Abstract:
AIM:This study aims to investigate the therapeutic effects of Xiaoyao pill(XYP)on ulcerative colitis(UC)in mice and its influence on nicotinamide adenine dinucleotide phosphate(NADPH)oxidases through 5-hy-droxytryptamine(5-HT).METHODS:Mouse models of liver-depression and spleen-deficiency type UC were created using the 2,4,6-trinitrobenzene sulfonic acid-ethanol enema method in conjunction with restraint stress and an improper diet.A total of 30 mice were randomly divided into 5 groups:control group,model group,model+XYP(0.15 mg/kg)group,model+XYP(0.3 mg/kg)group,and model+XYP(0.6 mg/kg)group,with 6 mice in each group.The mice in the latter 3 groups received XYP suspension at concentrations of 0.15,0.3 and 0.6 kg/L,respectively,via gavage once daily for 1 week.Additionally,24 mice were divided into 4 groups:control group,model group,model+XYP group(where mice received XYP at 0.6 kg/L via gavage after modeling),and model+XYP+5-HT group(where mice received XYP at 0.6 kg/L via ga-vage and were administered 1.0 mg/kg of 5-HT by enema on experimental days 1 and 4),with 6 mice in each group.Patho-logical changes of the colon tissues were assessed using hematoxylin-eosin staining.Colon length,disease activity index,body weight,and myeloperoxidase activity were also measured.The levels of inflammatory cytokines and 5-HT were quan-tified using ELISA.The levels of reactive oxygen species were determined through dihydroethidium staining,while oxida-tive stress indexes were measured using colorimetric methods.Protein expression of NADPH oxidases was analyzed by Western blot.RESULTS:Treatment with XYP significantly attenuated pathological damage in a concentration-dependent manner,reduced inflammation,and decreased oxidative stress in the colon tissues of UC mice(P<0.05).Moreover,treatment with XYP resulted in a decrease in the expression of 5-HT and NADPH oxidases(P<0.05).Following the ad-ministration of high-concentration XYP via gavage combined with 5-HT enema,there was a notable increase in NADPH oxi-dase expression(P<0.05),while pathological damage in colon tissues,inflammatory response and oxidative stress were exacerbated compared with treatment with high concentration of XYP alone(P<0.05).CONCLUSION:Treatment with XYP may modulate NADPH oxidase signaling to alleviate inflammation and oxidative stress in UC mice through the path-way involving 5-HT.
