Effects of transforming growth factor-β2 on the stress and apoptosis of human lens epithelial cells endoplasmic reticulum

Abstract:
Objective To investigate the effects of transforming growth factor-β2 (TGF-β2) on human lens epithelial cells endoplasmic reticulum,so as to provide evidence for the decrease of clinical cataracts incidence.Methods Human lens epithelial cells were cultured in vitro with different concentrations of transforming growth factorβ2 in the medium,and then,cell proliferation in the medium and the expression of associated markers of endoplasmic reticulum stress and apoptosis were analyzed statistically.Results When TGF-β2 concentrations were respectively at the rates of 0,100,200 and 500ng/L,corresponding OD values were respectively (0.519 ± 0.017),(0.459 ± 0.015),(0.357 ± 0.022) and (0.340 ± 0.028).Statistical significance could be seen in the OD and SR values of the 4 experimental wells,when SR values corresponded 100%,81.46%,61.79% and 58.57% (P < 0.05).With the increasing level of TGF-β2,the expression levels of caspase-3 and GRP 78 were increased,while their expression levels were decreased with the increase of Hrd1.There was statistical significance in the expression rates of 3 biomarkers of 4 experimental wells(P < 0.05).Conclusions TGF-β2 could inhibit cell survival and induce apoptosis,and at the same time,it could cause occurrence of the stress and apoptosis of human lens epithelial cells endoplasmic reticulum.
