Abstract:
Objective To study the regulatory effects of Escin on macrophage polarization factors,inflammatory factors and inflammation-related pathways.Methods RAW 264.7 macrophages were treated with Escin at concentrations of 0,0.5,1.0,2.0,5.0,10.0,and 20.0 μmol/L respectively.The effects of different concentrations of Escin on cell viability were determined by the MTT assay.The inflammation model of RAW264.7 macrophages was induced by lipopolysaccharide(LPS).The cells were divided into 6 groups according to treatment methods:Blank group,model group(500ng/ml LPS),treatment group 1(500ng/ml LPS+0.5μmol/L Escin),treatment group 2(500ng/ml LPS+1.0μmol/LEscin),treatment group 3(500ng/ml LPS+2.0μmol/L),and treatment group 4(500ng/ml LPS+4.0μmol/L Escin).Western blotting was used to detect the effect of Escin on the expression of mitogen-activated protein kinase(MAPK)signaling pathway proteins and the expression of macrophagy-related molecules in the LPS-induced RAW264.7 macrophage inflammation model.Results 0.5-5.0μmol/L Escin had no toxicity to RAW264.7 macrophages.The results of the Western blotting experiment showed that 4.0μmol/L Escin significantly downregulated the expression of tumor necrosis factor-α(P<0.05),and 0.5-4.0μmol/L Escin significantly upregulated the expressions of CD206 and CD 163(P<0.05).0.5-4.0μmol/L Escin significantly downregulated the expression of extracellular signal-regulated kinase(P<0.05),and 4.0μmol/L Escin significantly downregulated the expression of c-Jun N-terminal kinase(P<0.05).Conclusion In vitro,Escin can to a certain extent inhibit LPS-induced M1-type polarization of RAW264.7 macrophages,promote M2-type polarization,and exert the therapeutic effect of Escin on rheumatoid arthritis by regulating the MAPK signaling pathway.
