Abstract:
Objective To investigate the expression of ADAM17 in human placental villus and its effect on embryo implantation.Methods Semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and Western blot were applied to detect the expression of ADAM17mRNA and the protein ADAMl7 in villus tissues of normal pregnant woman and normal non-fertile endometria respectively at week 6,7,8,9 in the secretive phase.Results Semi-quantitative RT-PCR detection results indicated that the expression level of ADAM17mRNA in villus was obviously higher than that of the non-fertile endometria,with statistical significance (P < 0.01).Furthermore,its expression level reached peak at week 9 after pregnancy.The expression pattern and level of ADAM17 by Western blot detection was generally identical to that by semiquantitative RT-PCR,and no statistical significance could be noted,when comparisons were made between the two(P >0.05).Conclusions ADAM17 could be successfully expressed in placental villus and it might play an important role in the process of embryo implantation.
