Abstract:
Objective:To investigate effect of plumbagin(PLB)on angiogenesis in retina of diabetes retinopathy(DR)rats by regulating stromal cell-derived factor-1(SDF-1)/chemokine receptor 4(CXCR4)signaling pathway.Methods:Rats were randomly grouped into normal control group(NC),model group(DR),low,medium and high doses plumbagin groups(PLB-L,M,H),Qiming granules group(positive control).Retinal angiogenesis of rats in each group was observed by fluorescence fundus angiogra-phy;HE staining was applied to observe pathological changes of retina;serum levels of VEGF,Ang-1,IL-1β,IL-6 and TNF-α were detected by ELISA;oxidative stress level in retina was detected by micro-method kit;Western blot was applied to detect expressions of SDF-1 and CXCR4 proteins in retina.Results:Compared with NC group,retinal neovascularization increased in DR group,with obvious fluorescein leakage,serious retinal structure damage,thickening of optic nerve fiber layer with edema,and obvious pathologi-cal damage,serum levels of VEGF,Ang-1,IL-1β,IL-6,TNF-α,MDA contents in retinal tissue,SDF-1 and CXCR4 proteins expressions increased obviously,SOD activity decreased obviously(P<0.05);compared with DR group,retinal neovascularization and fluorescence leakage in PLB group decreased,and pathological damage improved obviously,serum levels of VEGF,Ang-1,IL-1β,IL-6,TNF-α,MDA content in retinal tissue,SDF-1 and CXCR4 proteins expressions decreased obviously,SOD activity increased obviously(P<0.05);there was no statistical difference in the above indexes between Qiming Granule group and PLB-H group(P>0.05).Conclusion:PLB can block expression of SDF-1/CXCR4 signaling pathway,alleviate inflammation,oxidative stress and pathological damage in DR rats,and reduce retinal neovascularization.
