Expression and clinical significance of DJ-1 in patients with rheumatoid arthritis

Abstract:
Objective:To detect DJ-1 expression in rheumatoid arthritis(RA)patients,collagen-induced arthritis(CIA)model mice and LPS-induced inflammation model RAW264.7 cells,and to explore its feasibility and clinical significance as a diagnostic bio-marker for RA.Methods:Peripheral blood of 24 RA patients and 22 healthy people from Jiangsu Province Hospital of Traditional Chinese Medicine(Affiliated Hospital of Nanjing University of Traditional Chinese Medicine)were collected,peripheral blood mono-nuclear cell(PBMC)and serum were isolated,and DJ-1 mRNA expression in PBMC was detected by qPCR.DJ-1 protein concentra-tion in serum were assayed by ELISA.Receiver operating characteristic(ROC)curve was generated,and diagnostic efficacy was evaluated by area under the curve(AUC),specificity and sensitivity.Pearson and Spearman correlation coefficient analysis were con-ducted to explore relationship between DJ-1 concentration and RA disease activity.CIA mice model and LPS-induced RAW264.7 cells inflammation model were established.DJ-1 mRNA expression in mouse synovial tissue and RAW264.7 cells were measured by qPCR.DJ-1 protein concentration in mouse serum and RAW264.7 cells supernatant were determined by ELISA.Malondialdehyde(MDA)and glutathione(GSH)levels were also measured,as well as activity of superoxide dismutase(SOD)in cell supernatant.Results:DJ-1 expression in PBMC from RA patients,synovial tissues of CIA mice and LPS-induced RAW264.7 cells were significantly lower than normal control group(P<0.05);ELISA results showed that DJ-1 concentration in RA patients' serum,CIA mice' serum and RAW264.7 cells supernatant were lower than normal control group(P<0.05).Patients with active RA showed lower DJ-1 concentra-tion in comparison to inactive RA(P<0.05).Additionally,DJ-1 concentration was negatively correlated with disease activity of RA.RAW264.7 cells treated with LPS showed an increase in MDA,while SOD and GSH significantly decreased(P<0.05).AUC of serum DJ-1 protein was 0.90,sensitivity was 75.00%,and specificity was 90.91%.Conclusion:DJ-1 concentration decreases in RA patients and negatively correlated with disease activity of RA,which is expected to become a novel biomarker for diagnosis of RA.
