Effect of hyperbaric oxygen on the expression of granzyme-b in neonatal rats with hypoxia-ischemia brain damage

Abstract:
Objective To investigate effects of hyperbaric oxygen (HBO) on the expression of granzyme-b after hypoxia-ischemia brain damage (HIBD) in neonatal rats. Methods One hundred and ten rats aged 7 days were randomly divided into the sham group (n = 10), the HIBD group (n =50) and the HBO group (n =50). The HIBD model was developed by the Rice method. Samples were obtained following injury, and changes in the expression of granzyme-b were measured by immunohistochemical assay and apoptosis of injured brain cells were detected with Epics XL flow cytometer. Results Six hours after hypoxia - ischemia, the expression of granzyme-b and cell apoptosis rate in the injured brain tissue in the HIBD and HBO groups increased markedly, peaked 1d after hypoxia ischemia (HI), then, gradually decreased. However, it was still significantly higher than those of the sham group 7days after HI (P <0.05). The expression of granzyme-b in the damaged brain and apoptosis rate 12 h, 1 d, 3 d and 7 d after HI for the HBO group were all significantly lower than those of the HIBD group (P < 0.05). Conclusions Expression of granzyme-b in the brain tissue was up-regulated following HIBD. HBO might be effective in the protection of brain damage induced by HI through inhibiting the expression of granzyme-b and apoptosis of neurons.
