κB inhibitors, the promoting effect of MBE on macrophage phagocytosis and the expression levels of IL-1β and TNF-α were down-regulated.ConclusionMBE can activate macrophages and regulate immune function through NF-κB/MAPKs signaling pathway.
